Abstract

Se describe el uso de agentes que modulan la expresion de amiloide A en suero. Reivindicacion 1: Un método para diagnosticar el glaucoma, caracterizado porque comprende analizar una muestra biologica para determinar el nivel aberrante, bioactividad o mutaciones del gen que codifica a la proteína amiloide A sérica (SAA), su region promotora, o productos genéticos, en donde dicho gen que codifica SAA comprende la secuencia expuesta en SEC ID Ns:1 o SEC ID Ns:3, dicha region promotora comprende la secuencia expuesta en SEQ ID Ns:12 o SEQ ID Ns:13, y en donde SAA comprende la secuencia expuesta en SEC ID Ns:2 o SEQ ID Ns:4; en donde el nivel aberrantemente alto o bioactividad o mutaciones de los genes de SAA o los productos genéticos indica un diagnostico de glaucoma. Reivindicacion 4: Un método para diagnosticar el glaucoma en un paciente, comprendiendo dicho método: a) aislar ácido nucleico de las células provenientes de un paciente, b) poner en contacto a la muestra con uno o más cebadores los cuales se hibridan específicamente 5' y 3' a por lo menos un alelo de SEQ ID Ns:1, SEC ID Ns:3, SEQ ID Ns:12, o SEQ ID Ns:13, bajo condiciones tales que se produce la hibridacion y la amplificacion del alelo; y c) detectar el producto de amplificacion; en donde el nivel aberrante o mutaciones de SEC ID Ns:1 o SEC ID Ns:3 en la muestra indica un diagnostico de glaucoma. Reivindicacion 5: Un método para identificar agentes potencialmente utiles para tratar el glaucoma, caracterizado porque comprende: a) mezclar una sustancia candidato con células que expresan SAA (SEC ID Ns:1 o SEC ID Ns:2) o células que contienen gen promotor/ informante de SAA de modo que el gen reportero es expresado; b) determinar el nivel de proteína SAA (SEC ID Ns:2 o SEC ID Ns:4) o el nivel de expresion genética en las células; en donde un incremento o disminucion de la produccion de la proteína de SAA o expresion genética en presencia de dicha sustancia candidato indica un agente potencialmente util para el tratamiento del glaucoma.
