Abstract

In the production of sustained release pharmaceutical pellets, substantially spherical innocuous cores, e.g. of sugar, synthetic resin or nitrocellulose, or naturally-occurring seeds, are coated with a medicament, the cores are subdivided into groups, each group is provided with a slowly digestible coating to produce pellets of 0,1-2 mm. diameter to provide different times of release of the medicament on ingestion, and the pellets are combined e.g. by enclosure in a gelatin capsule, if desired with uncoated pellets. The cores are first wetted in a coating pan with gelatin &c.-sugar solution, and sprinkled with medicament, the operation being repeated as necessary; alternatively, the adhesive solution may contain dissolved or suspended medicament. The time delay material may be applied as a solution in various specified organic solvents of glyceryl monostearate and a wax, beeswax being preferred, or wax-like substances such as chlolesterol. Fatty acids and alcohols may be used in place of glyceryl monostearate. The various delay times may be provided by varying the thickness of the coatings, or by using different coating materials. Medicaments specified are salts of racemic and dextro-amphetamine, racemic and dextro-desoxyephedrine, and 2-amino-1-(4 methylphenyl)-propane sulphate, or potassium iodide. The medicament may be extended with calcium sulphate dehydrate, starch, acacia. Specifications 523,594, as open to inspection under Sect. 91, and 715,305 are referred to.
