Claims (14)
1. Ein chimeres Isoprenoidsynthase-Polypeptid, umfassend eine erste Domäne aus
einer ersten Isoprenoidsynthase, gebunden an eine zweite Domäne aus einer zweiten
heterologen Isoprenoidsynthase, wobei die chimere Isoprenoidsynthase in der Lage
ist, die Bildung von Isoprenoid-Reaktionsprodukten zu katalysieren, die in
Abwesenheit der zweiten Domäne der zweiten heterologen Isoprenoidsynthase nicht gebildet werden.
2. Die chimere Isoprenoidsynthase nach Anspruch 1, wobei die chimere
Isoprenoidsynthase in der Lage ist, mindestens zwei verschiedene Isoprenoid-Reaktionsprodukte
zu katalysieren.
3. Die chimere Isoprenoidsynthase nach Anspruch 1, wobei die zweite Domäne der
zweiten heterologen Isoprenoidsynthase auch das Verhältnis der Isoprenoid-Reaktionsprodukte
der chimeren Isoprenoidsynthase bestimmt.
4. Die chimere Isoprenoidsynthase nach Anspruch 1, wobei die erste Domäne der
ersten Isoprenoidsynthase eine Pflanzen-Isoprenoidsynthase und die zweite Domäne
der zweiten heterologen Isoprenoidsynthase eine Pflanzen-Isoprenoidsynthase ist.
5. Die chimere Isoprenoidsynthase nach Anspruch 4, wobei die chimere
Isoprenoidsynthase ausgewählt ist aus der Gruppe bestehend aus (a) der Tabak- Hyoscyamus-CH4 chimeren Isoprenoidsynthase; (b) der Tabak-Hyoscyamus-CHIO
chimeren Isoprenoidsynthase; (c) der Tabak-Hyoscyamus-CHll chimeren
Isoprenoidsynthase; (d) der Tabak-Hyoscyamus-CHU chimeren
Isoprenoidsynthase; (e) der Tabak-Hyoscyamus-CHtä chimeren Isoprenoidsynthase;
oder (f) der Tabak-/^o5cyür/ni/5-CH14 chimeren Isoprenoidsynthase.
6. Die chimere Isoprenoidsynthase nach Anspruch 1, wobei die chimere
Isoprenoidsynthase die Bildung eines Mittels gegen Pilze katalysiert.
7. Die chimere Isoprenoidsynthase nach Anspruch 1, wobei die chimere
Isoprenoidsynthase die Bildung eines antibakteriellen Mittels katalysiert.
8. Die chimere Isoprenoidsynthase nach Anspruch 1, wobei die chimere
Isoprenoidsynthase die Bildung eines Antitumormittels katalysiert.
9. DNA kodierend ein chimeres Isoprenoidsynthase-Polypeptid nach Anspruch 1.
10. Ein Vektor, enthaltend die DNA nach Anspruch 9.
11. Eine Zelle, enthaltend die DNA nach Anspruch 9.
12. Die Zelle nach Anspruch 11, wobei die Zelle E. coli ist.
13. Ein chimeres Isoprenoidsynthase-Polypeptid, umfassend eine asymmetrisch
angeordnete homologe Domäne, wobei die chimere Isoprenoidsynthase in der Lage ist, die Bildung von Isoprenoid-Reaktionsprodukten zu katalysieren, die nicht
gebildet werden, wenn diese Domäne an ihrer natürlich vorkommenden Stelle in
dem Isoprenoidsynthase-Polypeptid angeordnet ist.
14. Verfahren zur Herstellung eines chimeren Isoprenoidsynthase-Polypeptids,
umfassend
(a) die Bereitstellung einer Zelle, die durch DNA nach Anspruch 9 transformiert
ist, wobei die DNA zur Expression in der Zelle angeordnet ist;
(b) Züchten der transformierten Zelle unter Bedingungen zur Expression der
DNA; und
(c) Isolieren der chimeren Isoprenoidsynthase.
