Abstract
Se trata del uso innovador de un sistema de retención de célula basado en membrana externa conjuntamente con cultivo celular de perfusión para expresión celular mejorada de proteínas recombinantes, particularmente proteínas de coagulación, tales como rFVIII, rFVIII con Dominio B Eliminado, rFIX o rFVII/rFVIIa. El uso de tal sistema en alta densidad de celula resulta en un cultivo celular más homogéneo debido a las fuerzas mecánicas inducidas durante la operación del sistema de retención, tales como la circulación celular inducida por bombeo a través de las fibras, además que la retención de algunos componentes celulares y de medio permite un ambiente ideal para las células. El uso del método proporcionado resulta en mejoras en productividad celular y en productividad volumétrica. El sistema de retención de célula basado en membrana es descartable y cuando es conectado a un biorreactor descartable, el mismo permite que todo el sistema de perfusión sea descartable cuando el medio y la recolección son desechados en sacos descartables. El sistema biorreactor de perfusión descartable puede ser operado como un sistema cerrado y no requiere limpieza, vaporización o el uso de una instalación con tubería rígida.
