Abstract

526,951. Cholesterol. ARMOUR & CO. March 21, 1939, No. 8989. Convention date, May 3, 1938. [Class 2 (iii)] Cholesterol is extracted from fatty animal tissue by reacting either in solution or in the dry state the animal tissue with an alkaline saponifying agent chosen from the group consisting of the alkaline earth metal oxides and hydroxides at a temperature above 100‹C. and extracting the reaction mixture with an organic solvent for the cholesterol. The preferred saponifying agent is calcium hydroxide, and the preferred solvent is a chlorinated hydrocarbon such as ethylene dichloride. Other solvents mentioned include ether, alcohol, acetone, petroleum ether, chlorinated benzol and ethyl acetate. It is preferred to use the saponifying agent in a substantially dry condition. In examples (1) minced beef spinal cord is mixed with slaked lime and heated in an autoclave for three hours at 100‹-125‹C. The reaction mixture is then extracted with ethylene dichloride. The extract is cooled, filtered, and evaporated. (2) A mixture of spinal cord and strontium hydroxide is heated for three hours in an autoclave at 15 lb. steam pressure. Water is removed by filtration and the solid residue is extracted with ethyl acetate. (3) Spinal cord is mixed with magnesium oxide and after saponification and removal of water, the reaction product is extracted with orthodichlorbenzene.
