Claims (1)
- 【特許請求の範囲】 1.天然ヌクレオチド、修飾ヌクレオチド、及びそれらの組み合わせか らなる群からの1つまたは2つの分子、並びに、第1のフルオロホア及び発色団 からなる非FRETのラベル対を含んでなり、核酸鎖とハイブリダイゼーション することができるプローブであって、前記発色団が、フルオロホア及び消去剤か らなる群から選択され、このプローブとターゲットとの相互作用が、プローブの 第1の立体配座から第2の立体配座への変化を引き起こし、それにより、前記ラ ベル対のラベル間の距離を変化させ、一方の立体配座のみにおいて、前記第1の フルオロホアの蛍光を少なくとも25パーセント消光させるのに十分にラベルが 相接することを特徴とするプローブ。 2.前記プローブが、互いにハイブリダイズしてハイブリッドを形成で きる2つの分子を含み、前記ラベル対が、前記分子の一方が、前記ハイブリッド の末端において前記分子の一方に結合し、ハイブリッドにおける消光効率が少な くとも60%であり、スペクトルのオーバーラップに基づいて予想される消光効 率より少なくとも10パーセント高いことを特徴とする請求項1記載のプローブ 。 3.前記プローブが、1つの分子を含む請求項1記載のプローブ。 4.前記非FRETラベル対の成員が、2本鎖DNAのハイブリダイズ した末端ヌクレオチドに結合したとき、少なくとも60%であり、スペクトルの オーバーラップに基づいて予想される消光効率より少なくとも10パーセント高 い消光効率を与えることを特徴とする請求項3記載のプローブ。 5.前記非FRETラベル対の成員が、2本鎖DNAのハイブリダイズ した末端ヌクレオチドに結合したとき、少なくとも70%であり、スペクトルの オーバーラップに基づいて予想される消光効率より少なくとも30パーセント高 い消光効率を与えることを特徴とする請求項4記載のプローブ。 6.発色団が消去剤であることを特徴とする請求項4または5記載のプ ローブ。 7.消去剤が、DABCYLであることを特徴とする請求項6記載のプ ローブ。 8.発色団が、前記第1のフルオロホアの最大励起より短い最大励起波 長を有するフルオロホアであることを特徴とする請求項4または5記載のプロー ブ。 9.前記分子が、互いにハイブリダイズして3−25ヌクレオチド長の ハイブリッドを形成することのできる腕に隣接した、7−140ヌクレオチドの ターゲットと相補的な領域を有する自己ハイブリダイズするオリゴヌクレオチド であることを特徴とする請求項4から7のいずれかに記載のプローブ。 10.2つの非FRETラベル対を含むことを特徴とする請求項1記載 のプローブ。 11.請求項1から10のいずれかに記載のプローブを含む検出試薬を 含むアッセイ用試薬キット。 12.核酸増幅を行う試薬をさらに含むことを特徴とする請求項11記 載のキット。 13.前記増幅が、PCR、3SR、LCR、Q−ベータ複製、及びN ASBAからなる群から選択されることを特徴とする請求項12記載のキット。 14.前記増幅がPCRであることを特徴とする請求項13記載のキッ ト。 15.請求項1から10のいずれかに記載のプローブを該プローブのタ ーゲットを含むと考えられるサンプルに接触させ、蛍光の変化を測定することを 含んでなる検出過程を含むことを特徴とするアッセイ。 16.多重アッセイであることを特徴とする請求項15記載のアッセイ 。 17.PCR法による増幅を含み、検出がリアルタイムで行われること を特徴とする請求項15または16記載のアッセイ。 18.少なくとも1つの一本鎖ハイブリダイゼーションプローブを、該 プローブのターゲットを含むと考えられるサンプルに接触させ、吸収を測定する ことにより検出することを含み、前記プローブが、天然オリゴヌクレオチドとハ イブリダイズすることができ、前記プローブが、天然ヌクレオチド、修飾ヌクレ オチド、及びそれらの組み合わせからなる群から選択されるヌクレオチドを含み 、前記プローブが、発色団の対を含み、このプローブとターゲットとの相互作用 が、 プローブの第1の立体配座から第2の立体配座への変化を引き起こし、それによ り、前記発色団の間の距離を変化させ、1つの立体配座のみにおいて、検出可能 な吸収変化させるのに十分に発色団を相接することを特徴とするアッセイ。 19.前記プローブが、互いにハイブリダイズして3−25ヌクレオチ ド長のハイブリッドを形成することのできる腕に隣接した、7−140ヌクレオ チドのターゲットと相補的な領域を有する自己ハイブリダイズするオリゴヌクレ オチドであることを特徴とする請求項18記載のアッセイ。
