Abstract
<p>La liberación retardada de un fármaco al colon se logra de una formulación de liberación retardada que comprende un núcleo y un recubrimiento para el núcleo. El núcleo comprende un fármaco y un recubrimiento que comprende: (I) una capa exterior y una capa de aislamiento entre el núcleo y la capa externa, o, (II) una capa exterior y una capa interior entre el núcleo y la capa externa, o (III) una capa exterior y tanto una capa de aislamiento como una capa interior entre el núcleo y la capa exterior. La capa exterior comprende una mezcla de un primer material polimérico que es susceptible a ataques por parte de la bacteria colónica, y un segundo material polimérico que tiene un umbral de pH en el intervalo de pH 6 a 8. La capa interior comprende un tercer material polimérico que es soluble en el fluido intestinal o en el fluido gastrointestinal, tal tercer material polimérico es un ácido policarboxílico que está al menos parcialmente neutralizado con 10% a 100% de los grupos ácido carboxílico de dicho polímero de ácido policarboxílico en forma de aniones carboxilato. La capa de aislamiento comprende un polímero no iónico que es soluble en el fluido intestinal o fluido gastrointestinal. La capa exterior se aplica directamente a la capa interior o la capa de aislamiento usando una preparación de recubrimiento formada por la combinación del primer material polimérico en medio acuoso con un segundo material polimérico en un medio orgánico.
