Abstract

La presente invención proporciona métodos para diferenciar células pluripotentes en célula beta mediante el use de suspensión de agrupaciones; los métodos de la invención usan control de uno o más de pH, concentración de células, y concentración del retinoide para generar una población de células que coexpresan PDX1MKX6.1 casi homogéneas mediante la supresión de la expresión precoz de NGN3 y la promoción de la expresión de NKX6.1; además, la población casi homogénea de células que coexpresan PDX1/NKX6.1 puede diferenciarse, además, in vitro para formar una población de células endocrinas pancreáticas que coexpresan PDX1, NKX6.1, insulina y MAFA.
