Abstract

背景技术
诸如牙膏的口腔护理组合物通常有两种类型:不透明的和透明的。不透明的牙膏通常含有基于钙的磨料,诸如碳酸钙。相比之下,透明的(例如凝胶)牙膏通常含有二氧化硅作为磨料。这些制剂中的任一者可以有益地包含抗菌剂,诸如5-氯-2-(2,4-二氯苯氧基)苯酚(三氯生)。然而,抗菌剂的递送通常需要使用乙烯基甲基醚和马来酸聚合物,例如GANTREZTM。广泛用于基于二氧化硅的牙膏的此类聚合物通过例如截留抗菌剂然后将其粘附到牙齿和牙龈来提高抗菌剂的吸收率和保持率。然而,此类聚合物的用途仅限于基于钙的洁齿剂,因为它们可导致钙离子螯合和沉淀,如例如WO2013/007571中所述。另外,递送乙烯基甲基醚和马来酸聚合物的媒介物可能是昂贵的,从而降低基于钙的洁齿剂作为大众市场产品的商业可行性。
采用降低量的可能对三氯生递送有效的GANTREZTM的基于钙的磨料制剂是已知的。据报道,这些制剂含有约0.75重量%的GANTREZTM与蒙脱石粘土诸如HV(通常约0.8重量%)的组合;粘度构造剂,诸如羧甲基纤维素;增稠剂二氧化硅,诸如-P;以及碱化剂,诸如用于中和GANTREZTM的硅酸钠(1.75重量%)。
然而,包含甚至更低量的GANTREZTM与HV粘土的组合可能过高地增大制剂成本。另外,已知含有粘土的制剂由于混合和水合要求而增加批次循环时间。因此,本领域需要在不使用GANTREZTM或类似聚合物并且不进一步使用大量粘度改性剂和/或硅酸钠的情况下提高基于钙的口腔护理组合物中的活性成分诸如三氯生的吸收率,以获得可快速且简易制备的具有成本效益的基于钙的口腔护理制剂。
发明内容
本公开涉及一种口腔护理组合物,其包含(i)基于钙的磨料;(ii)聚乙二醇,其平均分子量在约1500至约6000道尔顿的范围内;(iii)抗菌剂以及(iv)口腔可接受的媒介物。
本文还提供了一种制备口腔护理组合物的方法,其包括:将湿润剂和水组合以形成口腔可接受的媒介物;b)将基于钙的磨料加入口腔可接受的媒介物;c)将平均分子量在约1,500至约6,000道尔顿范围内的聚乙二醇、抗菌剂以及任选地选自调味剂和防腐剂的成分组合以形成预混物;d)将步骤c)的预混物加入步骤b)的含有基于钙的磨料的口腔可接受的媒介物以形成口腔护理组合物。
本公开还提供了一种促进口腔健康的方法,该方法包括使口腔表面与包含以下组分的口腔护理组合物接触:(i)基于钙的磨料;(ii)聚乙二醇,其平均分子量在约1500至约6000道尔顿的范围内;(iii)抗菌剂以及(iv)口腔可接受的媒介物。

除非另外规定,否则本文和在本说明书中其他处表达的所有百分比和量应理解为是指重量百分比。给定的量是以材料的有效重量计。
组合物
本发明人意外地认识到,在含有诸如PEG 6000的高分子量聚乙二醇的基于钙的口腔护理组合物中可以获得增大的抗菌剂(诸如三氯生)的保持率和吸收率。本发明人还意外地认识到,在例如小于9.0的pH值下也可获得提高的抗菌剂(诸如三氯生)的吸收率和保持率。此外,有利地可在不需要大量粘度改性剂、与现有技术制剂相比更少或无硅酸钠的情况下以及缩短的批次循环时间内制备本公开的基于钙的口腔护理组合物。
如本文所用的“口腔护理组合物”是指这样的组合物,其在口腔中保留足够的时间以接触基本上所有的牙表面和/或口腔组织,达到口腔活性。本公开的口腔护理组合物可呈各种形式,包括漱口液或洁齿剂。洁齿剂可包括牙用片剂、牙膏(牙霜剂)、牙粉、凝胶、液体、液状凝胶或本领域中技术人员已知的任何其他形式。
基于钙的磨料
在一些实施方案中,本公开的口腔护理组合物包含基于钙的牙科磨料或基于钙的牙科磨料的组合。如本文所用,术语“磨料”或“研磨剂”包括常常称为“抛光剂”的材料。可使用任何口腔可接受的基于钙的磨料,但是通常应选择磨料的类型、细度(粒度)和量使得牙齿釉质在正常使用组合物中不被过度地磨损。合适的基于钙的磨料抛光剂包括沉淀碳酸钙(PCC)、白垩、磷酸二钙及其二水合物形式、焦磷酸钙、磷酸三钙、正磷酸二钙、焦磷酸钙、n-焦磷酸钙(n-calcium pyrophosphate)、天然碳酸钙(NCC)等等。通常,使用PCC。
通常,本公开的口腔护理组合物仅包含基于钙的磨料。然而,还可以想到仅包含基于二氧化硅的磨料的口腔护理组合物。仅包含二氧化硅磨料的制剂还可有利地在如本文所述的合适的pH下与所述聚乙二醇结合使用,从而提高抗菌剂吸收率和保持率。合适的基于二氧化硅的磨料包括但不限于硅胶、水合二氧化硅或沉淀二氧化硅形式的二氧化硅。
其他可以想到的实施方案包括基于钙的磨料和基于二氧化硅的磨料的混合物。此类混合物还可以在如本文所述的合适的pH下与高分子量聚乙二醇、抗菌剂组合,从而提高抗菌剂的吸收率和保持率。
