Abstract
Tablets or granules of medicament having a room temperature water-solubility of about 1% or less are prepared by applying to the surface of particles of the medicament a partial or complete envelope of a non-toxic film-forming substance, admixing the particles with a water-soluble ingestible ingredient selected from one or more of the following: glucose, dextrose, mannitol, sorbitol, lactose, glycine, leucine and delta glucono-lactone and with a pharmaceutically acceptable water-soluble solid acid or acid salt and then compressing the mixture so formed into tablets and granules. Specified film-forming substances are dimethyl phenyl silanes, ethyl cellulose, cellulose acetate phthalate, polyacrylic acid, polyacrylamide, polyvinylacetate, polyvinyl alcohol acetate, polyvinyl acetate stearate, polyvinyl methyl ether and ethylene glycol distearate, preferably on a dry basis used in an amount which is 1-10% by weight of the medicament. Specified acids and acid salts are tartaric acid, potassium acid tartrate, citric acid, mono- and di-sodium citrates, fumaric acid, adipic acid, malic acid and monosodium phosphate, preferably in an amount ranging from 1-6% by weight of the finished tablets or granules. Specified medicaments are 2-(b -chloroethyl)-2,3-dihydro-4-oxo-(benzo-1,3-oxazine), dihydroxy aluminium carbonate, phenacetin, acetyl salicylic acid, salicylamide, codeine, antibiotics, e.g. oxytetracycline, vitamins, e.g. Vitamin A, sulphathiazole and steroid hormones, e.g. hydrocortisone. The tablets or granules may also contain water-soluble drugs such as water-soluble vitamins, e.g. nicotinamide and stimulants, e.g. caffeine. Specified tableting and granulating additives are lubricants, e.g. stearic acid talc and magnesium stearate, sweetening agents, e.g. saccharin and sodium saccharin, lemon flavour, spearmint oil and sodium chloride. The coated medicament particles should have a size in the range 16-100 mesh B.S.S.
