Abstract

La presente invención describe un procedimiento para la preparación de rh(PTH) (1-34) también conocido como teriparatida por medio de la construcción de un nucleótido novedoso, como un fragmento NcoI.lXol como se establece en SEQ. ID. NO.: 1 que codifica una proteína de fusión quimérica como se establece en SEQ. ID. NO.: 2 que comprende un miembro de fusión que consiste en 41 aminoácidos que pertenecen al gen ß- galactosidasa (LacZ) Escherichia coIi, un sitio de escisión endopeptidasa, fragmento del gen rhPTH(1-34), clonando dicho nucleótido en un vector de expresión bajo el control del promotor T7, transformando Escherichia coli con dicho vector y expresando la proteína de fusión quimérica en fermentación alimentada por lotes; la presente invención además describe una inducción de lactosa con baja velocidad de alimentación para expresión optimizada de rhPTH(1-34)en Escherichia coli; la presente invención también describe un procedimiento de purificación ortogonal de dos pasos, novedoso y único para rhPTH(1-34) que comprende una cromatografía de intercambio catiónico opcionalmente seguida por cromatografía preparativa seleccionada de HIC o RP-HPLC para producir una proteína objetivo con pureza del =99%; la presente invención describe un método ambientalmente benigno, efectivo en cuanto a costo y simple para producir rhPTH(1-34) de alta pureza.