在另外其他可以想到的实施方案中,口腔护理组合物包含多相洁齿剂。此类多相洁齿剂包括例如包含基于钙的磨料的不透明相和包含二氧化硅磨料的凝胶相。不透明相、凝胶相或两个相可以包含一种或多种如本文所述的抗菌剂和聚乙二醇,诸如合适的pH下的PEG-6000,从而提高抗菌剂(诸如三氯生)的吸收率和保持率。
在一些实施方案中,磨料的平均粒度通常为约0.1至约30μm,例如约1至约20μm或约5至约15μm。
在一些实施方案中,本公开的口腔护理制剂中包含量在约5至约75重量%范围内(诸如约30至约60重量%,诸如约35至约55重量%)的一种或多种基于钙的磨料和/或基于二氧化硅的磨料。
聚乙二醇
在一些实施方案中,本公开的口腔护理组合物包含聚乙二醇(PEG),其分子量在约1,500至约6,000道尔顿范围内,诸如约4,000至约6,000道尔顿,诸如约5,000至约6,000道尔顿,更通常约6,000道尔顿。
如本文所述的与本公开的口腔护理组合物一起使用的聚乙二醇的量在约0.1重量%至约0.5重量%范围内,诸如约0.1重量%至约4重量%,诸如约0.1重量%至约0.3重量%。通常,使用约0.3重量%的聚乙二醇,诸如PEG 6000。
在一些实施方案中,与本公开一起使用的分子量在1,500至6,000道尔顿范围内的聚乙二醇在20℃下为固体形式,例如薄片、粉末或蜡状物。合适的市售聚乙二醇聚合物诸如PEG 6000可得自Clariant,India和BASF,USA。其他市售来源可得自例如Sigma-Aldrich,St.Louis,MO,USA;Santa Cruz Biotechnology,Inc.,Dallas Texas,USA和Hallstar,Chicago,Il,USA。
不受理论的约束,据信,本文所述的聚乙二醇(例如PEG-6000)附着到牙齿表面而在该表面上形成连续膜,从而防止细菌附着到牙齿表面。还可以想到,如本文所公开的抗菌剂与聚乙二醇形成络合物或其他形式的缔合,从而在牙齿表面上形成络合物膜等。抗菌剂的递送和保持以及本文所述的聚乙二醇的成膜特性似乎使得牙齿表面不利于细菌积聚,特别是因为与聚乙二醇络合的抗菌剂的直接抑菌作用控制了细菌生长。因此,通过1)增强的递送、2)在牙齿表面上较长的保持时间以及3)防止细菌附着到牙齿表面的组合,本公开的口腔护理组合物能够有效减少牙斑。此外,在不导致钙螯合或沉淀的情况下实现这种牙斑的减少。因此,本公开的聚乙二醇可与包含基于钙的磨料的本公开的口腔护理组合物结合使用。
在一些实施方案中,本公开的口腔护理组合物不含具有不同酯基的聚(甲基乙烯基醚/马来酸)的单烷基酯的共聚物,诸如聚乙烯基甲基醚/马来酸(PVM/MA)共聚物的丁酯、PVM/MA共聚物的异丙酯和PVM/MA共聚物的乙酯,例如可从例如Ashland Inc.Covington,KY商购获得的GANTREZTM系列聚合物。
抗菌剂
在一些实施方案中,本公开的口腔组合物包含抗菌剂。可经由本文所述的聚乙二醇递送和保留的抗菌剂包括卤代二苯醚、酚类化合物、倍半萜烯醇和卤代二苯脲或它们的组合。本公开的口腔护理组合物中存在的抗菌剂的有效抗牙斑量为约0.5重量%至约1.0重量%,诸如约0.25重量%至约0.35重量%,通常约0.3重量%。
与本公开的基于钙的口腔护理组合物一起使用的卤代二苯醚的实例包括但不限于2′,4,4′-三氯-2-羟基二苯醚(三氯生)、2,2′-二羟基-5,5′-二溴-二苯醚、卤代水杨酸苯胺诸如4′5-二溴水杨酸苯胺、3,4′,5-三氯水杨酸苯胺、3,4′,5-三溴水杨酸苯胺、2,3,3′,5-四氯水杨酸苯胺、3,3′,5-四氯水杨酸苯胺、3,5-二溴-3′-三氟甲基水杨酸苯胺、5-正辛酰基-3′-三氟甲基水杨酸苯胺、3,5-二溴-4′-三氟甲基水杨酸苯胺、3,5-二溴-3′-三氟甲基水杨酸苯胺(氟苯)、苯甲酸酯、对羟基苯甲酸甲酯、对羟基苯甲酸乙酯、对羟基苯甲丙酸酯、对羟基苯甲酸丁酯、卤代二苯脲、3,4,4′-三氯二苯脲、3-三氟甲基-4,4′-二氯二苯脲、3,3′,4-三氯二苯脲或它们的组合。
在一些实施方案中,本公开的口腔护理组合物包含酚类化合物和酚类化合物的同系物。实例包括2-甲氧基-4-(2-丙烯基)-苯酚(丁子香酚)、2-异丙基-5-甲基苯酚(麝香草酚)、单烷基卤酚和多烷基卤酚和芳烷基卤酚、甲基对氯苯酚、正丙基对氯苯酚、正丁基对氯苯酚、正戊基对氯苯酚、仲戊基对氯苯酚、正己基对氯苯酚、环己基对氯苯酚、正庚基对氯苯酚、正辛基对氯苯酚、3,4,5,6-四溴-2-甲基苯酚、5-甲基-2-戊基苯酚、4-异丙基-3-甲基苯酚和5-氯-2-羟基二苯基甲烷。
还可以想到倍半萜烯醇,诸如金合欢醇、橙花叔醇、没药醇、檀香醇;以及卤代二苯脲,诸如3,4,4′-三氯二苯脲、3-三氟甲基-4,4′-二氯二苯脲、3,3,4′-三氯二苯脲。与本公开的口腔组合物一起使用的其他理想的抗菌剂在美国专利No.5,728,756中有所描述,该专利以引用方式整体并入本文。通常,本公开的口腔护理组合物中包含三氯生。
pH
