しかしながら、本発明者らの知見によれば、DHAを構成脂肪酸として高含有するグリセリン脂肪酸エステルを高濃度で含有する自己乳化型の油脂含有組成物を得ようとすると、油脂含有組成物中に含有される油性成分が離液してしまい安定性が損なわれ、また、自己乳化性にも劣ることが判明した。
本発明は、上記の状況に鑑みたものであり、ドコサヘキサエン酸を構成脂肪酸として高含有するグリセリン脂肪酸エステルを高濃度で含有させた場合であっても、安定性が良好であり、且つ自己乳化性に優れた油脂含有組成物、及び該油脂含有組成物を用いた経口製剤を提供することを目的とする。
[1] 少なくとも、(A)油性成分、(B)グリセリン、及び(C)乳化剤を含有する油脂含有組成物であって、
前記(A)油性成分として構成脂肪酸の30質量%以上75質量%以下がドコサヘキサエン酸であるグリセリン脂肪酸エステルを含み、
前記(C)乳化剤として構成脂肪酸がオレイン酸であるデカグリセリン脂肪酸エステルを含み、且つ、
前記(A)油性成分、(B)グリセリン、及び(C)乳化剤の含有比率が、前記(A)油性成分、(B)グリセリン、及び(C)乳化剤の合計質量に対して以下の範囲内にある油脂含有組成物。
(A)油性成分:55質量%以上78質量%以下
(B)グリセリン:10質量%以上35質量%以下
(C)乳化剤:5質量%以上13質量%以下
[2] 前記構成脂肪酸の30質量%以上75質量%以下がドコサヘキサエン酸であるグリセリン脂肪酸エステルは、グリセリン骨格が有するヒドロキシ基の平均脂肪酸エステル化数が2.5以上3.0以下のグリセリン脂肪酸エステルである前記[1]に記載の組成物。
[3] 前記[1]又は[2]に記載の組成物を含有する経口製剤。
(A)油性成分:55質量%以上78質量%以下
(B)グリセリン:10質量%以上35質量%以下
(C)乳化剤:5質量%以上13質量%以下
ここで、「油脂含有組成物が自己乳化能を有する」とは、油脂含有組成物が、水、消化液等の水性液体に接触した際において、機械的操作により加えられる外力を必要することなく自然に乳化することを意味する。
本明細書において「~」を用いて示された数値範囲は、「~」の前後に記載される数値をそれぞれ最小値及び最大値として含む範囲を示す。
本明細書において「工程」との語は、独立した工程だけでなく、他の工程と明確に区別できない場合であっても本工程の所期の作用が達成されれば、本用語に含まれる。
以下、本発明の組成物が含有する必須成分及び任意成分について説明する。
本発明の油脂含有組成物は、(A)油性成分〔成分(A)〕を含有し、該油性成分として、構成脂肪酸の30質量%以上75質量%以下がドコサヘキサエン酸であるグリセリン脂肪酸エステル(以下、適宜「特定グリセリン脂肪酸エステル」と称する。)を含む。
なお、後述する構成脂肪酸がオレイン酸であるデカグリセリン脂肪酸エステル等の(C)乳化剤に包まれる油性成分は、成分(A)である油性成分には含まれない。
<(a)試料のけん化、及び脂肪酸成分の抽出>
測定対象となる試料を、1mol/L水酸化カリウム-エタノール(1質量%ピロガロール含有)溶液に溶解し、けん化する。なお、内部標準としてヘプタデカン酸を用いる。
けん化後の試料溶液に、水、30質量%硫酸、ジエチルエーテルを添加した後、ジエチルエーテル層(有機層)を分液し、水洗する。水洗後のジエチルエーテル層から溶媒(ジエチルエーテル)を留去し、脂肪酸成分を得る。
<(b)けん化>
上記(a)にて得られた脂肪酸成分に、0.5mol/L水酸化ナトリウムのメタノール溶液を添加し、けん化する。
<(c)メチルエステル化>
上記(b)にてけん化後の試料に、三フッ化ホウ素メタノール錯体メタノール溶液を添加して、試料中に含まれる脂肪酸成分をメチルエステル化する。
<(d)ガスクロマトグラフ法(水素炎イオン検出器)による脂肪酸の同定>
上記(c)にてメチルエステル化後の試料に、ヘキサン、飽和食塩水を添加した後、ヘキサン層(有機層)を分液する。分液後のヘキサン層を測定試料として、水素炎イオン検出器を用いたガスクロマトグラフ法により、試料中に含まれる脂肪酸の種類及び量を分析する。
平均脂肪酸エステル化数は、平均エステル化率(%)×3÷100で算出できる。
平均エステル化率(%)は、「基準油脂物性試験法」(日本油化学協会制定)により測定した、グリセリン脂肪酸エステルのケン化価(SV)、酸価(AV)、水酸基価(OHV)を用い、次式より算出する。
エステル化率(%)={(SV-AV)×100}/(OHV+SV-AV)
SV:ケン化価、AV:酸価、OHV:水酸基価を表す。
成分(A)の含有比率は、併用される他の成分の含有量を考慮して、上記の範囲内において適宜設定することができる。
本発明の油脂含有組成物は、(B)グリセリン〔成分(B)〕を、該組成物中の成分(A)、(B)及び(C)の合計質量に対して、10質量%以上35質量%以下含有する。
