本发明提供一种由含有来源于己糖的酸、其非毒性盐及其分子内酯化合物中的至少一种作为有效成分的生物体内有害物质的生成抑制剂组成的动物饮料水及来源于己糖的酸、其非毒性盐及其分子内酯化合物中的至少一种在制备生物体内有害物质的生成抑制剂的用途。
Description
本申请是申请号为00817058.4,申请日为2000年10月18日,发明名称为动物的育肥促进剂及育肥促进方法的分案申请。
技术领域
本发明涉及动物的育肥促进剂及育肥促进方法。更具体地说,本发明涉及一种含有由以葡萄糖酸为代表的来源于己糖的酸、其无毒性盐及其分子内酯化合物中的至少一种成分作为有效成分的动物育肥促进剂以及使用该动物育肥促进剂的育肥促进方法。
背景技术
过去,为了促进家畜等动物的生长,可以使用添加有四环素、阿伏霉素等抗生物质作为育肥促进剂的饲料。然而,这些抗生物质在动物体内不仅容易诱发细菌由于突然变异而导致的耐药性,而且,这些抗生物质残留在动物体内还存在通过吃肉等方式而被人体摄取的危险性,其结果,这些抗生物质作为医药的效力恐怕会逐步降低。
因此在近年来,人们正在开展使用抗生物质以外的物质作为动物育肥促进剂的研究,例如,使用富马酸作为育肥促进剂。
另一方面,关于以葡糖酸为代表的来源于己糖的酸,有一种属于分子内酯化合物的葡聚糖-δ-内酯(glucono-δ-lactone)已被广泛地作为豆腐的凝固剂使用,而葡糖酸钙则被广泛地作为钙的补给剂使用,进而在近年来,葡糖酸钠和葡糖酸钾也作为食品添加剂被认可,可以在各种食品中使用(国际专利申请WO 94/09650号)。
发明内容
本发明者们认为,可以通过向动物投与对生物体的安全性很高的葡糖酸及其衍生物来研究该葡糖酸及其衍生物对生物体的活性。于是,本发明者们尤其是调查了该葡糖酸及其衍生物在动物肠脏内的投与效果,结果意外地观察到特定的肠内细菌发生增殖,而且发现,这种肠内细菌能明显地促进那些对生物体生长有用的短链脂肪酸的产生。并且还观察到,那些对生物体有害的物质的生成也相应地减少或消失。
也就是说,当将本发明来源于己糖的酸、其非毒性盐和分子内酯化合物(下文将这些化合物统称为“来源于己糖的酸类”)向动物投喂时,在该动物的肠内细菌中,巨型球菌属(Megasphaera)和光岗菌属(Mitsuokella),具体地为埃氏巨球形菌(Megasphaera elsdenii)和多酸光岗菌(Mitsuokella multiacidus),有特异性的增殖。
另一方面,与不存在来源于己糖的酸类的情况相比,对作为短链脂肪酸的乙酸的生成促进约2倍,对丙酸的生成促进约4倍,对丁酸的生成促进约8倍。另外,与此同时,以乳酸为首,那些被认为能导致生物体发生痢疾、消化管溃疡、腔管酸中毒(lumen acidosis)等有害作用的琥珀酸、氨和硫化氢的生成变为减少或消失。
由此可以看出,当来源于己糖的酸类一旦被动物摄取,就能促进动物体内的短链脂肪酸的生成和显著地改善饲料效率,从而能促进动物的育肥。