在一些实施方案中,可以用酸性材料或碱性材料调节本公开的口腔护理组合物的pH,以实现特定的pH值。在一些实施方案中,使用缓冲剂保持本公开的口腔护理组合物的所需pH范围。
pH调节剂的实例包括柠檬酸、磷酸、苹果酸、焦磷酸、乳酸、酒石酸、甘油磷酸、乙酸、硝酸、硅酸、葡萄糖酸、马来酸、天冬氨酸、琥珀酸、葡萄糖醛酸、富马酸、己二酸、谷氨酸或它们的化学可接受的盐(诸如钠盐和钾盐)、盐酸、氢氧化钠、氢氧化钾等。这些pH调节剂可单独使用或两种或更多种结合使用,以便将本公开的口腔护理组合物的pH调节到上述范围内。
可用于保持本公开的口腔护理组合物的所需pH的合适缓冲剂包括例如可溶性磷酸盐和硅酸钠。在一些实施方案中,使用硅酸钠作为缓冲剂。缓冲剂(通常为硅酸钠)的量通常在本公开的口腔护理组合物的约0.01重量%至5重量%的范围内,诸如1%至3%。
在一些实施方案中,将口腔护理组合物的pH保持在约8.0至约10.5的范围内,诸如约8.5至约9.4。更通常,将含有基于钙的磨料的本公开的组合物的pH保持在低于约9.0的范围内,诸如低于约8.9,诸如低于约8.7,诸如低于约8.6。
在一些实施方案中,通过省略缓冲剂诸如通过从本公开的口腔护理组合物中省略硅酸钠来获得本公开的口腔护理组合物的pH。因此,在一些实施方案中,本公开的口腔护理组合物由于不含硅酸钠而具有比本领域已知的基于钙的洁齿剂更低的pH。例如,在一些实施方案中,在不存在缓冲剂(诸如硅酸钠)的情况下,含有基于钙的磨料的本公开的口腔护理组合物的pH低于9.0,诸如低于8.9,诸如低于8.7,诸如低于8.6。
任选的另外的活性成分
防龋剂
在一些实施方案中,本公开的口腔护理组合物除基于钙的磨料和抗菌剂之外还包含活性成分。例如,在一些实施方案中,本公开的口腔护理组合物包含充当防龋剂的口腔可接受的氟离子源。可以存在一种或多种此类源。合适的氟离子源包括氟化物、单氟磷酸盐和氟硅酸盐以及胺氟化物,包括奥拉氟(N′-十八烷基三乙烯二胺-N,N,N′-三(2-乙醇)-二氢氟化物)。
作为防龋剂,一种或多种释放氟化物的盐任选地按提供总共为约100至约20,000ppm、约200至约5,000ppm、或约500至约2,500ppm的氟离子的量存在。在氟化钠或单氟磷酸钠是存在的唯一释放氟化物的盐的情况下,按重量计,组合物中氟化钠的存在示例量可以为约0.01重量%至约5重量%、约0.05重量%至约1重量%或约0.1重量%至约0.8重量%。通常,本公开的口腔护理组合物中所用的单氟磷酸钠的量在约0.7至约0.8重量%的范围内,诸如约0.76重量%。
碳酸氢盐
在一些实施方案中,组合物包含至少一种碳酸氢盐,其可由于泡腾和释放二氧化碳而用于向牙齿和牙龈赋予“清洁感”。可使用任何口腔可接受的碳酸氢盐,包括但不限于碱金属碳酸氢盐,诸如碳酸氢钠和碳酸氢钾、碳酸氢铵等。一种或多种碳酸氢盐任选地以组合物的约0.1重量%至约50重量%,例如约0.5重量%至约5重量%的总量存在。
抗牙结石剂
在一些实施方案中,本公开的基于钙的口腔护理组合物中包含另外的活性成分。例如,可以包含抗牙结石剂。合适的抗牙结石剂包括但不限于磷酸盐和聚磷酸盐(例如焦磷酸盐)、聚氨基丙磺酸(AMPS)、六偏磷酸盐、柠檬酸锌三水合物、多肽、聚烯烃磺酸盐、聚烯烃磷酸盐、二磷酸盐。在一些实施方案中,抗牙结石剂的存在量为约0.1重量%至约30重量%。在一些实施方案中,本公开的口腔护理组合物包含抗牙结石剂的混合物。在一些实施方案中,使用焦磷酸四钠(TSPP)和三聚磷酸钠(STPP)作为抗牙结石剂。在一些实施方案中,抗牙结石剂包含约1重量%至约2重量%的TSPP和约0重量%至约7重量%的STPP。
本公开的口腔护理组合物的另一个任选的组分为合成的阴离子聚合聚羧酸盐(SAPP),其充当聚磷酸盐抗牙垢剂的稳定剂并且可帮助阻挡令人不快的或引起疼痛的材料诸如糖进入牙神经。
增白剂
在一些实施方案中,本公开的口腔护理组合物包含增白剂。可在本公开的口腔护理组合物中使用任何已知或已开发的增白剂,包括过氧化物、金属亚氯酸盐和过硫酸盐。示例性过氧化物包括氢过氧化物、过氧化氢、碱金属和碱土金属的过氧化物、有机过氧化合物、过氧酸、它们的药学上可接受的盐以及它们的混合物。
合适的碱金属和碱土金属的过氧化物包括过氧化锂、过氧化钾、过氧化钠、过氧化镁、过氧化钙、过氧化钡以及它们的混合物。有机过氧化合物包括过氧化脲、甘油基过氧化氢、烷基过氧化氢、二烷基过氧化物、烷基过氧酸、过氧酯、二酰基过氧化物、过氧化苯甲酰和单过氧邻苯二甲酸盐以及它们的混合物。过氧酸及其盐包括有机过氧酸诸如烷基过氧酸和单过氧邻苯二甲酸盐以及它们的混合物,以及无机过氧酸盐诸如碱金属和碱土金属(诸如锂、钾、钠、镁、钙和钡)的过硼酸盐,以及它们的混合物。通常,增白剂包含过氧化氢。
