Abstract
Un método para la cuantificación de una cepa de vacuna y/o una cepa virulenta del Serotipo-1 de Virus de Enfermedad de Marek (MDV-1) en una muestra de un ave que comprende las etapas de: (i) proporcionar una muestra biológica del ave y opcionalmente aislar el ácido nucleico de la muestra biológica; (ii) someter la muestra biológica de (i) a PCR cuantitativo de tiempo real (qPCR) que comprende: (a) amplificación de la región del gen pp38 dentro de la muestra de ácido nucleico de (i), tal región contiene una diferencia de polimorfismo de nucleótido simple (SNP) consistente entre las cepas de vacuna y virulenta del MDV-1; y (b) poner en contacto el ácido nucleico amplificado de (a) con una sonda de ácido nucleico detectable específica para el SNP de la cepa de vacuna del MDV-1 y/o una sonda de ácido nucleico detectable específica para el SNP de la cepa virulenta del MDV-1; e (iii) medir los cambios en la señal detectable producida por la sonda de (ii). Los métodos también se proporcionan para la cuantificación absoluta de los virus de vacuna y virulencia.
