Claims (9)
1. Förfarande för f raitista lining av fusionsproteiner med en C- eller N-terminal del som motsvarar en interleu-5 kin-2 (IL-2) -molekyl, företrädesvis humant IL-2, men som innehäller mindre än 100 aminosyror, med undantag av de delar som innehäller 96 - 99 aminosyror, känne-t e c k n a t därav, att en gen som kodar för fusionspro-teinet bringas till expression i en värdcell. 10
2. Förfarande enligt patentkrav 1, känne- t e c k n a t därav, att genen som kodar för IL-2-delen omfattar en del av DNA-sekvensen I (bilaga).
3. Förfarande enligt patentkrav 2, känne- t e c k n a t därav, att genen som kodar för IL-2-delen 15 bestär av ett, tvä eller tre av segmenten A - F i IL-2-genen (EcoRI)-A-Pstl-B-Mlul-C-Xbal-D-Sacl-E-Pvul-F-(Sali) i valbar ordningsföljd ooh eventuellt sammankopplade me-delst adaptor- eller kopplingssekvenser.
4. Förfarande enligt nägot av patentkraven 1-3, 20 kännetecknat därav, att mellan IL-2-sekvensen ooh det önskade proteinets aminosyrasekvens finns en ami-nosyra eller aminosyrasekvens, som möjliggör kemisk eller enzymatisk avspjälkning av det önskade proteinet frän IL-2-delen, företrädesvis aminosyran Met, Cys, Trp, Lys eller 25 Arg eller en aminosyrasekvens, som innehäller nägon (näg-ra) av dessa aminosyror i C-terminalen.
5. Förfarande enligt patentkrav 4, kännetecknat därav, att aminosyrasekvensen är Asp-Pro eller Ile-Glu-Gly-Arg eller innehäller denna sekvens i C- . 30 terminalen.
6. Förfarande enligt nägot av patentkraven 1-5, kännetecknat därav, att genen som kodar för fusionsproteinet bringas till expression i en bakterie-cell, företrädesvis i E. coli. 93734
7. Användning av fusionsprotein soin kan erhällas enligt nägot av patentkraven 1-6 för framställning av önskade proteiner.
8. Genstruktur, kännetecknad därav, 5 att den kodar för ett fusionsprotein som kan erhällas enligt nägot av patentkraven 1-6.