在一些实施方案中,增白剂占本公开的口腔护理组合物的约0.1重量%至约50重量%,诸如约0.1重量%至约40%重量%,更通常约0.1重量%至约30重量%。在其他实施方案中,氧化剂基本上由过氧化氢组成。在一些实施方案中,过氧化氢占本公开的口腔护理组合物的约0.1重量%至约3重量%,诸如约0.1重量%至约2重量%,更通常约1重量%至约4重量%。
在一些实施方案中,固体增白剂诸如过硼酸钠、过氧化脲、亚氯酸钙、亚氯酸钡、亚氯酸镁、亚氯酸锂、亚氯酸钠、亚氯酸钾或它们的混合物用于本文所述的口腔护理组合物中。
在一些实施方案中,增白剂与例如聚合物诸如PVP(聚(N-乙烯基吡咯烷酮))结合。合适的PVP络合物在例如美国专利No.5,122,370中进行了公开,该专利的内容以引用方式整体并入本文。
亚锡离子
本公开的口腔护理组合物还可包含亚锡离子或亚锡离子源以减轻钙损失。合适的亚锡离子源包括但不限于氟化亚锡、其他卤化亚锡诸如二水氯化亚锡、焦磷酸亚锡、有机亚锡羧酸盐诸如甲酸亚锡、乙酸亚锡、葡萄糖酸亚锡、乳酸亚锡、酒石酸亚锡、草酸亚锡、丙二酸亚锡和柠檬酸亚锡、乙醛酸亚锡(stannous ethylene glyoxide)等等。一种或多种亚锡离子源任选地和示例性地以约0.01重量%至约10重量%,例如约0.1重量%至约7重量%或约1重量%至约5重量%的总量存在。
口腔可接受的媒介物
如本文所述的基于钙的磨料、聚乙二醇、抗菌剂(诸如三氯生)以及任何另外的所需活性剂或其他口腔护理成分可与口腔可接受的媒介物组合或结合。如本文所用,“口腔可接受的媒介物”是指安全用于本公开的组合物中的材料或材料组合,与合理的效/险比相称,任何所需的活性成分可在保留显著功效的同时与所述效/风险比相关联。
在一些实施方案中,口腔可接受的媒介物是低水含量的口腔可接受的媒介物并且可包含任何已知的成分或添加剂。例如,口腔可接受的媒介物可包括水和湿润剂(诸如甘油和/或山梨糖醇)的液体混合物。在其他实施方案中,口腔可接受的媒介物可包括水和山梨糖醇的液体混合物。
在制备本公开的合适的口腔护理组合物时采用的水应优选为去离子的并且不含有机杂质。在一些实施方案中,本公开的口腔组合物的水含量低于约20重量%、低于约10重量%、低于约8重量%、低于约4重量%、低于约2重量%、低于约1重量%或低于约0.1重量%。通常,本公开的口腔护理组合物的水含量低于约17%,诸如低于约16重量%。
湿润剂(诸如甘油和/或山梨糖醇)的存在总量可在约20重量%至约50重量%的范围内,诸如约25重量%至约45重量%,诸如约30重量%至约35重量%。在一个实施方案中,本公开的口腔护理组合物的湿润剂是存在量为约35重量%的山梨糖醇。
在一些实施方案中,口腔可接受的媒介物是基本上无水的。适用于此类实施方案的口腔可接受的湿润剂包括但不限于聚乙二醇,诸如PEG400、PEG600;PEG/PPG共聚物,诸如PEG/PPG 38/8共聚物、PEG/PPG-1 16/66共聚物(分别由BASF公司(Wyandotte,Michigan)以L4370知L1220销售)。在一些实施方案中,湿润剂的存在总量为约2重量%至约55重量%,并且通常为约2重量%至约35重量%。丙二醇或甘油的存在量还可以为约0重量%至约15重量%。
在一些实施方案中,口腔可接受的媒介物是高水含量的口腔可接受的媒介物并且可包含任何已知的成分或添加剂。例如,口腔可接受的媒介物可包括水、湿润剂(诸如甘油和/或山梨糖醇)和氯化钠的液体混合物。在此类实施方案中,水的存在量通常为本公开的口腔护理组合物的至少约21重量%,诸如约25重量%至约50重量%,诸如约40重量%至约75重量%,诸如在约77重量%至约88重量%的范围内。
其他口腔护理成分
增稠剂
在一些实施方案中,本公开的口腔护理组合物包含增稠剂。可以使用任何口腔可接受的增稠剂,其包括但不限于也称为羧基乙烯基聚合物的卡波姆、也称为爱尔兰藓并且更具体地为-角叉菜胶(ι-卡拉胶)的角叉菜胶、纤维素聚合物(诸如羟乙基纤维素、羧甲基纤维素(CMC)及其盐,例如CMC钠)、天然树胶(诸如低酰基结冷胶、黄原胶、刺梧桐胶、阿拉伯树胶和黄蓍胶)、胶体硅酸镁铝、胶体和/或增稠剂二氧化硅以及它们的混合物。

在一些实施方案中,特别是当口腔护理组合物为洁齿剂时,增稠剂包括角叉菜胶和增稠剂二氧化硅。角叉菜胶的存在量可在约0.05重量%至2.5重量%的范围内,诸如约1.0重量%至约2重量%,诸如约0.85重量%。增稠剂二氧化硅的存在量通常在约0.05重量%至约2.5重量%的范围内,诸如约1.0重量%-2重量%,诸如约1.75重量%。
结构剂
在一些实施方案中,本公开的口腔护理组合物包含结构剂,所述结构剂诸如在结构化漱口液(即包含提供流变学和结构化有益效果的材料的漱口液)中使用。此类结构剂包括但不限于可在水合时溶胀或膨胀形成独立的微凝胶粒子的随机分散体的聚合物或树胶结构剂。聚合物和树胶结构剂的实例包括:结冷胶、果胶、藻酸盐、阿拉伯半乳聚糖、角叉菜胶、黄多醣胶、瓜耳胶、鼠李胶、红藻胶、羧甲基纤维素和纤维素。另见PCT公开No.WO2014088534,其以引用方式整体并入本文。在一些实施方案中,可以使用硅酸钠作为结构剂。
