Abstract
Polinucleótido aislado adecuado para la expresión condicional de un gen efector en una línea germinal masculina de artrópodos, que comprende: una primera unidad de expresión que comprende un gen efector y un promotor para dicho gen operativamente unido a dicho gen y una segunda unidad de expresión que comprende una secuencia codificante para un factor de transcripción y un elemento regulador corriente arriba unido operativamente a dicha secuencia, donde el factor de transcripción es capaz de actuar sobre el promotor en la primera unidad de expresión para dirigir la expresión del gen efector, donde el elemento regulador corriente arriba comprende: un promotor para el factor de transcripción, donde el promotor es de matotopetli (topi), always early (aly) o b-2 tubulin (B2T), o un homólogo del mismo y un 5 UTR adyacente a un sitio de inicio para la secuencia codificante del factor de transcripción, donde el 5 UTR es de la proteína de choque térmico hsp83, topi, aly, o B2T, o un homólogo de la misma, donde, cuando tanto el promotor como el 5 UTR son de B2T, las señales de retraso transcripcional en la secuencia B2T se eliminan del promotor o del 5 UTR y donde el elemento regulador corriente arriba impulsa la expresión del factor de transcripción de tal manera que la proteína del factor de transcripción a su vez impulsa la transcripción del gen efector antes de la meiosis; método para introducir de manera recombinante una proteína efectora en una gónada o esperma, excluyendo la gónada o esperma transformado por este método; método de supresión de una población de artrópodos nativos, excluyendo la población de artrópodos nativos y las gónadas transformadas por este método; método para retardar o revertir la resistencia a insecticidas y/o bioplaguicidas en una población de artrópodos, excluyendo la población de artrópodos y las gónadas transformadas por este método; método de control de calidad, excluyendo los individuos analizados en este método; y método para determinar el estado de apareamiento de un artrópodo hembra. Reivindicación 23: El polinucleótido aislado de acuerdo con la reivindicación 22, caracterizado porque la secuencia codificante heteróloga comprende una secuencia que codifica una proteína que tiene un efecto letal, perjudicial o esterilizante, y/o una secuencia que codifica un marcador seleccionable. Reivindicación 24: El polinucleótido aislado de acuerdo con la reivindicación 22 o 23, caracterizado porque la mediación del empalme alternativo es específica del sexo y confiere una letalidad específica femenina. Reivindicación 25: El polinucleótido aislado de acuerdo con cualquiera de las reivindicaciones 22 - 24, caracterizado porque la tercera unidad de expresión es inducible y la inducción se produce por provisión o ausencia de una entidad química, tal como tetraciclina o uno de sus análogos incluyendo doxiciclina. Reivindicación 30: Un método de control de calidad, excluyendo los individuos analizados en este método, caracterizado porque comprende incluir una proteína fluorescente indicadora como, o además de, la proteína efectora codificada por el polinucleótido aislado de acuerdo con cualquiera de las reivindicaciones 1 - 26, de modo que los individuos en que se ha inducido o des-reprimido la expresión de la proteína efectora se hacen visibles en longitudes de onda adecuadas.
