Abstract
本发明公开了一种含聚谷氨酸的口腔护理牙膏,包括以下重量份数的原料:γ‑聚谷氨酸0.2‑0.8份,普鲁兰多糖1‑3份,山梨糖醇10份,甘油9份,月桂醇硫酸酯钠3份,香精1份,纤维素胶1份,焦磷酸钠0.2份,糖精钠0.2份,苯甲酸钠0.02份,维生素C 0.2份,维生素E 0.5份,SOD 0.05份,CPP 0.02份。本发明使用聚谷氨酸作为功效因子,不仅营养口腔,美白牙齿,平衡菌群,预防口腔问题,同时聚谷氨酸降解后的谷氨酸是无毒的,对牙齿和牙龈有营养作用。聚谷氨酸作为生物活性物质的保护剂,可以最大程度的保护其在牙膏体中的活性,延长其作用时间,发挥其功效,对人体无任何毒副作用,更天然、更健康。

技术领域
本发明涉及日化用品领域,具体说是一种含聚谷氨酸的口腔护理牙膏及其制备方法。
背景技术
随着人类生活水平的逐渐提高,越来越多的人们对口腔的保健观念和习惯发生了较大变化。同时,现代生活的快节奏、高压力让中国人的口腔状况日渐复杂化、多样化,以含氟牙膏、中草药牙膏、脱敏牙膏为代表的传统功效型牙膏逐渐被细分。从2005年开始,在云南白药牙膏的带动下,相当数量的知名牙膏品牌陆续推出了高端功效牙膏,牙膏的功效、品质、包装得到进一步提升。目前功效型牙膏的功能主要分为两个大类:一是预防和辅助治疗某些口腔疾病,如蛀牙、牙敏感、牙龈炎、牙周炎、口腔溃疡、牙齿缺损等。二是帮助调整和改善口腔状况,如增白牙齿、去口腔异味、去牙结石、去牙渍和牙斑等。未来随着消费者对口腔问题的关注变化和医学机构对口腔状况更深度地了解,还会有一些新的功能需求。如抗口干牙膏、补牙牙膏被挖掘出来。专利CN105267234A中提出了一种缓解口干症状的组合物及其制备方法,其中提出了采用透明质酸、普鲁兰多糖、聚谷氨酸的一种或多种。可以减轻由于疾病、使用药物、放化疗及老化引起的慢性和暂时性口干症,能够润滑口腔,并且保持口腔湿润。
而聚谷氨酸,又称纳豆胶,是一种水溶性,生物降解,不含毒性,使用微生物发酵法制得的生物高分子。其保湿锁水功效是透明质酸的500倍,由于其特殊的分子结构,使其具有极强的保湿能力,能有效地增加皮肤的保湿能力,促进皮肤健康,其效果由于透明质酸以及胶原蛋白。作为增白剂具有长效持久的抗敏性能。添加了聚谷氨酸的牙膏是一种天然口腔护理产品,既可以实现营养口腔,平衡菌群,预防口腔问题的作用,又避免了化学药物的抗药性和中草药有效成分偏低的缺点。并且对人体无任何毒副作用,更天然、更健康。
发明内容
本发明的目的是提供一种营养口腔,美白牙齿,平衡菌群,预防口腔问题的口腔护理牙膏及其制备方法,该牙膏是以天然、无公害、无副作用的聚谷氨酸高分子作为有效成分加入膏体中,复配以维生素,氨基酸和溶菌酶、SOD、CPP等生物活性物质。
本发明解决上述技术问题所采用的技术方案是:一种含聚谷氨酸的口腔护理牙膏的制备方法,其特征在于,包括以下步骤:
(1)将γ-聚谷氨酸和普鲁兰多糖先溶解于蒸馏水中,并搅拌均匀,使其成为粘稠状的液体;
(2)在室温条件下,向步骤(1)中得到的粘稠状液体中加入维生素,氨基酸、SOD、CPP,混合均匀;
(3)将步骤(2)得到的液体与牙膏基质混合,送入三合一真空制膏机,高速搅拌研磨30分钟脱气;再加入香精搅拌研磨15分钟混匀,获得牙膏膏体,通过无菌灌装后包装;
所述的牙膏基质包括山梨糖醇、甘油、月桂醇硫酸酯钠、纤维素胶、焦磷酸钠、糖精钠、苯甲酸钠。