在一些实施方案中,口腔护理组合物的粘度在200cps至1,000,000cps的范围内。例如,洁齿剂的粘度为60,000cps至1,000,000cps。
盐
在某些实施方案,特别是漱口液实施方案中,口腔可接受的媒介物包含0.10重量%至1.0重量%的单价或二价氯化物盐,诸如CaCl2、NaCl或它们的混合物,以达到例如在200至1000cps的范围内的粘度。
表面活性剂
在一些实施方案中,本公开的口腔护理组合物包含表面活性剂,可以添加该表面活性剂来增强制剂的稳定性,通过去垢作用帮助清洁口腔表面,并且在搅拌时例如在用本公开的口腔护理组合物刷洗或淋洗过程中提供泡沫。表面活性剂可通过将抗菌剂完全分散在例如整个口腔中来实现增加的预防作用。在各种实施方案中,合适的表面活性剂可用作表面活性剂、乳化剂和/或泡沫调节剂。
可使用任何口腔可接受的表面活性剂,它们大部分是阴离子型、非离子型或两性离子型的。合适的阴离子型表面活性剂包括但不限于C8-20烷基硫酸酯、磺化的C8-20脂肪酸甘油单酯的水溶性盐、肌氨酸盐、牛磺酸盐等。这些和其他种类的示例性实例包括月桂基硫酸钠、椰油基甘油单酯磺酸钠、月桂基肌氨酸钠、月桂基羟乙基磺酸钠(sodium laurylisoethionate)、月桂醇聚醚羧酸钠(sodium laureth carboxylate)和十二烷基苯磺酸钠。合适的非离子型表面活性剂包括但不限于泊洛沙姆(诸如泊洛沙姆407)、聚氧乙烯脱水山梨糖醇酯(诸如聚山梨醇酯20)、脂肪醇聚氧乙烯醚、烷基酚聚氧乙烯醚、叔胺氧化物、叔膦氧化物、二烷基亚砜等。合适的两性型表面活性剂包括但不限于具有阴离子基团(诸如羰酸根、硫酸根、磺酸根、磷酸根或膦酸根)的C8-20脂肪族仲胺和叔胺的衍生物。合适的实例为椰油酰胺基丙基甜菜碱。通常,在本公开的口腔护理组合物中使用月桂基硫酸钠作为表面活性剂。
在一些实施方案中,一种或多种表面活性剂的存在总量可以为约1.8重量%至约4重量%。在一些实施方案中,一种或多种表面活性剂的存在总量可以为约1.9重量%至约3重量%。在一些实施方案中,一种或多种表面活性剂的存在总量可以为约2.5重量%。
调味剂
可与本公开的口腔组合物一起使用的可用调味剂包括可操作用于增强组合物的味道的任何材料或所述材料的混合物。可使用任何口腔可接受的天然或合成的调味剂,诸如调味油、调味醛、酯、醇、类似材料及其组合。调味剂包括香草醛、鼠尾草、马郁兰、欧芹油、留兰香油、肉桂油、冬青油(水杨酸甲酯)、薄荷油、丁香油、月桂油、茴香油、桉树油、柑橘油、水果油和水果香精(包括源自柠檬、桔子、酸橙、葡萄柚、杏子、香蕉、葡萄、苹果、草莓、樱桃、菠萝等的那些)、源自豆类和坚果的调味剂(诸如咖啡、可可、可乐果、花生、杏仁)等,吸附的和包封的调味剂以及它们的混合物。可使用食品级磷酸来酸化本公开的口腔护理组合物并且提供浓烈且酸性的味道。还涵盖在本文的调味剂之内的是在口腔中提供芳香和/或其他感官效果(包括凉爽或温热效果)的成分。此类成分包括薄荷醇、乙酸薄荷酯、乳酸薄荷酯、樟脑、桉树油、桉油醇、茴香脑、丁香酚、肉桂、覆盆子酮(oxanone)、x-紫罗兰酮、丙烯基乙基愈创木酚(propenyl guaiethol)、麝香草酚、里哪醇、苯甲醛、肉桂醛、N-乙基-对-薄荷烷-3-甲酰胺、N,2,3-三甲基-2-异丙基丁酰胺、3-1-薄荷氧基丙烷-1,2-二醇、肉桂醛甘油缩醛(CGA)、薄荷酮甘油缩醛(MGA)以及它们的混合物。
在一些实施方案中,一种或多种调味剂任选的存在总量为约0.01重量%至约5重量%。在一些实施方案中,一种或多种调味剂任选的存在总量为约0.05重量%至约3重量%.在一些实施方案中,一种或多种调味剂任选的存在总量为约0.1重量%至约2.5重量%。在一些实施方案中,一种或多种调味剂任选的存在总量为约0.1重量%至约1.5重量%,诸如约0.95重量%。
甜味剂
可用于本文中的甜味剂包括口腔可接受的天然或人造、营养性或非营养性甜味剂。此类甜味剂包括右旋糖、聚右旋糖、蔗糖、麦芽糖、糊精、干的转化糖、甘露糖、木糖、核糖、果糖、左旋糖、半乳糖、玉米糖浆(包括高果糖玉米糖浆和玉米糖浆固体)、部分水解的淀粉、氢化淀粉水解产物、山梨糖醇、甘露醇、木糖醇、麦芽糖醇、异麦芽酮糖醇、阿斯巴甜、纽甜、糖精及其盐、三氯蔗糖、基于二肽的强增甜剂、环己氨基磺酸盐(cyclamate)、二氢查耳酮以及它们的混合物。一些实施方案中任选地包含一种或多种甜味剂。在一些实施方案中,一种或多种任选的甜味剂的存在总量为约0.005重量%至约5重量%。在一些实施方案中,一种或多种任选的甜味剂的存在总量为约0.01重量%至约1重量%。在一些实施方案中,使用约0.3%的糖精钠作为甜味剂。