用于实施发明的最佳方案
本发明来源于己糖的酸可以是D-体、L-体或其混合物中的任一种。具体地可以举出:葡糖二酸、甘露聚糖酸、グラル酸、イダル酸之类的糖酸,或葡糖酸、半乳糖酸、甘露糖酸、塔龙酸、阿洛糖酸之类的醛糖酸。其中特别优选的是葡糖酸。
作为这些酸的非毒性盐,例如可以举出与钠、钾之类的碱金属形成的盐,与钙、镁之类碱土金属形成的盐,与铜、铁、锌之类过渡金属形成的盐,其中特别优选的是葡糖酸钠和葡糖酸钙。
另外,作为上述那些的酸的分子内酯化合物,可以举出内酯化合物,具体地是葡糖酸的分子内酯化合物,例如葡聚糖-δ-内酯、葡聚糖-γ-内酯,其中特别优选葡聚糖-δ-内酯。
这些来源于己糖的酸类可以单独使用,也可以任意两种以上组合使用。
作为短链脂肪酸,可以举出:乙酸、丙酸、丁酸、异丁酸、戊酸、异戊酸、辛酸等碳原子数2~4的脂肪酸。对生物体内的吸收机构和生物体本身会带来不同影响的乳酸和琥珀酸不包含在本发明的短链脂肪酸内。
另外,所谓“饲料效率的改善”主要是指,即便对于使用粗杂谷物制造的营养价值低的动物用饲料,通过向其中加入来源于己糖的酸类,便可以获得营养价值高的优质饲料以及同样程度的育肥促进效果,而在向营养价值高的饲料中加入来源于己糖的酸类的情况下,可以获得更显著的育肥促进效果。
来源于己糖的酸类可以直接使用,也可以与那些在该领域中公知的固体或液体赋形剂一起作为动物的育肥促进剂、动物用肠内细菌增殖促进剂和生物体内有害物质的生成抑制剂使用,所说赋形剂的例子有:乳糖、蔗糖、葡萄糖、玉米淀粉、明胶、淀粉、水、甘油、脂肪油、山梨糖醇等。
可以将药剂根据需要与抗菌剂、防霉剂、驱虫剂、抗氧化剂、色素、香料、调味剂、酶之类常用的添加剂混合,并按照常规方法制成粉剂、散剂、颗粒剂、液剂、片剂等形态。
另外,可以将药剂本身直接向动物投喂,或者也可以将其添加到动物所要摄取的饲料或饮料水中来向动物投喂,但优选是将其配合到动物用的饲料或饮料水中使用。
所说的饲料或饮料水只要是通常使用的即可,对此没有特殊限定。作为饲料的一个例子,可以举出将下述物质按适宜比例混合配制而成的饲料,所说的物质包括:玉米、米、麦、高粱、大豆饼、麸子、脱脂米糠、鱼粉、脱脂奶粉、干燥乳清、油脂、苜蓿粉、北洋鱼粉(northern-seameal)、大豆油脂、粉末精制牛脂、小麦粉、菜籽油、肉骨粉(羽毛粉)、动物性油脂、磷酸钙、玉米麸粉(コ-ングルテンミ-ル)、糖浆、玉米胚粉(コ-ンジャムミ-ル)、碳酸钙、磷酸三钙、氯化钠、氯化胆碱、维生素类(维生素A、维生素B1、维生素B2、维生素B6、维生素B12、维生素D、维生素E、泛酸钙、烟酰胺、叶酸等)、氨基酸类(赖氨酸、蛋氨酸等)、微量无机盐类(硫酸镁、硫酸铁、硫酸铜、硫酸锌、碘化钾、硫酸钴等)、微生物等。
来源于己糖的酸类的用量可根据作为对象的动物的种类和体重等来决定,但通常每天可按20~1300mg/kg,优选按20~600mg/kg的比例使用,在配合到动物用饲料或饮料水中使用的情况下,通常对其有效成分按0.05~2.0重量%(下文仅仅以“%”表示)的比例使用,优选按0.1~1.0%的比例使用。
本发明还提供了上述育肥促进剂和把配合有该育肥促进剂的动物用饲料或饮料水向动物饲喂的动物饲养方法。
作为适用该方法的动物,可以举出那些属于通常饲养动物的牛、猪、马、羊、山羊、兔、水貂、鸡、火鸡、家鸭、鹌鹑、野鸭、甲鱼、蛙、虾、鲥、鲷、河豚、鳗、鲑、鳟、竹英鱼等,特别优选是以牛、猪或鸡作为饲养对象。配合有本发明的育肥促进剂和配合有该育肥促进剂的动物用饲料和饮料水可以按一般的方法向上述这些动物投喂。
在使用本发明的育肥促进剂的同时,还可以把巨球形属和光岗菌属本身作为例如干燥制剂直接给动物投喂,在此情况下,可以认为,伴随着对肠内细菌的增殖促进作用,可以获得更好的育肥促进效果。
实施例
下面举出实施例来更详细地说明本发明,但本发明不受这些实施例的限定。
实施例1
向猪用饲料(日配哺乳期幼猪喂养用配合饲料コロミ-ルGS、日本配合饲料(株)制)中添加相当于饲料重量0.0625%的葡糖酸钠。
实施例2~5
向猪用饲料(日配哺乳期幼猪喂养用配合饲料コロミ-ルGS、日本配合饲料(株)制)中分别添加相当于饲料重量0.125%(实施例2)、0.25%(实施例3)、0.5%(实施例4)、1.0%(实施例5)的葡糖酸钠。
比较例1
猪用饲料(日配哺乳期幼猪喂养用配合饲料コロミ-ルGS、日本配合饲料(株)制)直接用作饲料。
比较例2
向猪用饲料(日配哺乳期幼猪喂养用配合饲料コロミ-ルGS、日本配合饲料(株)制)中分别添加相当于饲料重量2%的富马酸(武田药品工业(株)制)。
试验例1
用实施例1~5和比较例1和2的饲料给各组4头(雄2头和雌2头)的幼猪(约35日龄的杂交种猪)连续喂养4周。在喂养开始时、喂养开始第2周、喂养结束时共分3次测定各个幼猪的体重,算出各组幼猪的体重增加量。另外,对各组幼猪分别调查在测量各个幼猪重量的当天的饲料摄取量,然后根据体重的总增加量和饲料的总摄取量来算出饲料的要求率。如此反复进行4次,算出各饲料供应组的体重增加量和饲料要求率的4次反复的平均值。结果示于下面的表1和图1中。

