随着科学技术的不断发展,人们生活水平的不断提高,对珍珠粉的利用开发也有新的设想,国内报导的珍珠水解液、高效珍珠粉等,仅仅是将难溶的碳酸钙制成可溶的有机钙,实质上它是碳酸钙及部分微量元素,而壳角蛋白质还未利用。而发明名称为“水溶性全成分珍珠粉系列产品新制备方法”CN1051394A的专利申请用可食性羧酸或氨基酸置换珍珠中矿物质的碳酸根,使珍珠硬蛋白分离出来,然后用酸、酶二重水解法制得几种水溶性全成分珍珠产品,但均存在有一定的不足之处。
由于珍珠粉未能被完全水解成人体能容易吸收的成分,因此目前还存在一定的缺点,特别它的主要成分之一的壳角蛋白质还未被人体完全吸收和充分利用。本发明目的在于发现一种天然活性物质能使珍珠粉酶解成人体极易吸收的氨基酸及人体有益的小肽分子,使珍珠粉完全被利用,充分发挥它的特殊功能。
本发明先将珍珠粉用乳酸处理,使其成为可溶性的乳酸钙液,它的主要成分为珍珠活性钙和微量元素,而不溶解部分为珍珠壳角蛋白质,将珍珠壳角蛋白质用天然植物活性物质木瓜蛋白酶进行水解,使其获得二十种的氨基酸,其中有八种人体必需的氨基酸,还有多肽物质,大大提高了珍珠粉的功能。
本发明通过以下步骤实施:
将乳酸加蒸馏水稀释充分混匀后,成为乳酸溶液,将珍珠粉分次加入到乳酸溶液中,珍珠粉边加入,边搅拌,待反应完全后,离心,所得上清液即为珍珠粉活性钙和微量元素,沉淀部分为珍珠壳角蛋白质。
将蒸馏水加入珍珠壳蛋白质中,微微加温至35-39℃,然而加入木瓜酶、半胱氨酸、乙二胺四乙酸二钠盐,进行酶解。酶解后,离心,得酶解珍珠液,测得其总含氮量及氨基酸成分后,分装在容器中高压消毒后,即得产品酶解珍珠液。
取酶解珍珠液0.2ml加入0.2ml 6%磺基水杨酸,清液稀释10倍,取0.05ml测定它的氨基酸及多肽含量,氨基酸总含量为17.58mg/ml。表1为酶解珍珠液氨基酸分析结果。

酶解珍珠液中含有12-13肽物质,它的分子量为1500左右,还含有一个次要成分,它的分子量≥5000。小肽与氨基酸的混合物在机体内的吸收率和速度都比单纯氨基酸佳,而它们的生物效价高于纯的氨基酸。
酶解珍珠液按上海药品检验所对药品毒性试验法进行小白鼠灌胃急性毒性LD50试验,结果如下:
采用昆明杂种小白鼠,体重为20克,分成雌、雄二组,每组10只,同剂量给药,实验观察七天。酶解珍珠液以最大耐受量灌胃给药每只1ml,每日灌胃二次,连续三天灌后观察小白鼠的毛色光滑,活动状态良好,其它均属正常,得出急性毒性小白鼠灌胃1ml/20克,小白鼠体重相当于50ml/kg液量灌入到动物胃内,均未出现死亡。本品属无毒级。
酶解珍珠液的主要原料为名贵中药珍珠粉、木瓜蛋白酶,珍珠粉是人们长期服用的,未见任何副作用发生,而木瓜蛋白酶早在1911年美国华勒斯坦公司已生产作为啤酒澄清剂,而它的ADI无需特殊规定,而反应后加热使剩余的酶失活通过离心除去。木瓜蛋白酶还作为药用,它的LD50为12.5g/kg属于无毒级品,我国国家标准作为食品添加剂。
本发明所采用的原料本身均是十分安全,无任何毒性,经酶解处理后又得到的均为对人体有益的氨基酸及小肽,例如:门冬氨酸可治疗肝病、苏氨酸有抗脂肪肝作用其铁盐可治疗贫血、组氨酸可抗溃疡和促进细胞再生作用、精氨酸用于肝昏迷的治疗和精子缺乏引起的男性不育症、胱氨酸可促进毛发生长,预防早衰和解毒作用等等。多肽同样具有一定人体的生理功能,例如:具有镇痛作用的吗啡五肽和十三肽P物质,具有睡眠促进作用的δ睡眠诱导肽,能促进子宫收缩和泌乳功能的催产素等的生理功能。
本发明采用木瓜蛋白酶对珍珠中的角壳蛋白质进行酶解,它的酶解条件极其温和,酶解后处理方法简便,酶解后所得产品组分稳定,木瓜蛋白酶价格便宜,可降低生产成本。本发明采用的方法克服了现有传统的方式服用珍珠粉,而使原来不易被吸收的壳角蛋白变成容易被人体吸收的氨基酸及多肽,大大提高了珍珠粉对人体的生理功能。
下面用实施例更详细地说明本发明,应理解为实施例在任何方面不对本发明范围限制。
实施例:
取乳酸3.2立升,加入蒸馏水42立升,充分混匀后成为乳酸溶液,将1公斤珍珠粉分次加入乳酸溶液中,边搅拌边加入,待反应完成后,离心,得上清液和不溶部分,上清液即为珍珠活性钙及微量元素部分,不溶部分即为珍珠壳角蛋白质,再取湿的壳角蛋白质1公斤,加入蒸馏水10立升在35-38℃下,加入15克木瓜蛋白酶,1克盐酸半胱氨酸,1克乙二胺四乙酸二钠盐,加毕后35-38℃下酶解3-5小时后,离心,得酶解珍珠液,测它的全氮含量后,分装到容器中,高压消毒灭菌即为产品。
取酶解珍珠液0.2ml加入6%磺基水杨酸,清液稀释10倍,进行氨基酸分析结果如下:

Claims (4)
1.一种酶解珍珠液的制备方法,由:
A珍珠粉在稀乳酸溶液中搅拌反应、离心,制成珍珠活性钙;
B离心不溶部分壳角蛋白质、蒸馏水加温35-38℃下分别加入木瓜蛋白酶、盐酸半胱氨酸、乙二胺四乙酸二纳盐搅匀、酶解;
C酶解珍珠液灭菌组成。

4.根据权利要求1所述的酶解珍珠液制备方法,其特征在于酶解温度为35-38℃。
