Die vorliegende Erfindung bezieht sich auf die Verwendung von beta-Cyclodextrin zur Behandlung von Akne.
Akne ist eine Erkrankung der Talgdrüsen. Sie tritt ausserordentlieh häufig auf, besonders, wenn man auch ihre frühen Stadien mit
rechnet, die häufig als"Hautunreinheiten" bezeichnet werden.
Das Anfangsstadiuiii der Akne ist durch die Ansammlung von Horn-Talgpropfen in den Talgdrüsenausgängen und die Ausbildung von
Komedonen charakterisiert; später treten entzündliche Veränderungen hinzu, deren unterschiedliche Erscheinungsformen vor allem durch
die Tiefe und den Grad der Gewebszerstörung bedingt ist. Schliesslich kann es zur Narbenbildung bzw. Epithelwucherungen und Zystenbildung kommen.
Je nach Schweregrad und Ausprägung spricht man von Akne vulgaris,
Akne comedonica, Akne papulo pustulosa, Akne conglobata usw.
Obwohl die Ursachen dieser Hautveränderungen noch nicht ganz geklärt sind, hat es nicht an Vorschlägen zur kosmetischen
und/oder therapeutischen Behandlung der Akne gefehlt. Dies
liegt vor allem daran, weil Akne insbesondere auf der Gesichts-, Brust- und Rückenhaut auftritt, was von den meisten Patienten,
auch aus psychologischen Gründen, besonders unangenehm empfunden wird. Die Tatsache, dass Akne am häufigsten in der Pubertätsperiode
auftritt, liess vermuten, dass Hormone hierbei eine Rolle spielen,
und man hat zur Behandlung der Akne z.B. östrogene Hormone vorgeschlagen. Aber auch Bakterizide, Antibiotika und keratolytische

Mittel finden Anwendung. Diese Mittel haben aber, bei meist
geringer Anti-Aknewirksainkeit, /.!. beträchtliche Nebenwirkungen,
wie Hautreizung, Toxizität oder horninnel Ie Veränderungen zur Kolge.
fs war datier Aufgabe der vorliegenden Erfindung, einen Wirkstoff zu
beschaffen, der diese Nachteile nicht aufweist und eine deutliche
Wirkung gegen Aktie zeigt. Diese Aufgabe wird geniäss der Erfindung
nun dadurch gelöst, dass gefunden wurde, dass beta-Cyclodextrin
diese Erfordernisse erfüllt. Bei einer klinischen Prüfung hat
K) sich gezeigt, dass innerhalb kurzer Zeit eine beträchtliche
symptomatische Heilung der mit beta-Cyclodextrin behandelten Fälle
von Akne der verschiedensten Erscheinungsformen erzielt wird.
Die Erfindung betrifft somit die Verwendung von beta-Cyclodextrin als Anti-Akneinittel in kosmetischen und pharmazeutischen
Präparaten zur Behandlung von Akne.
beta-Cyclodextrin, u.a. auch als "Schardinger Dextrin" oder Cycloheptaamylose
bekannt, wird durch Fraktionierung von enzymatisch U abgebauter Stärke gewonnen. In Die Stärke", 29. Jahrg. 1977,
Nr. 1, Seiten 26-33, werden einige Anwendungsmöglichkeiten der
Cyclodextrine in der Arzneimittelindustrie besprochen, ir>
der die Cyclodextrine auf Grund ihrer Fähigkeit, Einschlussverbindungen zu bilden, zur Erhöhung der Löslichkeit von schwerlöslichen Arzneimitteln,
zur Verbesserung der Stabilität anderer Verbindungen oder zur Herabsetzung der Flüchtigkeit anderer Verbindungen vorteilhaft
sein können. Eine Verwendung von beta-Cyclodextrin an sich als pharmazeutischer Wirkstoff ist aber in dieser Veröffentlichung
nicht beschrieben. beta-Cyclodextrin ist wegen seiner Fähigkeit
Su zur Bildung von Einschlussverbindungen auch schon in desodorisierenden
kosmetischen Präparaten (JA-PS 41440/78) sowie in Badezusatzpräparaten
(JA-PS 63126/75) vorgeschlagen worden.
Es ist aus dem bisherigen Stand der Technik aber nicht bekannt, dass beta-Cyclodextrin ein wirksames Mittel zur Behandlung von Akne
ist. Da die Wirkung vermutlich rein physikaiischer Natur ist,
verursacht die Verwendung des beta-Cyclodextrins keine signifikanten unerwünschten Nebeneffekte auf Haut oder Hautflora,

wie dies bei den chemisch oder physiologisch wirkenden Substanzen
der Fall ist.
Das beta-Cyclodextrin kann in ieglicher Applikationsform angewendet
werden. Typische Beispiele sind Lotionen, Salben, Suspensionen, Cremes, Gele usw. Im allgemeinen beträgt der Gehalt an beta-Cyclodextrin in diesen Applikationsformen von 0,1-10, vorzugsweise
1-5 Gew.X.
Die Erfindung wird nun anhand folgender Versuchsergebnisse näher
erläutert.
Es wurden zwei wässrig-alkoholische Lösungen nachstehender Zusammensetzung zur Behandlung von 64 Akne-Fällen angewendet.

Behandelt wurden alle Variationen der Akne vulgaris, Akne
comedonica, Akne papulo pustulosa, Akne conglobata, einzeln stehende
Pusteln etc., d.h. sowohl die pubertären als auch die späten Akne-Fälle bei Patienten beider Geschlechter; und in allen Fällen
bestand die bereits mehrere Jahre.
Die Behandlung umfasste ein zweimal tägliches Behandeln der von
Akne befallenen Partien, und in wöchentlichen Intervallen wurde
die Anzahl der noch vorhandenen Pusteln und Mitesser registriert und ins Verhältnis zur Ausgangszahl gesetzt.
Unter den 54 mit Präparat A behandelten Fällen von Akne der verschiedensten Erscheinungsformen und Schweregrade wurde nach 1-4
Wochen schon bei 43 Probanden ( = 80/0) eine symptomatische Heilung
zu 60-100% beobachtet; mit Präparat B wurde in einem Versuch mit 10 Probanden in 3 Fällen eine geringfügige Besserung erreicht.