从示出的结果可以看出,与不添加葡糖酸钠的情况相比,在喂养添加了0.0625%~1.0%葡糖酸钠的饲料的情况下,猪的体重增大了2~12%,饲料效率改善了5~13%。
实施例6
在幼猪用饲料(幼猪饲养用配合饲料“幼猪顺利”(子豚とんとん),日本配合饲料(株)制)中添加了相当于饲料重量0.5%的葡糖酸钠。
实施例7
在幼猪用饲料(幼猪饲养用配合饲料“幼猪顺利”,日本配合饲料(株)制)中添加了相当于饲料重量0.5%的葡聚糖-δ-内酯。
实施例8
在幼猪用饲料(幼猪饲养用配合饲料“幼猪顺利”,日本配合饲料(株)制)中添加了相当于饲料重量0.5%的葡糖酸钙。
实施例9
将葡糖酸钠添加并溶解于幼猪在8小时/天(9:00~17:00)内需饮用量的自来水中,以便使小猪通过饮水对葡糖酸钠的摄取量达到与在饲料中添加0.5%葡糖酸钠时所摄取的量相等(200mg/kg·体重/天)。在其他时间则只喂饮不添加葡糖酸钠的自来水,如此循环地操作。
比较例3
把幼猪用饲料(幼猪饲养用配合饲料“幼猪顺利”,日本配合饲料(株)制)直接作为饲料。
试验例2
用实施例6~8和比较例3的饲料以及实施例9的饮用水向各组6头(雄3头和雌3头)的幼猪(约2月龄的三元杂交种)连续喂养8周。在喂养开始时、喂养开始第2周、喂养开始第4周、喂养开始第6周和喂养结束时共分5次测定各个幼猪的体重,算出各组幼猪的体重增加量。
另外,对各组幼猪分别调查在测量各个幼猪重量的当天的饲料摄取量,然后根据体重的总增加量和饲料的总摄取量来算出饲料的要求率。
将此操作在实施例6中重复3次,在实施例7中重复1次、在实施例8和9中各重复1次,在比较例3中重复2次,算出各饲料供应组的体重增加量和饲料要求率的平均值。结果示于表2中。

