本発明の一実施形態に係るポリ(3-ヒドロキシアルカノエート)の製造方法(以下、「本製造方法」と称する。)は、重量平均分子量が10万~70万であるポリ(3-ヒドロキシアルカノエート)の製造方法であり、(a)ポリ(3-ヒドロキシアルカノエート)を含有する菌体を含む培養液にアルカリ水溶液を添加して、pHを8.0~12.0に調整する工程、(b)前記工程(a)で得られた培養液に酵素を添加して、前記菌体を酵素処理する工程、および(c)前記工程(b)で得られた培養液にアルカリ水溶液を添加して、pHを10.0~12.0に調整する工程、を含むことを特徴とする。
・熱安定性に優れていること。
・分子量分布の観点から、同程度の分子量のP3HAを多く含むこと(単分散)。
・低分子量のP3HAを製造した場合、得られたP3HA粉体の表面が非多孔質となること。
本製造方法は、以下の工程(a)~(c)を含む、重量平均分子量が10万~70万であるポリ(3-ヒドロキシアルカノエート)のP3HAの製造方法である。
工程(a)は、P3HAを含有する菌体を含む培養液にアルカリ水溶液を添加して(「アルカリ処理」ともいう。)、pHを8.0~12.0に調整する工程である。すなわち、pHを8.0~12.0に調整することで、P3HAを加水分解し、P3HAの分子量を調整する工程であるともいえる。当該工程によれば、P3HAを含有する菌体において、菌体中で(換言すると、当該P3HAが結晶化していない(アモルファスな)状態で)、当該P3HAの分子量を調整することができる。工程(a)は、P3HAを含有する菌体を含む培養液を、pHが8.0~12.0の状態に一定時間維持する工程であるとも言える。また、工程(a)は、分子量調整工程であるとも言える。
本明細書において、「P3HA」とは、以下の式(2):

(式中、Rは、CnH2n+1で表されるアルキル基であり、nは、1~15の整数である。)で示される1種以上の単位からなる共重合体の総称を意図する。
本明細書において、「P3HAを含有する菌体」とは、P3HAを生産する微生物を意図する。すなわち、本発明の一実施形態において、P3HAは、微生物により産生される。本製造方法において用いられる微生物は、細胞内にP3HAを生成し得る微生物である限り、特に限定されない。例えば、天然から単離された微生物や菌株の寄託機関(例えば、IFO、ATCC等)に寄託されている微生物、またはそれらから調製し得る変異体や形質転換体等を使用できる。より詳しくは、例えば、カプリアビダス(Cupriavidus)属、アルカリゲネス(Alcaligenes)属、ラルストニア(Ralstonia)属、シュウドモナス(Pseudomonas)属、バチルス(Bacillus)属、アゾトバクター(Azotobacter)属、ノカルディア(Nocardia)属、アエロモナス(Aeromonas)属の菌等が挙げられる。中でも、アエロモナス属、アルカリゲネス属、ラルストニア属、またはカプリアビダス属に属する微生物が好ましい。特に、アルカリゲネス・リポリティカ(A.lipolytica)、アルカリゲネス・ラトゥス(A.latus)、アエロモナス・キャビエ(A.caviae)、アエロモナス・ハイドロフィラ(A.hydrophila)、カプリアビダス・ネカトール(C.necator)等の菌株がより好ましく、カプリアビダス・ネカトールが最も好ましい。
本発明の一実施形態において、アルカリ水溶液は、塩基性化合物を含む水溶液である。アルカリ水溶液に含まれる塩基性化合物としては、特に限定されないが、例えば、水酸化ナトリウム、水酸化カリウム等のアルカリ金属またはアルカリ土類金属の水酸化物;炭酸ナトリウム、炭酸カリウム等の金属炭酸塩;リン酸ナトリウム、リン酸カリウム、リン酸水素ナトリウム、リン酸水素カリウム等の金属リン酸塩または金属リン酸水素塩等が挙げられる。

