Abstract

<B>LINHAGEM CELULAR PRODUTORA DE ERITROPOETINA HUMANA RECOMBINANTE E A ERITROPOETINA HUMANA RECOMBINANTE PRODUZIDA POR ESTA CéLULA<D>. O gen codificante para eritropoetina humana (EPO) foi obtido a partir de DNA genómico humana. O gen utilizado não inclui fragmentos de regiões flanqueantes 5' e 3' do gen de EPO que não codificam para a proteína. O gen foi clonado em um plasmídeo de expressão para células eucarióticas que tem como únicos elementos de controle da expressão o promotor primário de vírus SV40 e seu sinal de poliadenilação. As células recombinantes resultantes da transfecção com as construções genéticas utilizadas produz inesperadamente mais de 50 mg de EPO recombinante por litro de meio de cultivo por dia.
