Abstract
. Oppfinnelsen angår 4-fenyl-6-substituert-pyrimidin-2-karbonitrilderivater som har Formel I, Formel I hvor R representerer 1-3 eventuelle substituenter uavhengig valgt fra (C1-4)alkyl (eventuelt substituert med ett eller flere halogenatomer), (C1-4)alkyloksy (eventuelt substituert med ett eller flere halogenatomer) og halogen; X er NR1, O eller S; R1 er H eller (C1-4)alkyl; Y er (C1-4)alkyl, benzyl eller (C2-6)alkyl, substituert med en gruppe valgt fra OH, (C1-4)alkyloksy, NR2R3, en 4-8 leddet mettet heterocyklisk ring omfattende 1 eller 2 heteroatomer valgt fra O, S og NR4 og en 5 eller 6-leddet aromatisk heterocyklisk gruppe omfattende 1-4 N atomer; eller R1 og Y danner sammen med nitrogenet som de er bundet til en 5-8 leddet mettet heterocyklisk ring, eventuelt omfattende et ytterligere heteroatom valgt fra O, S, NR4 og NO; idet ringen er eventuelt substituert med NR2R3 eller med 1 til 4 (C1-3)alkylgrupper; R2 og R3 er uavhengig H eller (C1-4)alkyl; eller R2 og R3 danner sammen med nitrogenet som de er bundet til en 4-8 leddet mettet heterocyklisk ring, eventuelt omfattende et ytterligere heteroatom valgt fra O, S og NR4; R4 er H, (C1-6)alkyl, (C3-6)cykloalkyl, (C3-6)cykloalkyl(C1-4)alkyl, (C1-4)alkyloksy-(C1-4)alkyl, benzyl, amido-(C1-4)alkyl, (C1-6)alkyloksykarbonyl(C1-4)alkyl eller karboksy(C1-4)alkyl; eller et farmasøytisk akseptabelt salt derav, farmasøytiske preparater omfattende disse, så vel som anvendelse av disse 4-fenyl-6-substituert-pyrimidin-2-karbonitrilderivater for fremstilling av et medikament for behandling av cathepsin K og cathepsin S relaterte lidelser, f.eks. osteoporose, aterosklerose, inflammasjon og immunlidelser så som revmatoid artritt og kronisk smerte.
