Abstract
Reivindicación 1: Una composición para la crioconservación de células derivadas de tejido de cordón umbilical, en donde la composición comprende: los aminoácidos L-arginina, L-glutamina, glicina, L-isoleucina, L-leucina, L-lisina, HCl, L-metionina, L-fenilalanina, L-serina, L-treonina, L-triptofano y L-valina; las vitaminas D-calcio pantotenato, cloruro de colina, ácido fólico, niacinamida, piridoxina, HCl, riboflavina y tiamina, HCl; las sales inorgánicas cloruro de potasio, USP, bicarbonato de sodio, USP, cloruro de sodio, USP y fosfato de sodio, monobásico, H₂O; las sales inorgánicas dihidrato de cloruro de calcio (CaCl₂.2H₂O), cloruro de magnesio (MgCl₂.6H₂O), bicarbonato de potasio (KHCO₃) y monofosfato de potasio (KH₂PO₄) y DMSO, ácido lactobiónico, glutatión, dextrosa, sacarosa y manitol. Reivindicación 7: La composición de la reivindicación 1, en donde las células derivadas de tejido de cordón umbilical humano se aíslan de tejido de cordón umbilical humano sustancialmente libre de sangre, tienen la capacidad de autorrenovación y de expansión en cultivo, tienen el potencial de diferenciarse, carecen la producción de CD117 o CD45, expresan y no expresan hTERT o telomerasa. Reivindicación 8: La composición de la reivindicación 7, en donde las células derivadas de tejido del cordón umbilical comprenden, además, una o más de las siguientes características: expresan el receptor de lipoproteína de baja densidad oxidado 1, reticulón, ligando del receptor de quimocina 3 y/o proteína quimiotáctica de no granulocitos; expresan CD31 o CD34; expresan, en relación con un fibroblasto humano, células madre mesenquimales o células de la médula ósea de la cresta iliaca, niveles elevados de interleucina 8 o reticulón 1; y expresan CD10, CD13, CD44, CD73 y CD90. Reivindicación 9: Un estuche que comprende la composición de la reivindicación 1 y células derivadas de tejido del cordón umbilical, en donde las células derivadas de tejido de cordón umbilical humano se aíslan de tejido de cordón umbilical humano sustancialmente libre de sangre, tienen la capacidad de autorrenovación y de expansión en cultivo, tienen el potencial de diferenciarse, carecen la producción de CD117 o CD45, expresan y no expresan hTERT o telomerasa. Reivindicación 13: Un método para crioconservar células derivadas de tejido de cordón umbilical humano, el método comprende: proporcionar una formulación que comprende la composición de crioconservación de la reivindicación 1 y células derivadas del tejido del cordón umbilical; enfriar la composición de crioconservación de una temperatura inicial de aproximadamente 4ºC a una velocidad de 1,0ºC/min. hasta que la composición alcance una temperatura de aproximadamente -45,0ºC; y además, enfriar la composición de crioconservación hasta que la composición alcance una temperatura de almacenamiento de aproximadamente -120,0ºC; en donde las células derivadas de tejido de cordón umbilical humano se aíslan de tejido de cordón umbilical humano sustancialmente libre de sangre, tienen la capacidad de autorrenovación y de expansión en cultivo, tienen el potencial de diferenciarse, carecen la producción de CD117 o CD45, expresan y no expresan hTERT o telomerasa. Reivindicación 17: Un método para crioconservar células derivadas de tejido de cordón umbilical humano, el método comprende: proporcionar una formulación que comprende la composición de crioconservación de la reivindicación 3 y células derivadas del tejido del cordón umbilical enfriar la composición de crioconservación de una temperatura inicial de aproximadamente 4ºC a una velocidad de -1,0ºC/min. hasta que la composición alcance una temperatura de aproximadamente -45,0ºC; y además, enfriar la composición de crioconservación hasta que la composición alcance una temperatura de almacenamiento de aproximadamente -120,0ºC; en donde las células derivadas de tejido de cordón umbilical humano se aíslan de tejido de cordón umbilical humano sustancialmente libre de sangre, tienen la capacidad de autorrenovación y de expansión en cultivo, tienen el potencial de diferenciarse, carecen la producción de CD117 o CD45, expresan y no expresan hTERT o telomerasa. Reivindicación 19: Un método para crioconservar células derivadas de tejido de cordón umbilical humano, el método comprende: proporcionar una formulación que comprende la composición de crioconservación de la reivindicación 6 y células derivadas del tejido del cordón umbilical; enfriar la composición de crioconservación de una temperatura inicial de aproximadamente 4ºC a una velocidad de -1,0ºC/min. hasta que la composición alcance una temperatura de aproximadamente -45,0ºC; y además, enfriar la composición de crioconservación hasta que la composición alcance una temperatura de almacenamiento de aproximadamente -120,0ºC; en donde las células derivadas de tejido de cordón umbilical humano se aíslan de tejido de cordón umbilical humano sustancialmente libre de sangre, tienen la capacidad de autorrenovación y de expansión en cultivo, tienen el potencial de diferenciarse, carecen la producción de CD117 o CD45, expresan y no expresan hTERT o telomerasa.
