Abstract

A synthetic thyroprotein is prepared by treating a protein containing tyrosine with iodine until the million test becomes negative or the biuret test yields a blue or blue-violet colour and then maintaining the iodinated protein at a temperature which is higher than that at which the iodination was effected and is also between 50-100 DEG C. for 12-24 hours. The treatment of the protein with iodine should be carried out at 30-45 DEG C., preferably 37-40 DEG C., and in the presence of a metal such as copper, tin, zinc or brass, as catalyst. Proteins which may be used include skim milk, casein, albumen, blood serum, albumin, globulin, egg albumen, meat meal, cottonseed meal, gluten meal, soyabean meal, peanut meal, cocoanut meal. Instead of iodine, a mixture of iodine and potassium iodide in aqueous solution. In examples: (1) pulverized iodine is added to an aqueous solution containing tyrosine at pH 6-8 at 30-45 DEG C., with sodium bicarbonate in the solution as a buffer. The solution is then incubated at 50-100 DEG C. for 12-18 hours in the presence of a metal rod as catalyst; (2) powdered iodine is added to an aqueous solution of skim milk in the presence of sodium bicarbonate at 38-40 DEG C. The solution was then incubated at 38 DEG C. for 18-20 hours. Dilute hydrochloric acid was added to adjust pH to 3.8-4.0 and the precipitated iodinated protein washed with water at pH 4.0.