着色剂
在一些实施方案中,本公开的口腔护理组合物中可以包含着色剂。如本文所用,术语“着色剂”是指向另一种物质赋予颜色的呈干粉或液体形式的物质。一般来讲,着色剂包括染料、色淀、颜料或它们的组合。在一些实施方案中,也可将云母、云母基珠光颜料、滑石、碳酸钙和二氧化硅用作着色剂。更通常地,着色剂为染料、色淀或一种或多种染料和/或一种或多种色淀的组合。甚至更通常地,将颜料掺入本公开的口腔护理组合物中。此类颜料包括无毒、水不溶性无机颜料,诸如二氧化钛和氧化铬绿、群青蓝和粉红以及氧化铁,例如胭脂树提取物、叶绿酸铜络合物、角黄素、β-胡萝卜素、合成的铁氧化物、TiO2以及它们的混合物。
在一些实施方案中,着色剂诸如颜料的所需量为使得本公开的口腔护理组合物中着色剂的最终浓度高达12%的着色剂量,例如约0.1重量%至约12重量%、约0.1重量%至约10重量%、约0.1重量%至约2重量%、或约0.1重量%至约1重量%。通常,本公开的口腔护理组合物中使用约1重量%的二氧化钛。
抗氧化剂
在一些实施方案中,本公开的口腔护理组合物中包含抗氧化剂。合适的抗氧化剂包括抗坏血酸、抗坏血酸棕榈酸酯、硫代二丙酸、抗坏血酸钙、二月桂基二硫代丙酸酯、愈创木胶、抗坏血酸钠、丁基化羟基甲苯、丁基化羟基苯甲醚和生育酚。也可使用抗氧化剂的混合物。
当存在时,抗氧化剂的添加水平有效减少或减轻原本可能因牙膏组分的氧化而造成的变色。当抗氧化剂存在时,典型的水平在约0.01重量%至约1重量%的范围内。
防腐剂
合适的防腐剂诸如苄醇也可包含在本公开的口腔护理组合物中。其他可用的防腐剂包括对羟基苯甲酸的甲酯、乙酯、丁酯、丙酯和异丙酯。其他合适的防腐剂包括甲醛、二甲基乙内酰脲和硅酸钠。在一些实施方案中,本公开的口腔护理组合物包含约0重量%至约0.8重量%、通常约0.05重量%至约0.5重量%、更通常约0.1重量%至0.3重量%的防腐剂。通常,本公开的口腔护理组合物中使用苄醇。
方法
本公开还提供了制备本公开的口腔护理组合物的方法。该方法包括将包括基于钙的磨料(诸如沉淀碳酸钙)、聚乙二醇(诸如PEG 6000)和抗菌剂(诸如三氯生)的各种成分与口腔可接受的媒介物组合。在一些实施方案中,包含缓冲剂和/或pH调节剂以获得如本文所公开的本公开的口腔护理组合物的pH,例如低于9.0,诸如8.68的pH。在其他实施方案中,省略缓冲剂(诸如硅酸钠)以获得所需的pH,例如低于9.0,诸如8.68。在各种实施方案中,本公开的口腔护理组合物还可以包含另外的活性成分,包括如本文所公开的氟化物、碳酸氢盐、抗牙结石剂、增白剂和亚锡离子。还可以包含另外的口腔护理成分,诸如增白剂、表面活性剂、甜味剂、风味剂、防腐剂和着色剂。
在一些实施方案中,用于制备口腔护理组合物的时间在1小时至4小时范围内,诸如约1小时至约3小时,诸如约1小时至约2小时。通常,用于制备本公开的口腔护理组合物的时间为约1.5小时或更短。
在一些实施方案中,通过在60℃至62℃范围内的温度下将湿润剂(诸如山梨糖醇)与水混合形成口腔可接受的媒介物来制备本公开的口腔护理组合物。通常,水是加热至85℃的经巴氏灭菌、过滤和纯化的水。
通常,将一种或多种增稠剂(诸如角叉菜胶和/或增稠二氧化硅)加入口腔可接受的媒介物。在一些实施方案中,在将增稠剂与口腔可接受的媒介物组合之前,将活性成分(诸如单氟磷酸钠和碳酸氢钠)加入增稠剂(诸如角叉菜胶)。还可以将甜味剂加入增稠剂(诸如角叉菜胶)和任选的活性成分。在与口腔可接受的媒介物组合之前,使增稠剂和任选的另外的成分在55℃-65℃范围内的温度(诸如55℃至60℃或诸如64℃至65℃)下受热混合。
在一些实施方案中,然后可以将缓冲剂(诸如硅酸钠)加入上述混合物。
在一些实施方案中,然后将基于钙的磨料(诸如沉淀碳酸钙)加入口腔可接受的媒介物和另外的成分,例如增稠剂、单氟磷酸钠、碳酸氢钠和甜味剂。可以在封闭真空中添加沉淀碳酸钙并且同时除气以制备顺滑的不含空气的糊剂。在各种实施方案中,着色剂(诸如二氧化钛)与基于钙的磨料同时添加。
在一些实施方案中,可以将表面活性剂(诸如月桂基硫酸钠)加入含有基于钙的磨料的上述混合物。通常,表面活性剂呈颗粒形式,并且在52℃至46℃范围内的温度下在开放式真空下缓慢添加同时搅拌。
在一些实施方案中,通过将PEG-6000与抗菌剂(例如三氯生)组合来制备PEG-6000预混物。在其他实施方案中,将防腐剂(诸如苄醇)和调味剂首先与PEG-6000混合直至PEG-6000溶解,然后添加抗菌剂。然后将PEG-6000预混物加入含有基于钙的磨料和任选的表面活性剂的上述混合物以形成本公开的口腔护理组合物。
