Abstract
本发明葛根素及其提取方法，其提取工艺流程为：原料葛根清杂粉碎→水煎煮→水浸膏→乙醇沉淀→醇浸膏→聚酰胺柱层析→水洗脱→浓缩→放置析晶→粗品→精制→干燥→葛根素精品。经此方法提取的葛根素精品含量达97-100％，收率达1％以上，工艺简单、成本降低，便于扩大生产。
Description
本发明为中药有效成份提取物，特别涉及的是从中药葛根中提取的尤其对心脑血管病有明显疗效的葛根素及其提取方法。
葛根作为中药有悠久的历史，药典中记载葛根性味甘、辛、凉，归脾、胃经，能解肌退热、生津、透疹、升阳止泻，用于外感发热头痛、项强、口渴、消渴、麻疹不透、热痢、泄泻、高血压、颈项强痛。现代药理学表明，葛根中所含以葛根素为主的黄酮类化合物能扩张心脑血管，对冠心病、心绞痛、高血压及缺血性脑血管病有显著疗效。我省八十年代曾将葛根有效成份的研究列入省科技计划，取得一定成绩，使葛根素成为一种治疗心血管病的新药面世。针对工业化生产葛根素存在的污染严重、含量低、收得率低等问题，本人于95年2月23日申请专利《葛根素及其提取方法和用途》，申请号为“95110343.1”这一专利技术解决了原工艺大量使用易燃及有毒的有机溶媒所导致的环境污染等问题，并将葛根素原料药的含量从不足80％提高到90％以上，根据实际需要葛根素的质量仍需继续提高。
本发明的目的是提供一种利用中药葛根提取的纯度为97-100％，收率为1％以上，提取方法简便可靠的葛根素及其提取方法。
本发明的目的是这样实现的：参照附图说明如下：
一种葛根素的提取方法，其提取的工艺流程为：将原料葛根粉碎→水煎煮→水浸膏→乙醇沉淀→醇浸膏→聚酰胺柱层析→水洗脱→浓缩→放置析晶→粗品→精制→干燥→葛根素精品。
附图为葛根素提取工艺流程方框图。
附图中葛根清杂粉碎系指将原料清除杂物，加工粉碎。
附图中水煎煮工序系指将葛根加水煎煮2-4遍，浓缩煎煮液至稠膏状；
附图中乙醇沉淀系指将乙醇加入水浸膏中使含醇量达70-80％，加热至微沸，搅拌，放置，过滤，回收乙醇，浓缩制得醇浸膏；
附图中柱层析所采用的吸附剂为聚酰胺类物质；
附图中水洗脱系指聚酰胺柱层析的洗脱溶媒为水或热水或PH为2-8的水或含有乙醇的水；
附图中所示柱层析-水洗脱-浓缩-放置析晶-粗品的工艺流程是指：聚酰胺柱层析为数根柱连续层析，是将已制得的醇浸膏加入到一组以管道首尾联连的层析柱首端，用水洗脱，通过薄层检测进行连续层析，收集未端的洗脱液，浓缩后放置析晶，得葛根素粗品；
附图中葛根素粗品提取中柱层析的条件要求是指：层析用聚酰胺为14-200目，总用量与生药量之比为1∶1-1∶3，柱径高比为1∶2-1∶10，层析柱根数为2-8根；
附图中精制系指将所得粗品经下列三种方法之一处理后可得葛根素精品：①将粗品溶解后再次进行聚酰胺柱层析，洗脱溶媒为去离子水或蒸馏水，选择收集洗脱液，浓缩并放置析晶，滤取结晶，干燥、制得葛根素精品；精制用聚酰胺为60-200目，总用量与粗品之比为5∶1-20∶1，柱径高比为1∶2-1-10，层析柱根数常用的为1-3根；②将葛根素粗品加热溶解于适量水中，放置析晶，滤取结晶，如此反复重结晶1-5次后，干燥，制得葛根素精品；③将干燥的葛根素粗品加热溶解于含乙醇的水溶液中，过滤，放置析晶，滤取结晶，干燥，制得葛根素精品。
附图中干燥系指葛根素的干燥除一般干燥方法外，还有冷冻干燥、喷雾干燥和微波干燥。
上述方法提取的葛根素不仅可以用于防治心血管疾病，对脑血管病也有明显疗效。本发明提取的中间体均可作药用。
葛根素的提取方法实施例如下：
实施例1
①将葛根原料清杂、洗净、粉碎成粗粉；
②将粗粉水煎3次，每次0.5小时，过滤、浓缩、制成稠膏；
③加乙醇使含醇量达70％，加热并保持微沸0.5小时，间歇搅拌，放置，过滤，回收乙醇，制得葛根醇浸膏；
