Abstract

Patente de Invenção: "MéTODO DE REALIZAçãO DETRANSGêNESE". A invenção provê um método para a geraçãode animais e células transgênicos através da co-inserção de ácidonucléico e um núcleo em um oócito não-fertilizado. De preferência,a co-inserção é através de microinjeção e com mais preferênciaatravés de microinjeção piezoeletricamente atuada. Os embriõesde expressão de transgene (tg) são aqui produzidos seguindo aco-injeção de oócitos de camundongos não-fertilizados comcabeças de espermatozóide e DNA exógeno codificando ou umaproteína verde fluorescente (GFP) ou um repórter ß-galactosidase.O oócito microinjetado pode ser deixado desenvolver em célulasdiferenciadas ou células de origem; em um embrião in vitro antesda transferência a uma mãe hospedeira substituta, ou ele pode sertransferido diretamente para uma mãe hospedeira substituta. Odesenvolvimento embriónico pode ocorrer até o final, de modo quea prole possua modificações transgênicas que possam alterar assuas características (fenótipo) e são, por sua vez, transmitidas paraa prole.