本文还提供了用于促进人或动物受试者的口腔健康的方法,包括使口腔表面与本公开的口腔护理组合物接触,该口腔护理组合物包含基于钙的磨料、如上所述的聚乙二醇、抗菌剂、口腔可接受的媒介物和如本文所公开的合适的pH下的其他任选成分。如本文所用,短语“促进口腔健康”涵盖预防、养护、逆转、减弱、减轻、改善、最小化、抑制或停止人或动物的一种或多种口腔病症的有害影响。短语“口腔病症”包括但不限于牙斑、牙垢、色斑、口臭、齿龈炎、牙周炎以及它们的组合。
如本文所用的“动物受试者”包括非人哺乳动物,诸如犬科、猫科和马科动物。本公开的口腔护理组合物与哺乳动物受试者的口腔表面接触,从而以高度有效的方式促进口腔健康。
在各种实施方案中,根据本公开制备的本公开的口腔护理组合物可定期施用于口腔表面,例如在每天的基础上,每天至少一次持续多天,或者每隔一天或每隔两天。在一些实施方案中,每天向口腔表面施用本公开的口腔护理组合物1至3次,持续至少2周至8周、从4个月至3年、或更长时间直至一生。
本文所述的实例和其他实施方案是示例性的并且不旨在限制描述本公开的组合物和方法的完全范围。可在本公开的范围内进行具体实施方案、材料、组合物和方法的等同改变、修改和变型,具有基本上类似的结果。
实施例
实施例1-洁齿剂制剂
如下文所述通过混合表1中指定量的成分制备七种洁齿剂制剂A、C、D、E、F、G和H。包含的竞品洁齿剂制剂称为配方B(表2)。
将配备有搅拌器的不锈钢容器(SS容器)装上70%山梨糖醇并加热至60℃,同时搅拌。将经巴氏灭菌、过滤、纯化的水加入山梨糖醇并加热至85℃。
将含有角叉菜胶、单氟磷酸钠、糖精钠和碳酸氢钠的预混物缓慢加入单独的容器(SS容器)以形成凝胶物质。在64℃至65℃范围内的温度下混合凝胶物质直至观察不到任何团块(约20分钟)。将0.5重量%的巴氏灭菌水加入SS容器以补偿水损耗。然后将凝胶物质转移至不锈钢混合容器(Alpro搅拌器)。
然后将液体硅酸钠(配方A、C、E、F和H)加入不锈钢混合容器(Alpro搅拌器)。将混合物搅拌2分钟。
然后将增稠二氧化硅(-P)装入不锈钢混合容器并混合直至完全分散于上述山梨糖醇、水、凝胶物质和液体硅酸钠混合物中(约7分钟)。
然后将两个单独批次的沉淀碳酸钙和二氧化钛加入上述混合物。在封闭式真空(650mm Hg)下搅拌混合物同时除气直至温度达到52℃。然后施加全真空(720mm Hg)并对混合物除气。该过程在约15分钟内完成。
在52℃下将月桂基硫酸钠颗粒缓慢加入上述混合物,同时在开放式真空(650mmHg)下搅拌直至温度下降至46℃(约10分钟)。将混合物搅拌15分钟。监测真空以确保压力不会下降至低于600mm Hg。当压力下降至低于600mm Hg(这时施加全真空(720mm Hg))时,中止添加月桂基硫酸钠颗粒,然后在开放式真空下继续添加月桂基硫酸钠。
将PEG-6000(细粉形式)与调味剂和苄醇组合并且搅拌直至PEG-6000溶解。然后添加三氯生并溶解形成PEG-6000预混物。PEG-6000预混物溶液略微混浊。不受理论的约束,据信混浊可能是由于PEG-6000和苄醇之间的乳化作用导致的固体完全分散。将上述PEG-6000预混物加入不锈钢混合容器并在封闭式真空(650mm Hg)下与此前添加的成分组合直至预混物完全分散。然后使混合物经受全真空(720mm Hg)、除气并且搅拌五分钟。
在完成上述过程后,停止搅拌器并释放真空。检查该批次的颗粒性。使发现为粒状的任何批次经受全真空(720mm Hg)并混合五分钟。抽取批次的样品并检查其味道、颜色、气味、质地、pH、比重和月桂基硫酸钠颗粒的溶解。
如上所述制备的所有配方A、C、D、E、F、G和H含有如表1中指定的39%碳酸钙和0.3%三氯生;然而,仅配方E、F和G含有PEG 6000。配方A(对照)、E、F和H各自含有1%硅酸钠。配方C含有3%硅酸钠。配方D和G不含硅酸钠。由于不含硅酸钠,配方D和G表现出比配方A、C、E、F和H更低的pH值,即分别为8.9和8.68。参见表1。

实施例2.吸收率和保持率
使用涂有唾液的羟基磷灰石(SCHAP)盘(即牙釉质的矿物相)作为人类牙齿的体外实验模型,评估本公开的口腔护理组合物中的聚乙二醇(诸如PEG 6000)对牙齿表面的三氯生吸收率和保持率的影响。据发现体外评估与抗菌剂的体内递送和在口腔表面上的保持有关。
为了测试SCHAP盘的三氯生吸收率,将石蜡膜刺激的全唾液收集到烧杯中并通过以10,000rpm离心10分钟来澄清。将每个羟基磷灰石盘在试管中用无菌水水合。然后去除水并替换为2毫升澄清唾液。通过将盘在37℃下温育过夜同时在水浴中连续振摇来形成唾液膜。在该处理后,通过抽吸移除过量的唾液,将盘转移到新的管中并用洁齿剂浆液处理。通过用10克水稀释5克如本文所述的洁齿剂配方A-G中的每一个然后在烧杯中用搅拌棒搅拌来制备洁齿剂浆液。水还被用作对照洁齿剂制剂。
