Abstract

Aquí se proveen ácidos nucleicos, composiciones y métodos que permiten la escisión de uno o más loci del genoma de una célula hospedera. En particular, aquí se provee un constructo de ácido nucleico excindible que comprende, en una orientación 5' a 3': un primer ácido nucleico de repetición en tándem, un primer sitio de reconocimiento de endonucleasa de inserción alélica, un ácido nucleico objetivo, un segundo sitio de reconocimiento de endonucleasa de inserción alélica, y un segundo ácido nucleico de repetición en tándem. En algunas modalidades, el constructo de ácido nucleico excindible es integrado en el genoma de la célula hospedera, y el ácido nucleico objetivo puede ser escindido del genoma de la célula hospedera al poner en contacto los sitios de reconocimiento de endonucleasa de inserción alélica con una o más endonucleasas de inserción alélica apropiadas.