上記式(3)および(4)中の各種パラメータは、実施例に記載の方法により測定される。
工程(b)は、前記工程(a)で得られた培養液に酵素を添加して、前記菌体を酵素処理する工程である。当該工程によれば、前記菌体由来の不純物(細胞壁、タンパク質等)を破壊および除去することで、前記菌体からP3HAを効率的に回収できる。
本明細書において、「アルカリ性タンパク質分解酵素」とは、アルカリ環境下(例えばpH8.5の溶液中)でタンパク質を分解する活性を有するタンパク質分解酵素を意図する。
本発明の一実施形態において、アルカリ性タンパク質分解酵素処理とは、溶菌酵素を前記培養液に添加し、前記菌体を酵素処理する工程である。
本製造方法は、工程(a)と(b)との間に、さらに工程(a’)を含み得る。
工程(c)は、前記工程(b)で得られた培養液にアルカリ水溶液を添加して、pHを10.0~12.0に調整する工程である。当該工程によれば、P3HAの分子量をさらに小さく調整し得るとともに、菌体を破砕する際に、菌体由来の不純物(核酸、タンパク質等)を分散および溶解することで、高純度のP3HAを菌体から分離することができる。工程(c)は、アルカリ洗浄工程とも言える。
本製造方法は、工程(c)の後に、さらに工程(c’)を含み得る。工程(c’)は界面活性剤処理工程であるとも言える。
本発明の一実施形態において、本製造方法は、工程(d)をさらに含み得る。
本発明の一実施形態において、本製造方法は、工程(e)をさらに含み得る。
・工程(e1):P3HA水性懸濁液に分散剤を添加する工程
・工程(e2):P3HA水性懸濁液のpHを7以下に調整する工程
工程(e1)と工程(e2)とを実施する順番は、特に限定されないが、工程(e2)におけるP3HAの凝集が抑制され、よりP3HAの分散安定性に優れた水性懸濁液が得られる観点で、工程(e1)の後に工程(e2)を実施することが好ましい。
本発明の一実施形態において、本製造方法は、工程(f)をさらに含み得る。
本発明の一実施形態に係るP3HA粉体(以下、「本P3HA粉体」と称する。)は、重量平均分子量が10万~60万であり、残タンパク質量が2000ppm以下であり、粒子表面が非多孔質であり、かつ、以下の式(1)で示される重量平均分子量保持率が50%以上である。

本P3HA粉体は、加工時に異物が発生せず、生産性に優れ、かつ、高い熱安定性を有するため、種々の分野において極めて有用である。

実施例および比較例における測定および評価を、以下の方法で行った。
熱安定性は、160℃で20分間加熱した後のP3HAの重量平均分子量保持率として評価した。P3HAの重量平均分子量保持率は、以下の式(1)によって算出した:

P3HA粉体10mgを、クロロホルム10mLに溶解させた後、不溶物を濾過により除いた。この溶液(濾液)を、「Shodex K805L(300x8mm、2本連結)」(昭和電工社製)を装着した島津製作所製GPCシステムを用い、クロロホルムを移動相として重量平均分子量を測定した。分子量標準サンプルには、昭和電工(株)製Shodex K-804(ポリスチレンゲル)を用いた。
P3HA粉体を、160℃で7分間予熱処理した後、5MPa加圧下、160℃で20分間加熱し、P3HA樹脂シートを作製した。当該P3HA樹脂シート10mgをクロロホルム10mLに溶解させたこと以外は、(加熱する前のP3HAの重量平均分子量の測定)に記載したのと同様の手順で、加熱した後のP3HAの重量平均分子量を測定した。
