Abstract
Se describe una célula T que coexpresa un primer receptor antigénico (CAR) y un segundo CAR en la superficie celular, y cada CAR comprende: (i) un dominio de unión a antígenos, (ii) un espaciador, (iii) un dominio transmembranal; y (iv) un endodominio, donde los dominios de unión a antígeno del primer y segundo CAR se unen a diferentes antígenos, y en donde uno del primer o segundo CAR es un CAR activador que comprende un endodominio activador, y el otro CAR es un CAR inhibitorio que comprende un endodominio inhibitorio de ligación encendida que comprende un dominio de tirosina fosfatasa de una proteína tirosina fosfatasa que contiene el dominio homologo Src (SH2); una secuencia de ácido nucleico que codifica dichos primer y segundo CAR; un kit que comprende las secuencia de ácido nucleico que codifican dichos primer y segundo CAR; un kit que comprende un primer y un segundo vector que comprenden dichas secuencias de ácido nucleico; un método para elaborar dicha célula T; una composición farmacéutica que comprende dichas células T y un vehículo, diluyente o excipiente farmacéuticamente aceptable; el uso de diha composición farmacéutica para preparar un medicamento útil para tratar y/o prevenir el cáncer; el uso de dichas células T para preparar un medicamento para tratar y/o prevenir el cáncer; una célula asesina natural (NK) que coexpresa un primer receptor antigénico (CAR) y un segundo CAR en la superficie celular, y cada CAR comprende: (i) un dominio de unión a antígenos, (ii) un espaciador, (iii) un dominio transmembranal; y (iv) un endodominio, donde los dominios de unión a antígenos del primer y segundo CAR se unen a diferentes antígenos, y en donde uno del primer o segundo CAR es un CAR activador que comprende un endodominio activador, y el otro CAR es un CAR inhibitorio que comprende un endodominio inhibitorio de ligación encendida que comprende un dominio de tirosina fosfatasa de una proteína tirosina fosfatasa que contiene el dominio homologo Src (SH2); y una composición de células que comprende las células T que expresan CAR y/o células NK que expresan CAR de acuerdo con la reivindicación 20, elaborada al transducir una muestra de sangre ex vivo con un ácido nucleico que codifica el primer y segundo CAR.