④将葛根醇浸膏进行连续柱层析，聚酰胺为30-80目，总用量为生药量的2/3，柱径高比为1∶5，层析柱为4根，以常水洗脱，薄层检测，收集未端洗脱液，浓缩，过滤，低温(0℃左右)放置析晶，滤取结晶，微波干燥，得类白色葛根素粗品，可用于配制各种口服制剂。
实施例2
将葛根醇浸膏进行连续柱层析的另一实例是：聚酰胺为60-100目，总用量与生药量之比为1∶1，柱径高比为1∶4，层析柱根数为7根，以去离子水洗脱，薄层检测，收集未端洗脱液，浓缩，趁热过滤，缓慢降温至35℃左右，放置析晶，滤取结晶，得白色葛根素粗品。
实施例3
葛根素粗品的精制方法为：将粗品溶解后加到一组聚酰胺层析柱首端，用去离子水洗脱，收集洗脱液，浓缩过滤，滤液加热至沸，放置析晶，滤取结晶，真空干燥，得葛根素精品，层析用聚酰胺为60-80目，总用量与粗品之比为15∶1，柱径高比为1∶5，层析柱根数为3根。
实施例4
葛根素粗品的精制的另一方法为：将粗品加2-3倍量水加热溶解，趁热过滤，缓慢降温至40℃左右，放置析晶，滤取结晶，再加3倍量水加热溶解，重复结晶一次，滤取结晶，80℃烘干，得葛根素精品。
实施例5
葛根素粗品精制的另一方法是将干燥的粗品溶解于适量的70％乙醇溶液中，过滤，放置析晶，滤取结晶，真空干燥，得葛根素精品。
本发明的优点很多，首先是纯度高，提取的葛根素纯度高达100％；工艺流程中不用有害物质，所以粗品即可作成各种口服制剂；精品可作成针剂；由于纯度高，可作为分析用试剂；由于本工艺简单、易行、无三废、收率高达1％以上，所以为葛根素的大生产和降低成本创造了目前最好的条件。
Claims (10)
1、一种葛根素的提取方法，其特征在于：其提取的工艺流程为：将原料葛根清杂粉碎→水煎煮→水浸膏→乙醇沉淀→醇浸膏→聚酰胺柱层析→水洗脱→浓缩→放置析晶→粗品→精制→干燥→葛根素精品。
2、根据权利要求1所述的葛根素提取方法，其特征在于：柱层析所用吸附剂为聚酰胺类物质。
3、根据权利要求1所述的葛根素提取方法，其特征在于：聚酰胺柱层析洗脱溶媒为水、热水、PH值为2-8的水，含有乙醇的水；
4、根据权利要求1所述的葛根素提取方法，其特征在于：工艺流程中聚酰胺柱层析→水洗脱→浓缩→放置析晶→粗品系指：聚酰胺柱层析为1根以上的柱连续层析，即将葛根醇浸膏加入到一组以管道首尾联接的层析柱首端，用水洗脱，通过薄层检测进行连续柱层析，收集末端的洗脱液、浓缩、放置折晶，得葛根素粗品。
5、根据权利要求4所述的葛根素提取方法，其特征在于：粗品提取的柱层析的条件要求为；层析用聚酰胺为14-200目，总用量与生药量之比为1∶1-1∶3，柱径高比为1∶2-1∶10，层析柱根数为2-8根。
6、根据权利要求1所述的葛根素提取方法，其特征在于：葛根素粗品的精制方法为：将粗品溶解后再次进行聚酰胺柱层析，洗脱溶媒为去离子水或蒸馏水，选择收集洗脱液，浓缩并放置析晶，滤取结晶，干燥、制得葛根素精品；精制用聚酰胺为60-200目，总用量与粗品之比为5∶1-20∶1，柱径高比为1∶2-1∶10，层析柱根数常用的为1-3根。
7、根据权利要求1所述的葛根素提取方法，其特征在于：葛根素粗品的精制方法为：葛根素粗品加热溶解于适量水中，放置析晶，滤取结晶，如此反复重结晶1-5次后，干燥，制得葛根精品。
8、根据权利要求1所述的葛根素提取方法，其特征在于：葛根素粗品的精制方法为：将干燥的葛根素粗品加热溶解于含乙醇的水溶液中过滤，放置析晶，滤取结晶，干燥，制得葛根素精品。
9、根据权利要求1所述的葛根素提取方法，其特征在于：葛根素的干燥除一般干燥方法外，还有冷冻干燥，喷雾干燥和微波干燥。
10、一种用权利要求1所述的葛根素提取方法所得到的葛根素，其特征在于：它是一种从中药葛根中提取的天然化合物，提取的中间粗品为无毒液体、浸膏为棕黄色直至白色的粗品，经精制后为白色结晶或粉末，其葛根素(C21H20O9)含量达97-100％，收得率达1％以上。
