Abstract
1494230 Insulin ELI LILLY & CO 30 Jan 1975 4200/75 Heading C3H A process for the purification of alkali metal insulin or ammonium insulin comprises swelling a gel having in the dry state a water-regain of at least 4% by weight and particle diameters less than 100 microns with an aqueous medium; packing a column with said swollen gel, adding to said packed column an aqueous solution of an alkali metal insulin or ammonium insulin of purity at least 80% wherein the pH of the solution is 7À5-9À5, the alkali metal insulin or ammonium insulin concentration is 1-8%, weight per volume and the total amount of alkali metal insulin or ammonium insulin is sufficient to provide a column loading of 0À8-6À7 g/litre of bed volume; and eluting the alkali metal insulin or ammonium insulin from the column at 5-30‹ C. with an aqueous eluant having a pH of 7À5 to 9À5. In the example, beef sodium insulin is purified on cross-linked dextran gel with aqueous sodium hydroxide. The alkali metal insulin or ammonium insulin may be converted into zinc insulin.