初期分子量を以下の手順によって測定した:不活化後のP3HAを含む培養液を蒸留水で希釈し、遠心分離を行った後、上清を除去した。得られた沈殿物(P3HA)にエタノールを加え、エタノールに分散させ、遠心分離を行った。上清を除去し、真空乾燥器で1時間以上乾燥させ、沈殿物を完全に乾燥させ、乾燥体(P3HA粉体)を得た。得られたP3HA粉体10mgを、クロロホルム10mLに溶解させた後、不溶物を濾過により除いた。この溶液(濾液)を、「Shodex K805L(300x8mm、2本連結)」(昭和電工社製)を装着した島津製作所製GPCシステムを用い、クロロホルムを移動相として初期分子量を測定した。分子量標準サンプルには、昭和電工(株)製Shodex K-804(ポリスチレンゲル)を用いた。
最終分子量および数平均分子量を以下の手順によって測定した:P3HA粉体10mgを、クロロホルム10mLに溶解させた後、不溶物を濾過により除いた。この溶液(濾液)を、「Shodex K805L(300x8mm、2本連結)」(昭和電工社製)を装着した島津製作所製GPCシステムを用い、クロロホルムを移動相として最終分子量および数平均分子量を測定した。分子量標準サンプルには、昭和電工(株)製Shodex K-804(ポリスチレンゲル)を用いた。
P3HAの収率を以下の手順によって測定した:
(a)後述する分子量調整前の培養液と分子量調整後の培養液とをそれぞれ2gずつ採り、水分計(株式会社エー・アンド・デイ製ML-50)を用いて固形分濃度を測定した。
収率(%)=(分子量調整後の培養液のP3HA含量/分子量調整前の培養液のP3HA含量)×100×(分子量調整前の培養液の固形分濃度(%)/分子量調整後の培養液の固形分濃度(%)) ・・・(4)。
残タンパク質量は、BCA Protein Assay Kit(Thermo Fisher Scientific社製)を用いて測定した。
SEM試料台の上にカーボンテープを貼り、その上から外添剤を含有したP3HA粉体、または比較例となるP3HA粉体を振りかけ後、過剰な粉体をブロアーで除去した。前処理は、オスミウム・プラズマコーターを用いて、20nmの厚みでコーティングを行った。上記P3HA粉体の前処理は、フィルジェン製 OPC60A-Gオスミウム・プラズマコーターを用いて行った。SEM観察は、日立ハイテクフィールディング製 S-4800を用いて行った。
(菌体培養液の調製)
国際公開第WO2019/142717号に記載のラルストニア・ユートロファを、同文献の段落〔0041〕~〔0048〕に記載の方法で培養し、P3HAを含有する菌体を含む菌体培養液を得た。P3HAの繰り返し単位の組成比(3-ヒドロキシブチレート単位/3-ヒドロキシヘキサノエート単位の組成比)を測定したところ、99.9/0.1~96.0/4.0(mol/mol)であった。
上記で得られた菌体培養液を内温70℃で8時間加熱・攪拌処理し、滅菌処理を行った。
上記で得られた不活化した培養液(不活化培養液)に対して30%水酸化ナトリウムを用いてpHを9.0±0.2に調整し、内温を75±2℃として、菌体中のP3HAの分子量調整を12時間行った。P3HAの収率は、97%であった(表1)。
上記で得られた分子量調整後の液(分子量調整後液)に対して95%硫酸を添加してpHを7.0±0.2に調整した。IW(工業用水)を添加して固形分濃度を18%に調整した後、細胞壁中の糖鎖(ペプチドグリカン)を分解する酵素(溶菌酵素)であるリゾチーム(富士フイルム和光純薬(株)社製)を添加し、50℃で2時間保持した。その後、アルカリ性タンパク質分解酵素であるアルカラーゼ2.5L(Novozyme社製)を添加し、次いで、50℃で30%水酸化ナトリウムを添加して、pH8.5に調整しながら2時間保持した。