如表2所示,投喂了一种葡糖酸钠、葡糖酸钙或葡聚糖-δ-内酯的添加量为0.5%的饲料或饮料水的情况,与不添加葡糖酸类的情况相比,猪的体重增大了10~18%,饲料要求率也改善了4~14%。
实施例10
在鸡用标准试验饲料(ブロイラ-试验用标准饲料SDB No.1和SDB No.2,日本配合饲料(株)制)中添加相当于饲料重量0.5%的葡糖酸钠。
实施例11
根据鸡需要饮用相当于鸡饲料3~4倍的水这一事实,于是向自来水中添加相当于自来水重量0.14%(相当于向饲料中添加0.5%)的葡糖酸钠。
比较例4
将鸡用标准试验饲料(ブロイラ-试验用标准饲料SDB No.1和SDB No.2,日本配合饲料(株)制)直接作为饲料。应予说明,在该饲料中,在ブロイラ-试验用标准饲料SDB No.1(ブロイラ-育肥前期用标准饲料)和ブロイラ-试验用标准饲料SDB No.2(ブロイラ-育肥后期用标准饲料)中含有0.1%的葡糖酸钠。
试验例3
将实施例10和比较例4的饲料以及按实施例11配制的饮料水给各组50只(雄25只和雌25只)的初生小鸡(ブロイラ-专用种,チヤンキ-)连续喂养8周。在喂养开始时、喂养开始第2周、喂养开始第4周、喂养开始第6周和喂养结束时共分5次测定各只鸡的体重,算出各组鸡的体重增加量。另外,对各组鸡分别调查在测量各个鸡重量的当天的饲料摄取量,然后根据体重的总增加量和饲料的总摄取量来算出饲料的要求率。结果示于表3中。

如表3所示,在喂给葡糖酸钠的含量为0.5的饲料或添加量为0.14%的饮料水的情况下,与不添加葡糖酸钠的情况相比,鸡的体重增大了6~10%,饲料要求率也改善了6%。
试验例4
在猪用饲料的人工乳后期((社)日本科学饲料协会制)用饲料和幼猪育成用饲料((社)日本科学饲料协会制)中添加相当于饲料重量的0.05、0.1、0.25、0.5和1.0%的葡糖酸钠。
将上述饲料给各组4头(雄2头和雌2头)的幼猪(体重约20kg)连续喂养8周(最初的2周内喂给人工乳后期用饲料,在随后的6周内喂给幼猪育成用饲料)。从喂养开始时起,每周都测定各个猪的体重,据此算出各组猪的体重增加。另外,对各组幼猪分别调查在测量各个幼猪重量的当天的饲料摄取量,并算出饲料的要求率。
将上述操作对喂养无添加剂饲料和添加剂含量为0.5%的饲料的幼猪组反复进行10次,而对添加剂为其它含量的幼猪组则反复进行5次,算出各组猪的体重增加和饲料要求率的平均值。结果示于表4和图2中。

如结果所示的那样,在喂给添加了0.05~1.0重量%葡糖酸钠的饲料的情况下,与没有添加葡糖酸钠的情况相比,猪的体重增大了2~7%。另外,与没有向饲料中添加葡糖酸钠的情况相比,饲料要求率改善了3~6%。
实施例12
从3头经过一周时间环境驯化的猪的大肠中取出内容物,用厌氧性磷酸缓冲液(50mM,pH6.5)稀释5倍,然后用4重纱布过滤。将25ml滤液加入到一个120ml容量的陶瓷瓶中,用氮气/二氧化碳(80∶20)的混合气体置换陶瓷瓶中的气相,用丁基橡胶栓和氧化铝密封料进行密封,然后在39℃的温度下静置培养(n=3)24小时。
应予说明,在加入大肠滤液之前,除了无添加剂的对照组(阴性对照)之外,其余向陶瓷瓶内添加了葡萄糖1w/v%(阳性对照)或葡糖酸钠1w/v%。
培养结束后,加入6N盐酸1ml,在停止发酵之后,进行以下的测定。
浓度:按照Ushida和Sakata(《动物科学技术(Anim.Sci.Technol.)》,69,571~575,1988)的条件进行前处理,短链脂肪酸、琥珀酸和乳酸的含量用HPLC法测定,而氨的含量用靛酚法(Weatherburn,AnalyticChem.,39,971~974,1967)测定,减去在培养前的测定值,将所获差值作为发生量。其结果示于表5中。

如表5所示,葡糖酸钠添加组,与无添加组相比,乙酸、丙酸、丁酸的浓度大幅地增加。即便与葡萄糖添加组相比,其效果也在同等以上,特别是丁酸的浓度可以观察到明显的增加。另外,在葡糖酸钠添加组中,由乳酸、琥珀酸、氨这些有害成分构成的物质可观察到有大幅度的减少。
实施例13
发酵气体的产生量按下述方法测定,即,使用一个装有注射针的注射器刺入在实施例12的培养步骤中使用的陶瓷瓶的顶部空间,使气体吸入到注射器中直至气体的压力变成常压,然后测定这时的注射器的刻度。
另外,气体组成以二氧化碳、甲烷、氢和硫化氢作为对象,使用TCD和FPD气体色谱法(Ushida等,日本,《动物技术科学杂志(J.Zootech.Sci.)》,53期,412~416(1982)和《动物科学技术(Anim.Sci.Technol.)》,69期,571~575(1998))进行测定。结果示于表6中(n=3,μl/g)。

