- 【特許請求の範囲】 1. 非哺乳類タンパク質をコードするDNA配列またはその断片を含んでなる合 成ポリヌクレオチドであって、該DNA配列が、哺乳類宿主内での発現のために最 適化されたコドンを含むことを特徴とする合成ポリヌクレオチド。 2. 該タンパク質が、HIVタンパク質、HSVタンパク質、HAVタンパク質、HBVタ ンパク質、HCVタンパク質、HPVタンパク質、HSVタンパク質、プラスモジウム(P lasmodium)タンパク質、マイコバクテリウム(Mycobacterium)タンパク質、ボ レリア(Borrelia)タンパク質およびロタウイルスタンパク質から選ばれる、請 求項1に記載のポリヌクレオチド。 3. 該タンパク質がHIVタンパク質である、請求項2に記載のポリヌクレオチ ド。 4. DNA配列: (配列番号1)を有する、請求項3に記載のポリヌクレオチド。 5. 該ポリヌクレオチドが、HIVgag、gag-プロテアーゼまたはenv遺伝子産物 をコードする遺伝子を含み、ヒト組識を含む脊椎動物組識内にインビボで導入さ れると抗HIV中和抗体、HIV特異的T細胞免疫応答または防御免疫応答を誘導する 、請求項3に記載のポリヌクレオチド。 6. 脊椎動物において免疫応答を誘導するための方法であって、請求項1に記 載のポリヌクレオチド1ng〜100mgを該脊椎動物の組識内に導入することを含んで なる方法。 7. 弱毒化病原体、不活化病原体、サブユニットワクチン、タンパク質ワクチ ンおよびそれらの組合せの投与を更に含む、請求項6に記載の方法。 8. 請求項3に記載のポリヌクレオチドおよび医薬上許容される担体および所 望によりアジュバントを含んでなる、HIV感染に対する免疫応答を誘導するため の免疫原性組成物。 9. 霊長類において抗HIV免疫応答を誘導するための方法であって、請求項3 に記載のポリヌクレオチドを該霊長類の組識内に導入し、同時にサイトカインを 非経口的に投与することを含んでなる方法。 10. HIV抗原に特異的なリンホカイン分泌を含むエフェクタ ー機能として細胞傷害性およびヘルパーT細胞増殖を刺激するよう抗原提示細胞 を誘導する方法であって、脊椎動物の細胞を請求項3に記載のポリヌクレオチド にインビボでさらすことを含んでなる方法。 11. 請求項3に記載のポリヌクレオチドを抗HIV抗ウイルス剤と共に患者に投 与することを含んでなる、治療を要する患者を治療する方法。 12. 請求項1に記載のポリヌクレオチドを含んでなる医薬組成物。