上記で得られた酵素処理液に対して30%水酸化ナトリウムを添加してpH8.5に調整した(アルカリ洗浄)。次いで、上記酵素処理液に対して0.6~1.0wt%になるようにドデシル硫酸ナトリウム(SDS、花王製)を添加した(界面活性剤処理)。その後、pHが11.0±0.2になるように30%水酸化ナトリウムを用いて調整するとともに、pH11.0の水酸化ナトリウム水溶液を添加して、上記酵素処理液に対して2倍になるよう希釈した。
上記で得られたP3HA水性懸濁液に、エチレンオキサイド/プロピレンオキサイド共重合体非イオン性分散剤(ポリエチレンオキサイド分子量8000、ポリプロピレンオキサイド分子量2000、商品名プロノン208)を1.0phr(水性懸濁液中に存在するP3HA100重量部に対して1重量部)添加し、混合した。この液を120分間撹拌した後、P3HA水性懸濁液100gあたりに対して、10重量%硫酸を0.3ml添加し、P3HA水性懸濁液を得た。
(分子量調整)までは、実施例1と同様の方法で分子量調整後液を得た。
上記で得られた分子量調整後の液(分子量調整後液)に対してIW(工業用水)を添加して固形分濃度を18%に調整した。次いで、アルカリ性タンパク質分解酵素であるアルカラーゼ2.5L(Novozyme社製)を添加し、50℃で2時間保持した。次いで、細胞壁中の糖鎖(ペプチドグリカン)を分解する溶菌酵素であるリゾチーム(富士フイルム和光純薬(株)社製)を添加し、50℃で2時間保持した。その後、アルカラーゼ2.5L(Novozyme社製)を添加し、次いで、50℃で30%水酸化ナトリウムを添加して、pH8.5に調整しながら2時間保持した。以降は、実施例1と同様の方法でP3HA粉体を得た。また、実施例1と同様の方法で、各種測定を行った。
P3HAの繰り返し単位の組成比(3-ヒドロキシブチレート単位/3-ヒドロキシヘキサノエート単位の組成比)が96.0/4.0~92.0/8.0(mol/mol)であるP3HAを用いたこと以外は、実施例1と同様の方法でP3HA粉体を得た。また、実施例1と同様の方法で、各種測定を行った。
P3HAの繰り返し単位の組成比(3-ヒドロキシブチレート単位/3-ヒドロキシヘキサノエート単位の組成比)が92.0/8.0~87.0/13.0(mol/mol)であるP3HAを用いたこと以外は、実施例1と同様の方法でP3HA粉体を得た。また、実施例1と同様の方法で、各種測定を行った。
(不活化)までは、実施例3と同様の方法で不活化培養液を得た。
上記で得られた不活化培養液に対してIW(工業用水)を添加して固形分濃度を18%に調整した後、細胞壁中の糖鎖(ペプチドグリカン)を分解する酵素であるリゾチーム(富士フイルム和光純薬(株)社製)を添加し、50℃で2時間保持した。その後、タンパク質分解酵素であるアルカラーゼ2.5L(Novozyme社製)を添加し、次いで、50℃で30%水酸化ナトリウムを添加して、pH8.5に調整しながら2時間保持した。
上記で得られた酵素処理後液に対して30%水酸化ナトリウムを用いてpHを11.5±0.2に調整し、内温を50±2℃として、P3HAの分子量調整を12時間行った。以降は、実施例1と同様の方法でP3HA粒子を得た。また、実施例1と同様の方法で、各種測定を行った。なお、P3HAの収率は、(溶菌、濃縮)の直前(換言すれば、pHを11.5±0.2に調整し、内温を50±2℃として12時間保持した直後)に計測した。

(酵素処理)までは、実施例1と同様の方法で酵素処理液を得た。
上記で得られた酵素処理液に対して0.6~1.0wt%になるようにドデシル硫酸ナトリウム(SDS、花王製)を添加し、2倍希釈した。次いで、pHが8.5になるように30%水酸化ナトリウムを用いて調整した。以降は、実施例1と同様の方法でP3HA粒子を得た。また、実施例1と同様の方法で、各種測定を行った。
