贫血是指人体外周血红细胞容量臧少,低于正常范围下限的一种常见的临床症状。由于红细胞容量测定较复杂,临床上常以血红蛋白(Hb)浓度来代替。贫血发生的主要原因为:(1)缺铁;( 2 ) EPO生成减少;(3)代谢毒物对红细胞的增殖过程产 生抑制作用;(4) 炎症因子加快清除循环中抗体或免疫复合物包裹的红细胞,使其寿命缩短; (5) CKD患者体内活性氧产生增多使红细胞易发生溶血等。临床表现:(1)神经系统:头昏、耳鸣、头痛、失眠、多梦、记忆减退、注意力不集中等,乃是贫血缺氧导致神经组织损害所致常见的症状。小儿贫血时可哭闹不安、躁动甚至影响智力发育。(2)皮肤黏膜:苍白是贫血时皮肤、黏膜的主要表现。贫血时机体通过神经体液调节进行有效血容量重新分配,相对次要脏器如皮肤、黏膜则供血减少;另外,由于单位容积血液内红细胞和血红蛋白含量减少,也会引起皮肤、黏膜颜色变淡。粗糙、缺少光泽甚至形成溃疡是贫血时皮肤、黏膜的另一类表现,可能还与贫血的原发病有关。溶血性贫血,特别是血管外溶血性贫血,可引起皮肤、黏膜黄染。(3)呼吸循环系统:贫血时红细胞内合成较多的2,3-二磷酸甘油酸(2,3-DPG),以降低血红蛋白对氧的亲和力,使氧解离曲线右移,组织获得更多的氧。气急或呼吸困难,大都是由于呼吸中枢低氧或高碳酸血症所致。故轻度贫血无明显表现,仅活动后引起呼吸加快加深并有心悸、心率加快。贫血愈重,活动量愈大,症状愈明显。重度贫血时,即使平静状态也可能有气短甚至端坐呼吸。长期贫血,心脏超负荷工作且供氧不足,会导致贫血性心脏病,此时不仅有心率变化,还可有心律失常和心功能不全。(4)消化系统:贫血时消化腺分泌减少甚至腺体萎缩,进而导致消化功能减低、消化不良,出现腹部胀满、食欲减低、大便规律和性状的改变等。长期慢性溶血可合并胆道结石和脾大。缺铁性贫血可有吞咽异物感或异嗜症。巨幼细胞贫血或恶性贫血可引起舌炎、舌萎缩、牛肉舌、镜面舌等。
复方红衣口服液是市场常用的补血药物,具备功效稳定、毒副作用小等优点。申请人之前的专利技术2008101590994和2015101684898均涉及复方红衣口服液的改进,其中专利技术2008101590994涉及对提取工艺的改进,通过对不同原料提取工艺的改进,提高了药效,但其中,将花生红衣通过所述工艺可以获得较高量的原花青素、白藜芦醇等,但是枸杞子、大枣不易采用上述工艺,会造成多糖有效成分的流失;专利技术2015101684898在已有配方的基础上,添加了多种中草药成本,但是该方法大幅度增加了原料成本。
发明内容
为了克服现有技术的缺陷,本发明提供了复方红衣补血口服液的制备工艺。
本发明是通过如下技术方案来实现的:
复方红衣补血口服液的制备工艺,其包括如下步骤:步骤1)制备木耳提取液,步骤2)制备枸杞子提取物,步骤3)制备大枣提取物,步骤4)制备花生红衣提取液,步骤5)煮沸、滤过、分装以及灭菌。
具体地,所述步骤1)制备木耳提取液,包括如下步骤:取木耳20g,加16倍重量的水,浸泡30分钟,加热微沸2小时,滤过,药渣再加10倍重量的水,加热微沸1小时,滤过,合并滤液,浓缩至密度为1.02g/ml,得到木耳提取液。
具体地,所述步骤2)制备枸杞子提取物,包括如下步骤:取枸杞子100g,粉碎,然后加入5倍重量的水,用高速剪切机剪切60min获得浸泡液,剪切转速为5000rpm;将浸泡液置于提取罐中,添加两倍体积的60%乙醇,并且控制压强为0.8Mpa,温度为60-70℃,300rpm搅拌提取120min,过滤收集滤液,然后对滤液进行减压蒸发去除乙醇,收集残余液,干燥6-9h,干燥温度70℃,得到枸杞子提取物。