进一步,所述牙膏中物质的重量份数如下:γ-聚谷氨酸0.2-0.8份,普鲁兰多糖1-3份,山梨糖醇10份,甘油9份,月桂醇硫酸酯钠3份,香精1份,纤维素胶1份,焦磷酸钠0.2份,糖精钠0.2份,苯甲酸钠0.02份,维生素C 0.2份,维生素E 0.5份,SOD 0.05份,CPP 0.02份。
进一步,所述γ-聚谷氨酸的制备方法包括以下步骤:
(1)菌种活化:将地衣芽孢杆菌接种在斜面培养基上,37℃培养16小时,反复活化2-3次,得到成熟的斜面种子;
(2)种子液的制备:将步骤(1)中活化后的斜面种子接种于装有50ml液体种子培养基的500ml三角瓶中,37℃,220rpm培养16小时至对数生长期,得到种子液;
(3)发酵罐发酵:将步骤(2)中的种子液以培养基体积分数5%的接种量接种于发酵罐中,30L发酵罐发酵培养基装液量为15L,初始发酵温度38-40℃,罐压0.01-0.05Mpa,通风量0.5-1.0vvm,转速400-600rpm;当发酵液OD660在0.550-0.600时,并控制发酵温度30-35℃,调节转速至200-350rpm,通风量1.5-2.0vvm,控制pH在7.2-7.5,发酵2-6小时,得到含γ-聚谷氨酸的发酵液;
(4)聚谷氨酸的提取:将步骤(3)中得到的发酵液离心去菌体,将上清液加入5倍上清液体积的乙醇,沉析出聚谷氨酸,过滤后得到γ-聚谷氨酸,然后在复溶于上清液等体积的蒸馏水中,利用喷雾干燥方法制备出干粉γ-聚谷氨酸。同时将上清液用流动相稀释数倍至γ-聚谷氨酸的含量在1-3g/L,通过高效液相对其分子量进行检测,分子量在20万到200万道尔顿。
进一步,所述步骤(2)中种子培养基组成为:葡萄糖30g/L,酵母膏7g/L,胰蛋白胨10g/L,K2HPO4 0.5g/L,MgSO4·7H2O 0.5g/L,pH7.2,余量纯净水。
进一步,所述步骤(3)中发酵培养基组分为:葡萄糖80g/L,氯化钠10g/L,MgSO4·7H2O 1g/L,无水氯化钙1g/L,FeSO4·7H2O 0.02g/L,KCl 0.5g/L,精氨酸0.8g/L,组氨酸0.5g/L,苏氨酸0.5g/L,甲硫氨酸0.4g/L,谷氨酰胺0.5g/L,胆碱0.31g/L,吡哆醇0.001g/L。
一种含聚谷氨酸的口腔护理牙膏,其特征在于:由上述任意一项的制备方法得到。
本发明生产高分子量γ-聚谷氨酸的菌株具体为地衣芽孢杆菌(Bacilluslicheniformis)CGMCC NO.3336。该菌株已于2009年10月14日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏号为CGMCC NO.3336。
本发明的优点在于:本发明所使用的聚谷氨酸作为功效因子,不仅营养口腔,美白牙齿,平衡菌群,预防口腔问题,同时聚谷氨酸降解后的谷氨酸是无毒的,对牙齿和牙龈有营养作用。聚谷氨酸作为生物活性物质的保护剂,可以最大程度的保护其在牙膏体中的活性,延长其作用时间,发挥其功效。而且对人体无任何毒副作用,更天然、更健康。
其中普鲁兰多糖的作用,能够快速形成保水膜,和聚谷氨酸配合使用能更加保湿美白,增加口腔润滑感,延长功效因子作用时间。
