1293757 Preparing samples for microscopy PAINES & BYRNE Ltd and M LEE 22 Oct 1969 [22 Oct 1968] 50050/68 Heading G2J [Also in Division B1] A nerve-stain composition is prepared by forming a solution of methylene blue and a reducing agent, such as ascorbic acid, in a solvent, preferably water, such that the methylene blue is reduced to lenco-methylene blue, the composition having a p-H of from 3-6 preferably about 4. On application to tissue, oxidation of the lencomethylene blue to methylene blue takes place more quickly where nerve tissue is present than elsewhere so that the nerve tissue is differentiated by colour from surrounding tissue.