具体地,所述步骤3)制备大枣提取物,包括如下步骤:取大枣200g,粉碎,过100目筛,然后添加到反应釜中,再添加两倍重量的水,搅拌均匀,加热至60-70℃,并且控制压强为1-2Mpa,保温保压30min,然后降压至常压,保温条件下添加1-2wt%的活性炭,200rpm搅拌10min,过滤收集滤液,然后添加占滤液两倍体积的无水乙醇进行醇沉,静置6-9h,收集沉淀,干燥,得到大枣提取物。
具体地,所述步骤4)制备花生红衣提取液,包括如下步骤:取花生红衣100g,加水煎煮两次,第一次加10倍重量的水,第二次加8倍重量的水,每次加热微沸1小时,滤过,合并煎液,滤液浓缩至密度为1.07g/ml,再加无水乙醇使浓缩液中乙醇含量体积比为70%,冷藏24小时,滤过,滤液回收乙醇,补加等重量的水,搅匀,静置8小时,滤过,收集滤液,即得花生红衣提取液。
具体地,所述步骤5)煮沸、滤过、分装以及灭菌,包括如下步骤:往步骤4)所得花生红衣提取液中加入木耳提取液、枸杞子提取物、大枣提取物、蜂蜜30g和蔗糖140g,煮沸20min,滤过,滤液放冷至室温再加入柠檬酸2g,加水调至1000ml,分装,灭菌,即得。
本发明复方红衣补血口服液,每支装10ml;口服。一次10ml,一日3次。
本发明取得的有益效果主要包括但是并不限于以下几个方面:
现有技术中采用将红衣、枸杞子以及大枣混合,一并煮沸取其过滤液,没有考虑到各种药物组分的制备参数存在较大差异,导致制备出的补血口服液的营养成分流失,不能达到最佳的治疗效果;传统水煎法虽然工艺简单,易于操作,但是其提取效率低,并且过高的温度容易破坏或分解多糖的结构,降低多糖的生活活性;通过两次煎煮并且合理地控制煎煮时间,可以提取和保留了花生红衣原花青素、白藜芦醇等物质的生物学活性,同时大大提高了上述物质的释放率;但是该工艺并不适合大枣和枸杞子。
本发明制备方法简单可行,对不同的中药成分采用不同技术进行处理,提高了各原料的有效成分,增加了药效,减少了原料浪费;本发明应用了高速剪切技术提取枸杞多糖,高速剪切破除了枸杞表面的蜡质层和细胞壁,辅助适合强度的高压,使枸杞多糖最大程度得释放出来,极大地提高了枸杞多糖的提取率,同时明显缩短了提取时间;本发明大枣提取方法采用破碎成合适粒径进行辅助破壁,提高了有效成分的溶出度,同时采用高压以及适宜温度辅助水提,时间短效率高,避免采用昂贵的酶处理,提高了多糖提取率,降低了能耗和企业成本;本发明方法独特,是针对现有技术已知药物的工艺改进,避免了原料浪费,提高了药效,优于现有技术已知的产品和工艺,具备较好的应用前景。
具体实施方式
为了使本技术领域的人员更好地理解本申请中的技术方案,下面将结合本申请具体实施例,对本发明进行更加清楚、完整地描述,显然,所描述的实施例仅仅是本申请一部分实施例,而不是全部的实施例。基于本申请中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其他实施例,都应当属于本发明保护的范围。
实施例1
复方红衣补血口服液的制备工艺,其包括如下步骤:
1)取木耳20g,加16倍重量的水,浸泡30分钟,加热微沸2小时,滤过,药渣再加10倍重量的水,加热微沸1小时,滤过,合并滤液,浓缩至密度为1.02g/ml,得到木耳提取液;