该方法制备聚谷氨酸工艺流程简单,尤其是简洁有效的下游提取工艺,这种提取工艺能够大幅度的减少生产成本,同时也能减少聚谷氨酸的损失率,使得聚谷氨酸的最后产率提高;另外,该方法能够获得高分子量的聚谷氨酸,有利于制作滋养修复的洗发水。专利CN105267234A一种缓解口干症状的组合物及其制备方法中主要用于缓解口干症状。本发明中的聚谷氨酸除复配了普鲁兰多糖外,还添加了维生素、氨基酸、SOD、CPP等生物活性物质,更加能够满足口腔护理的多种要求。
具体实施方式
为使本发明的目的、技术方案和优点更加清楚,下面将对本发明的技术方案进行清楚、完整的描述,所描述的实施例是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1
一、制备高分子聚谷氨酸
(1)菌种活化:将地衣芽孢杆菌CGMCC NO.3336原始菌株接种在斜面培养基上,于37℃培养16小时,如此活化2-3次,制备成熟的斜面种子;
(2)种子液的制备:将一环活化后的斜面种子接种于装有50ml液体培养基的500ml三角瓶中,37℃,220rpm培养16小时至对数生长期;
所述种子培养基组成为:葡萄糖30g/L,酵母膏7g/L,胰蛋白胨10g/L,K2HPO40.5g/L,MgSO4·7H2O 0.5g/L,pH7.2,不足部分纯净水补足;
(3)发酵罐发酵:将步骤(2)中的种子液以10%的接种量接种于发酵罐,30L发酵罐发酵培养基装液量为15L,初始发酵温度38-40℃,罐压0.01-0.05Mpa通风量0.5-1.0vvm,转速400-600rpm;当发酵液OD660在0.550-0.600时,并控制发酵温度30-35℃,调节转速至200-350rpm,通风量1.5-2.0vvm,控制pH在7.2-7.5至发酵结束前2-6小时。
所述发酵培养基组分为:葡萄糖80,氯化钠10g/L,MgSO4·7H2O 1g/L,无水氯化钙1g/L,FeSO4·7H2O 0.02g/L,KCl 0.5g/L,精氨酸0.8g/L,组氨酸0.5g/L,苏氨酸0.5g/L,甲硫氨酸0.4g/L,谷氨酰胺0.5g/L,胆碱0.31g/L,吡哆醇0.001g/L,
(4)聚谷氨酸的提取:发酵结束后离心去菌体,然后用5倍上清液体积的乙醇,醇沉析出聚谷氨酸,然后在复溶于上清液体积的蒸馏水,利用喷雾干燥方法制备出干粉聚谷氨酸。同时将上清液用流动相稀释数倍至γ-聚谷氨酸的含量在1-3g/L,通过高效液相对其分子量进行检测,分子量在20万到200万道尔顿。
二、制备牙膏
本发明所述的牙膏,包括以下重量份数的组分:γ-聚谷氨酸0.2份,普鲁兰多糖1份,山梨糖醇10份,甘油9份,月桂醇硫酸酯钠3份,香精1份,纤维素胶1份,焦磷酸钠0.2份,糖精钠0.2份,苯甲酸钠0.02份,维生素C 0.2份,维生素E 0.5份,SOD 0.05份,CPP 0.02份
实施例2
高分子聚谷氨酸的制备方法和牙膏的制备方法基本同实施例1;
所述的牙膏,包括以下重量份数的组分:γ-聚谷氨酸0.5份,普鲁兰多糖3份,山梨糖醇10份,甘油9份,月桂醇硫酸酯钠3份,香精1份,纤维素胶1份,焦磷酸钠0.2份,糖精钠0.2份,苯甲酸钠0.02份,维生素C 0.2份,维生素E 0.5份,SOD 0.05份,CPP 0.02份
实施例3
高分子聚谷氨酸的制备方法和牙膏的制备方法基本同实施例1;