然后在37℃下用1毫升洁齿剂浆液温育这些盘。在30分钟后,从每个盘抽取上清液,然后将这些盘各自转移到新的管中并向每根新的管加入5毫升水,然后将盘涡旋振摇五秒。然后将这些盘各自转移到新的管中,并且重复洗涤程序总共三次。为了评估三氯生吸收率,向每个盘加入1.0ml乙醇并在室温下放置1小时,以将被吸收的三氯生提取到乙醇中,并且涡旋振摇十秒。抽取乙醇并转移到HPLC(高效液相色谱)小瓶中,以测定三氯生的存在浓度。
为了评估三氯生保持率,程序与如上针对三氯生吸收率所述的相同,不同的是在将盘洗涤三次后,将每个盘置于1毫升唾液中并在37℃下温育四小时。如上针对三氯生吸收率所述,然后向每个盘加入1.0ml乙醇并在室温下放置1小时,以将被吸收的三氯生提取到乙醇中,并且涡旋振摇十秒。然后抽取乙醇并转移到HPLC(高效液相色谱)小瓶中,以测定保持在盘上的三氯生的浓度。
实施例3.结果
如表2中证明,高分子量聚乙二醇(诸如PEG 6000)意外地提高了抗菌剂(诸如三氯生)的吸收率(0小时)和保持率(4小时)。如表2中所示,在羟基磷灰石(HAP)盘上,含有PEG6000的制剂在0小时具有比对照配方A更好的吸收率。例如,如表2中所见,相比于对照配方A(即在0小时18μg三氯生/HAP),含有PEG-6000的制剂在0小时导致羟基磷灰石盘上存在24μg(配方E)、20μg(配方F)和38μg(配方G)三氯生。另见图1。
此外,表2还显示,在羟基磷灰石盘上,含有PEG 6000的制剂在4小时具有比对照配方A更好的保持率。例如,如表2中所见,相比于对照配方A(即在4小时7μg三氯生/HAP),含有PEG-6000的制剂在4小时导致羟基磷灰石盘上存在11μg(配方E)、10μg(配方F)和23μg(配方G)三氯生。另见图1。
令人意外的是,省略硅酸钠(其导致略微降低的pH)也出人意料地提高了三氯生的吸收率。例如,如表2中所见,相比于对照配方A,pH 9.37(即,在0小时18μg三氯生/HAP),不含硅酸钠的制剂在0小时导致羟基磷灰石盘上存在30μg(配方D,pH 8.9)和38μg(配方G,pH8.68)三氯生。另见图1和图2。
此外,省略硅酸钠并因而得到较低的pH也出人意料地提高了三氯生的保持率。例如,如表2中所见,相比于对照配方A(即,在4小时仅7μg三氯生/HAP),不含硅酸钠的制剂在4小时导致羟基磷灰石盘上存在19μg(配方D)和23μg(配方G)三氯生。另见图1和图2。
此外,表2显示,将PEG 6000与较低pH(无硅酸钠)相结合以协同方式(大于相加作用)增大了三氯生的吸收率和保持率。参见例如,相比于对照配方A(即,在0小时18μg三氯生/HAP并且在4小时7μg三氯生/HAP),配方G(其含有0.3%PEG 6000并且pH为8.68(无硅酸钠))在0小时保持了38μg三氯生/HAP并且在4小时候保持了23μg三氯生/HAP。另见图1和图2。
根据上文,本公开的实施例证明,使用本公开的口腔护理组合物得到了优异的抗菌剂(诸如三氯生)吸收率和保持率。

Claims (24)
1.一种口腔护理组合物,其包含:
(i)基于钙的磨料;
(ii)聚乙二醇,所述聚乙二醇的平均分子量在约1500至约6000道尔顿的范围内;
(iii)抗菌剂;以及
(iv)口腔可接受的媒介物。
2.根据权利要求1所述的口腔护理组合物,其中所述抗菌剂选自卤代二苯醚、酚类化合物、倍半萜烯醇和卤代二苯脲以及它们的组合。

a)将湿润剂和水组合以形成口腔可接受的媒介物;
b)将基于钙的磨料加入所述口腔可接受的媒介物;
c)将平均分子量在约1500至约6000道尔顿范围内的聚乙二醇、抗菌剂以及任选地选自调味剂和防腐剂的成分组合以形成预混物;
d)将步骤c)的所述预混物加入步骤b)的含有所述基于钙的磨料的所述口腔可接受的媒介物以形成所述口腔护理组合物。
17.根据权利要求16所述的方法,其中所述方法还包括在将所述基于钙的磨料加入步骤b)的所述口腔可接受的媒介物之前,将至少一种增稠剂加入步骤a)的所述口腔可接受的媒介物。
18.根据权利要求17所述的方法,其中所述至少一种增稠剂包括选自角叉菜胶、增稠二氧化硅以及它们的组合的增稠剂。
19.根据权利要求16-18中任一项所述的方法,其中所述聚乙二醇的平均分子量为6000道尔顿。
20.根据权利要求16-19中任一项所述的方法,其中所述抗菌剂为5-氯-2-(2,4-二氯苯氧基)苯酚(三氯生)。
21.根据权利要求16-20中任一项所述的方法,其中所述基于钙的磨料为沉淀碳酸钙。
22.根据权利要求16-21中任一项所述的方法,其中所述口腔护理组合物的pH在约8.6至约8.9的范围内。
23.根据前述权利要求中任一项所述的方法,其中所述口腔护理组合物不含聚(甲基乙烯基醚/马来酸)的单烷基酯的共聚物。
24.一种促进口腔健康的方法,所述方法包括:
使口腔表面与口腔护理组合物接触,所述口腔护理组合物包含:
基于钙的磨料,
聚乙二醇,所述聚乙二醇的平均分子量在约1500至约6000道尔顿的范围内,
抗菌剂,以及
口腔可接受的媒介物。
