The invention provides a method for the generation of animals and transgenic cells through the co-insertion of nucleic acid and a nucleus in an unfertilized oocyte. Preferably, co-insertion is through microinjection and more preferably through piezoelectrically operated microinjection. Transgene expression (tg) embryos are produced here following the co-injection of unfertilized mice oocytes with sperm heads and exogenous DNA encoding either a fluorescent green protein (GFP) or a ß-galactosidase reporter. The microinjected oocyte can be allowed to develop in differentiated cells or cells of origin; in an in vitro embryo prior to transfer to a surrogate host mother, or it can be transferred directly to a surrogate host mother. Embryonic development can occur until the end, so that the offspring has transgenic modifications that can alter their characteristics (phenotype) and are, in turn, transmitted to the offspring.