所述的牙膏,包括以下重量份数的组分:γ-聚谷氨酸0.8份,普鲁兰多糖2份,山梨糖醇10份,甘油9份,月桂醇硫酸酯钠3份,香精1份,纤维素胶1份,焦磷酸钠0.2份,糖精钠0.2份,苯甲酸钠0.02份,维生素C 0.2份,维生素E 0.5份,SOD 0.05份,CPP 0.02份。

Claims (6)
1.一种含聚谷氨酸的口腔护理牙膏的制备方法,其特征在于,包括以下步骤:
(1)将γ-聚谷氨酸和普鲁兰多糖先溶解于蒸馏水中,并搅拌均匀,使其成为粘稠状的液体;
(2)在室温条件下,向步骤(1)中得到的粘稠状液体中加入维生素,氨基酸、SOD、CPP,混合均匀;
(3)将步骤(2)得到的液体与牙膏基质混合,送入三合一真空制膏机,高速搅拌研磨30分钟脱气;再加入香精搅拌研磨15分钟混匀,获得牙膏膏体,通过无菌灌装后包装;
所述的牙膏基质包括山梨糖醇、甘油、月桂醇硫酸酯钠、纤维素胶、焦磷酸钠、糖精钠、苯甲酸钠。
2.根据权利要求1所述的一种含聚谷氨酸的口腔护理牙膏的制备方法,其特征在于,所述牙膏中各物质的重量份数如下:γ-聚谷氨酸0.2-0.8份,普鲁兰多糖1-3份,山梨糖醇10份,甘油9份,月桂醇硫酸酯钠3份,香精1份,纤维素胶1份,焦磷酸钠0.2份,糖精钠0.2份,苯甲酸钠0.02份,维生素C 0.2份,维生素E 0.5份,SOD 0.05份,CPP 0.02份。
3.根据权利要求1所述的一种含聚谷氨酸的口腔护理牙膏的制备方法,其特征在于,所述γ-聚谷氨酸的制备方法包括以下步骤:
(1)菌种活化:将地衣芽孢杆菌接种在斜面培养基上,37℃培养16小时,反复活化2-3次,得到成熟的斜面种子;
(2)种子液的制备:将步骤(1)中活化后的斜面种子接种于装有50ml液体种子培养基的500ml三角瓶中,37℃,220rpm培养16小时至对数生长期,得到种子液;
(3)发酵罐发酵:将步骤(2)中的种子液以培养基体积分数5%的接种量接种于发酵罐中,30L发酵罐发酵培养基装液量为15L,初始发酵温度38-40℃,罐压0.01-0.05Mpa,通风量0.5-1.0vvm,转速400-600rpm;当发酵液OD660在0.550-0.600时,并控制发酵温度30-35℃,调节转速至200-350rpm,通风量1.5-2.0vvm ,控制pH在7.2-7.5,发酵2-6小时,得到含γ-聚谷氨酸的发酵液;
(4)聚谷氨酸的提取:将步骤(3)中得到的发酵液离心去菌体,将上清液加入5倍上清液体积的乙醇,沉析出聚谷氨酸,过滤后得到γ-聚谷氨酸,然后在复溶于上清液等体积的蒸馏水中,利用喷雾干燥方法制备出干粉γ-聚谷氨酸。
4.根据权利要求3所述的一种含聚谷氨酸的口腔护理牙膏的制备方法,其特征在于,所述步骤(2)中种子培养基组成为:葡萄糖30 g/L,酵母膏7 g/L,胰蛋白胨10 g/L,K2HPO4 0.5g/L,MgSO4•7H2O 0.5 g/L,pH7.2,余量纯净水。
5.根据权利要求3所述的一种含聚谷氨酸的口腔护理牙膏的制备方法,其特征在于,所述步骤(3)中发酵培养基组分为:葡萄糖80 g/L,氯化钠10g/L,MgSO4•7H2O 1g/L,无水氯化钙1g/L,FeSO4•7H2O 0.02g/L,KCl 0.5g/L,精氨酸 0.8g/L,组氨酸0.5g/L,苏氨酸0.5g/L,甲硫氨酸0.4g/L,谷氨酰胺 0.5g/L,胆碱 0.31g/L,吡哆醇0.001g/L。
6.一种含聚谷氨酸的口腔护理牙膏,其特征在于:由权利要求1-5任意一项的制备方法得到。