2)取枸杞子100g,粉碎,然后加入5倍重量的水,用高速剪切机剪切60min获得浸泡液,剪切转速为5000rpm;将浸泡液置于提取罐中,添加两倍体积的60%乙醇,并且控制压强为0.8Mpa,温度为60℃,300rpm搅拌提取120min,过滤收集滤液,然后对滤液进行减压蒸发去除乙醇,收集残余液,干燥6h,干燥温度70℃,得到枸杞子提取物1.79g;采用用苯酚硫酸法进行含量测定,枸杞多糖的含量为75%;
3)取大枣200g,粉碎,过100目筛,然后添加到反应釜中,再添加两倍重量的水,搅拌均匀,加热至60℃,并且控制压强为1Mpa,保温保压30min,然后降压至常压,保温条件下添加1wt%的活性炭,200rpm搅拌10min,过滤收集滤液,然后添加占滤液两倍体积的无水乙醇进行醇沉,静置8h,收集沉淀,干燥,得到大枣提取物5.1g;利用苯酚-硫酸法测得固体物中多糖含量为83%;
4)取花生红衣100g,加水煎煮两次,第一次加10倍重量的水,第二次加8倍重量的水,每次加热微沸1小时,滤过,合并煎液,滤液浓缩至密度为1.07g/ml,再加无水乙醇使浓缩液中乙醇含量体积比为70%,冷藏24小时,滤过,滤液回收乙醇,补加等重量的水,搅匀,静置8小时,滤过,收集滤液,即得花生红衣提取液;
5)往步骤4)所得花生红衣提取液中加入木耳提取液、枸杞子提取物、大枣提取物、蜂蜜30g和蔗糖140g,煮沸20分钟,滤过,滤液放冷至室温再加入柠檬酸2g,加水调至1000ml,分装,灭菌,即得。
本发明复方红衣补血口服液,每支装10ml;口服。一次10ml,一日3次。
对比例1
取木耳20g加16倍重量的水,浸泡30分钟,加热微沸2小时,滤过,药渣再加10倍重量的水,加热微沸1小时,滤过,合并滤液,浓缩至密度为1.02;取花生红衣100g、枸杞子100g、大枣200g,加水煎煮两次,第一次加10倍重量的水,第二次加8倍重量的水,每次加热微沸1小时,滤过,合并煎液,滤液浓缩至密度为1.07,再加乙醇使浓缩液中乙醇含量重量比达65%,冷藏24小时,滤过,滤液回收乙醇,补加等重量的水,搅匀,静置8小时,滤过;然后在滤液中加入木耳浓缩液、蜂蜜30g和蔗糖140g,煮沸20分钟,滤过,滤液放冷至室温再加入柠檬酸2g,加水调至1000ml,分装,灭菌,即得。
用法和用量:每支装10ml;口服。一次10ml,一日3次。
实施例2
提取工艺对多糖有效物质的影响:
以对照例1为例,将枸杞子和大枣分别采用对照例1的工艺,苯酚-硫酸法测定多糖的含量,检测了不同工艺对多糖产率的影响;具体见表1:

实施例3
毒性试验:
本实验采用健康SD大鼠,口服给药实施例1制备的复方红衣补血口服液剂量梯度均为1ml/kg以及5ml/kg,连续给药45天,经观察大鼠给药期间及停药后1周之内动物生长状态、活动饮食、血液学、血液生化学、脏器组织结构以及尿常规等均无明显影响,表明临床用药安全可靠。
实施例4
动物学实验:
所选用的小鼠为SD大鼠50只;剂量选择:1ml/kg。
模型建立:将SD大鼠置于实验室环境适应5d后,10只给予正常大鼠基础饲料和正常饮用水,作为空白对照组,其余40只给予低铁饲料及饮用水,20d后采尾血检测血红蛋白(Hb),将血红蛋白(Hb)低于100g/L的大鼠作为营养性贫血模型动物。选择建模成功的营养性贫血模型动物30只;随机平均分为三组,分别是模型对照组,实施例1组,对比例1组。模型对照组给与相同体积的生理盐水,实施例1组和对比例1组给与药物;连续30d,用分光光度计测定血红蛋白(Hb);具体见表2。

实施例5
临床试验:
对象:全部病历均为市中医院2015-2016年门诊患者共计79例,其中实施例1组39例,对比例1组40例,两组病例在性别、年龄、病程等方面经统计学分析,具有可比性。全部病人均是由于血虚或气虚引起的面色苍白或微黄,倦怠乏力,心悸怔忡,失眠多梦等症状。
服药方法及疗程:
实施例1组,给予实施例1制备的药物制剂,每支装10ml;口服。一次10ml,一日3次,四周为一个疗程;
对比例1组,给予对比例1制备的药物制剂,每支装10ml;口服。一次10ml,一日3次,四周为一个疗程。
疗效评定标准:显著、有效和无效。
治愈:临床症状消除,无反弹。
显效:临床症状虽已消除,但疗效不够巩固。
有效:临床症状及体征有所改善。
无效:治疗前无变化。
具体结果见表3 表3

由上表可见:实施例1组的治愈率和总有效率均明显优于对比例1组,有显著性差异。
以上列举的仅是本发明的最佳具体实施例。显然,本发明不限于以上实施例,还可以有许多变形。本领域的普通技术人员能从本发明公开的内容直接导出或联想到的所有变形,均应认为是本发明的保护范围。
Claims (6)
1.复方红衣补血口服液的制备工艺,其包括如下步骤:步骤1)制备木耳提取液,步骤2)制备枸杞子提取物,步骤3)制备大枣提取物,步骤4)制备花生红衣提取液,步骤5)煮沸、滤过、分装以及灭菌。
2.根据权利要求1所述的制备工艺,其特征在于,所述步骤1)制备木耳提取液,包括如下步骤:取木耳20g,加16倍重量的水,浸泡30分钟,加热微沸2小时,滤过,药渣再加10倍重量的水,加热微沸1小时,滤过,合并滤液,浓缩至密度为1.02g/ml,得到木耳提取液。
3.根据权利要求1或2所述的制备工艺,其特征在于,所述步骤2)制备枸杞子提取物,包括如下步骤:取枸杞子100g,粉碎,然后加入5倍重量的水,用高速剪切机剪切60min获得浸泡液,剪切转速为5000rpm;将浸泡液置于提取罐中,添加两倍体积的60%乙醇,并且控制压强为0.8Mpa,温度为60-70℃,300rpm搅拌提取120min,过滤收集滤液,然后对滤液进行减压蒸发去除乙醇,收集残余液,干燥6-9h,干燥温度70℃,得到枸杞子提取物。
4.根据权利要求1-3任其一的制备工艺,其特征在于,所述步骤3)制备大枣提取物,包括如下步骤:取大枣200g,粉碎,过100目筛,然后添加到反应釜中,再添加两倍重量的水,搅拌均匀,加热至60-70℃,并且控制压强为1-2Mpa,保温保压30min,然后降压至常压,保温条件下添加1-2wt%的活性炭,200rpm搅拌10min,过滤收集滤液,然后添加占滤液两倍体积的无水乙醇进行醇沉,静置6-9h,收集沉淀,干燥,得到大枣提取物。
5.根据权利要求1-4任其一的制备工艺,其特征在于,所述步骤4)制备花生红衣提取液,包括如下步骤:取花生红衣100g,加水煎煮两次,第一次加10倍重量的水,第二次加8倍重量的水,每次加热微沸1小时,滤过,合并煎液,滤液浓缩至密度为1.07g/ml,再加无水乙醇使浓缩液中乙醇含量体积比为70%,冷藏24小时,滤过,滤液回收乙醇,补加等重量的水,搅匀,静置8小时,滤过,收集滤液,即得花生红衣提取液。
6.根据权利要求1-5任其一的制备工艺,其特征在于,所述步骤5)煮沸、滤过、分装以及灭菌,包括如下步骤:往步骤4)所得花生红衣提取液中加入木耳提取液、枸杞子提取物、大枣提取物、蜂蜜30g和蔗糖140g,煮沸20min,滤过,滤液放冷至室温再加入柠檬酸2g,加水调至1000ml,分装,灭菌,即得。