如表6所示,葡糖酸钠的添加组,与无添加组相比,可以看出作为有害物质的硫化氢大幅度地减少。另外还可看出,氢气和甲烷的生成量有所增加,这表明由于微生物引起的厌氧发酵变得活跃。
实施例14
从经过一周时间环境驯化的猪的大肠中取出内容物,用厌氧性磷酸缓冲液(50mM,pH6.5)稀释5倍,然后用4重纱布过滤。将1ml滤液接种到添加有1w/v%葡糖酸钠的PYF培养基(100ml)中,在39℃的厌氧条件下进行培养。
在培养期间内,每隔48小时注入一次10w/v%的葡糖酸钠水溶液5ml。
对无添加的和添加葡糖酸钠的培养物在第0、第8、第24、第48、第96和第144小时进行无菌取样,将其稀释至10-5,将该溶液0.05ml接种(コンラ-ジ)于BL琼脂培养基(ニツスイ制)上,在37℃下进行厌氧培养48小时,测定在琼脂平板上繁殖的厌氧性菌落数(表7)。
同时,根据繁殖的菌落的性状,把在第144小时之时只在接种有葡糖酸钠添加培养物的琼脂平板上看出有繁殖的菌株分离出来,进行菌数的测定和鉴定(表8)。

如表7和表8所示,可以看出,由于添加了葡糖酸钠而使总厌氧性菌数增加,特别是可以看出埃氏巨球形菌和多酸光岗菌的增殖。
实施例15
把在实施例14中可以看出增殖的菌株埃氏巨球形菌和多酸光岗菌,按照与实施例12同样的方法,在添加了1%w/v葡糖酸钠的PYF培养基(100ml)中于39℃进行24小时的厌氧培养和对培养液中的短链脂肪酸浓度(mM)的测定。结果示于表9中。

如表9所示,对于上述两个菌种,都可以利用葡糖酸钠来产生各种短链脂肪酸。特别是埃氏巨球形菌可以产生乙酸和丁酸,而多酸光岗菌可以产生大量的乙酸和丙酸。
根据本发明,可以提供一种由含有来源于己糖的酸、其非毒性盐及其分子内酯化合物中的至少一种作为有效成分构成的动物育肥促进剂及使用这种动物育肥促进剂的育肥促进方法。
该育肥促进剂可促使肠内细菌增殖,只生成对生物体的生长有用的短链脂肪酸,即使是营养价值低的饲料,通过向其中添加上述组合物,也能使其变成营养价值高的饲料并能产生同等的饲料效率,从而可以促进动物的育肥。

1.一种动物用饮料水,其特征在于,添加0.05~2.0重量%生物体内有害物质的生成抑制剂而构成的,其中所述生物体内有害物质的生成抑制剂含有来源于己糖的酸、其非毒性盐及其分子内酯化合物中的至少一种作为对生物体内的有害物质具有生成抑制功能的有效成分。
2.如权利要求1所述动物用饮料水,其中上述生物体内有害物质是氨、硫化氢、乳酸或琥珀酸。
3.如权利要求1或2所述动物用饮料水,其中上述来源于己糖的酸为葡糖酸。
4.如权利要求1至3任意一项所述动物用饮料水,其中上述葡糖酸的非毒性盐及其分子内酯化合物是葡糖酸钠、葡糖酸钙或葡聚糖酸-δ-内酯。
5.来源于己糖的酸、其非毒性盐及其分子内酯化合物中的至少一种在制造生物体内有害物质的生成抑制剂中的应用。
6.如权利要求5所述应用,其中上述生物体内有害物质是氨、硫化氢、乳酸或琥珀酸。
7.如权利要求5或6所述应用,其中上述来源于己糖的酸为葡糖酸。
8.如权利要求7所述应用,其中上述葡糖酸的非毒性盐及其分子内酯化合物是葡糖酸钠、葡糖酸钙或葡聚糖酸-δ-内酯。
