Abstract

Nuevas proteinas de shock por calor (HSPs) Streptococcus pneumoniae, Streptococcus pyogenes y Streptococcus agalactiae que tienen masas molecularesaparentes de 70-72 kDa, polipéptidos inmunológicamente relacionados, las secuencias de nucleótidos y aminoácidos derivadas de HSP72 de S.pneumoniae (SECID Ns: 4, SEC ID Ns: 5), las secuencias de nucleótidos y aminoácidos derivados de HSP70 de S. pyogenes (SEC ID Ns: 19; SEC ID Ns: 20), las secuencias denucleótidos y aminoácidos derivadas HSP70 de S. agalactiae (SEC ID Ns: 21; SEC ID Ns: 22), anticuerpos que se ligan con HSPs y métodos de ADN recombinantes,vectores de expresión y huéspedes recombinantes para la producción de los HSPs y los polipéptidos inmunológicamente relacionados. Los polipéptidos, lassecuencias de ADN y los anticuerpos de esta invención proveen nuevos medios para el diagnóstico, prevención y/o tratamiento de las enfermedades causadaspor estreptococos. Se describe un nuevo método para producir anticuerpos contra HSP72 y péptidos inmunológicamente relacionados. Estos péptidos son usados enla preparación de una vacuna y en un método de prevención de una infección causada por S. pneumoniae. Finalmente, también se describen métodos dedetección dela presencia de S. pneumoniae en muestras biológicas, haciendo uso de los polipéptidos bacterianos HSP72 y polipéptidos inmunológicamenterelacionados, de anticuerpos dirigidos contra el mismo o de una sonda de ADN contra los ácidos nucleicos quecodifican para los polipéptidos HSP72.New heat shock proteins (HSPs) Streptococcus pneumoniae, Streptococcus pyogenes and Streptococcus agalactiae having apparent molecular masses of 70-72 kDa, immunologically related polypeptides, nucleotide and amino acid sequences derived from S. pneumoniae HSP72 (SECID Ns: 4, Ns: 4, SEQ ID Ns: 5), the nucleotide and amino acid sequences derived from S. pyogenes HSP70 (SEQ ID Ns: 19; SEQ ID Ns: 20), the nucleotide and amino acid sequences derived HSP70 from S. agalactiae (SEQ ID Ns: 21; SEQ ID Ns: 22), antibodies that bind to HSPs and recombinant DNA methods, expression vectors and recombinant hosts for the production of the HSPs and immunologically related polypeptides. The polypeptides, DNA sequences, and antibodies of this invention provide new means for the diagnosis, prevention, and / or treatment of diseases caused by streptococci. A new method for producing antibodies against HSP72 and immunologically related peptides is described. These peptides are used in the preparation of a vaccine and in a method of preventing an infection caused by S. pneumoniae. Finally, methods for detecting the presence of S. pneumoniae in biological samples are also described, making use of the bacterial HSP72 polypeptides and immunologically related polypeptides, of antibodies directed against it or of a DNA probe against the nucleic acids that encode for the HSP72 polypeptides.