(不活化)までは、実施例1と同様の方法で不活化培養液を得た。

P3HAの繰り返し単位の組成比(3-ヒドロキシブチレート単位/3-ヒドロキシヘキサノエート単位の組成比)が92.0/8.0~87.0/13.0(mol/mol)であるP3HAを用いたこと以外は、比較例2と同様の方法でP3HA粉体を得た。また、実施例1と同様の方法で、各種測定を行った。
(分子量調整)で分子量調整を6時間行ったこと以外は、比較例3と同様の方法でP3HA粉体を得た。また、実施例1と同様の方法で、各種測定を行った。なお、P3HAの収率は、(溶菌、濃縮)の直前(換言すれば、pHを11.5±0.2に調整し、内温を50±2℃として6時間保持した直後)に計測した。
(分子量調整)で分子量調整を6時間行ったこと以外は、比較例4と同様の方法でP3HA粉体を得た。また、実施例1と同様の方法で、各種測定を行った。なお、P3HAの収率は、(溶菌、濃縮)の直前(換言すれば、pHを11.5±0.2に調整し、内温を50±2℃として6時間保持した直後)に計測した。
Claims (9)
- 重量平均分子量が10万~70万であるポリ(3-ヒドロキシアルカノエート)の製造方法であり、
(a)ポリ(3-ヒドロキシアルカノエート)を含有する菌体を含む培養液にアルカリ水溶液を添加して、pHを8.0~12.0に調整する工程、
(b)前記工程(a)で得られた培養液に酵素を添加して、前記菌体を酵素処理する工程、および
(c)前記工程(b)で得られた培養液にアルカリ水溶液を添加して、pHを10.0~12.0に調整する工程、
を含む、ポリ(3-ヒドロキシアルカノエート)の製造方法。 - 前記工程(b)における酵素処理が、アルカリ性タンパク質分解酵素処理、溶菌酵素処理、およびアルカリ性タンパク質分解酵素処理をこの順で行うことを含む、請求項1に記載のポリ(3-ヒドロキシアルカノエート)の製造方法。
- 前記工程(a)と(b)との間に、さらに
(a’)前記工程(a)で得られた培養液のpHを6.0~8.0に調整する工程、
を含み、
前記工程(b)における酵素処理が、溶菌酵素処理、およびアルカリ性タンパク質分解酵素処理をこの順で行うことを含む、請求項1または2に記載の製造方法。 - 前記工程(b)における酵素が、リゾチームおよびアルカラーゼである、請求項1~3のいずれか1項に記載のポリ(3-ヒドロキシアルカノエート)の製造方法。
- 前記工程(c)の後に、さらに
(c’)前記工程(c)で得られた培養液に界面活性剤を添加する工程、
を含む、請求項1~4のいずれか1項に記載のポリ(3-ヒドロキシアルカノエート)の製造方法。 - 前記工程(a)の反応時間が4時間~30時間である、請求項1~5のいずれか1項に記載のポリ(3-ヒドロキシアルカノエート)の製造方法。
- 前記ポリ(3-ヒドロキシアルカノエート)が、ポリ(3-ヒドロキシブチレート-コ-3-ヒドロキシヘキサノエート)である、請求項1~6のいずれか1項に記載のポリ(3-ヒドロキシアルカノエート)の製造方法。
- 重量平均分子量が10万~60万であり、残タンパク質量が2000ppm以下であり、粒子表面が非多孔質であり、かつ、以下の式(1)で示される重量平均分子量保持率が50%以上である、ポリ(3-ヒドロキシアルカノエート)粉体。
重量平均分子量保持率(%)=(160℃で20分間加熱した後のP3HAの重量平均分子量/160℃で20分間加熱する前のP3HAの重量平均分子量)×100 ・・・(1) - 重量平均分子量(Mw)と数平均分子量(Mn)との比(Mw/Mn)が、1.0~3.1である、請求項8に記載のポリ(3-ヒドロキシアルカノエート)粉体。