グリセリンとしては、濃度98%以上の濃グリセリンが特に好ましい。
本発明の油脂含有組成物は、(C)乳化剤〔成分(C)〕を含有し、該乳化剤として、構成脂肪酸がオレイン酸であるデカグリセリン脂肪酸エステル(以下、適宜「特定乳化剤」と称する。)を含む。
成分(C)として含まれる特定乳化剤は、本発明の油脂含有組成物が、水、消化液等の水性液体に接触して自己乳化能を発揮する際において、乳化剤として機能しうる。
疎水性基として脂肪鎖を有する乳化剤としては、一般的には、脂肪鎖が飽和脂肪酸である乳化剤の方が、脂肪鎖が不飽和脂肪酸である乳化剤に比べてHLBが低いことから、油脂への溶解性が高いことが知られている(吉田時行他 編「新版 界面活性剤ハンドブック」、工学図書株式会社、1987年、等の成書を参照。)。
しかしながら、本発明者らの知見によれば、DHAを構成脂肪酸として高含有する油脂を高濃度で含有させた自己乳化型の油脂含有組成物においては、一般に有効とされる脂肪鎖が飽和脂肪酸である乳化剤を用いた場合では、組成物の安定性や再分散性(自己乳化性)が発揮されず、脂肪鎖中に不飽和結合を有するオレイン酸を構成脂肪酸とするデカグリセリン脂肪酸エステルとグリセリンとを組み合わせた一定の領域においてはじめて、DHAを構成脂肪酸として高含有する油脂を高配合しても、安定性及び再分散性の双方に優れた自己乳化型の油脂含有組成物を得ることが可能となることが判明した。
また、本発明者らは、疎水性基である脂肪鎖としてオレイン酸を有する乳化剤であっても、親水基がショ糖であるショ糖脂肪酸エステルやポリソルベートでは、DHAを構成脂肪酸として高含有する油脂を高濃度で含有させた自己乳化型の油脂含有組成物の安定性及び再分散性は発揮されず、オレイン酸を構成脂肪酸とするデカグリセリン脂肪酸エステル(特定乳化剤)のみが選択的に乳化性能において優れた効果を発揮することを見出したものである。
本発明の油脂含有組成物は、成分(A)、(B)及び(C)の各成分と共に、必要に応じて他の成分を含有してもよい。他の成分としては、例えば、目的に応じた物性や機能性を有する他の油性成分(例えば、アスタキサンチン、ルテイン、リコピン等のカロテノイド色素を含有する油脂類、リン脂質類等)、香料、酸化防止剤、植物や果実等の抽出物などが挙げられる。
油類に関しては、成分(A)と混合して配合することができる。この場合、当該油類の含有量は、成分(A)の含有量に包含される。
本発明の油脂含有組成物、例えば、(B)成分と(C)成分とを混合し、湯せん中で均一に溶解して、混合液を得た後、該混合液に、(A)成分を十分撹拌しながら徐々に添加していき、全量添加する方法により調製することができる。
例えば、本発明の油脂含有組成物は、特定乳化剤を含む成分(C)を、成分(B)であるグリセリンに溶解したものを水相とし、成分(A)である油性成分を油相として、該油相を該水相にわずかずつ添加して乳化する方法よって好適に調製することができる。水相には、水を更に添加してもよい。
更に、カプセル製剤としては、ソフトカプセル製剤及びハードカプセル製剤のいずれであってもよいが、内容物の漏れや摂取容易性の点からは、ソフトカプセル製剤であることがより好ましい。
ソフトカプセル皮膜としては、ゼラチン等の動物原料に由来する成分、又は、デンプン、カラギーナン等の植物原料に由来する成分を主成分とするものであることが好ましい。
ソフトカプセル製剤の製造において、製造適性の点から、本発明の油脂含有組成物を封入(充填)させる際には、該油脂含有組成物の温度を上げて粘度を下げること、あるいは、水などを添加して粘度を下げることが望ましい。ソフトカプセル完成後における油脂含有組成物の経時安定性の観点からは、水を添加して粘度を下げることが好ましい。加える水の量は油脂含有組成物の粘度によって異なるが、油脂含有組成物の単位質量あたり3~8質量%程度が好ましい。
<油脂含有組成物の調製>
実施例1~14、比較例1~15の各油脂含有組成物を、以下のようにして調製した。
また、実施例14については、表1に示す種類及び量で、(B)成分、(C)成分及び水を混合して混合液を得た以外は、実施例1と同様にして油脂含有組成物を得た。
実施例1~14、比較例1~15の油脂含有組成物を用いて、初期安定性、及び自己乳化性についての評価を行った。
各油脂含有組成物を、蓋付ガラス容器中に、25℃で1日放置後、組成物の状態を目視にて観察し、下記の評価基準により評価した。結果を表1に併記する。
<評価基準>
A:分離はもちろんなく均一であり安定性が非常に優れている
B:油性成分が、組成物中から分離することなく、安定している
C:油性成分が分離して、組成物の表面に染み出ている
D:油性成分が、組成物中から完全に分離している
ここで、油性成分は、(A)成分等の油性成分である。
初期安定性の評価結果が「A」又は「B」であったものに関して、100mgの油脂含有組成物に25℃の水50mlを添加し、マグネティックスターラーで撹拌し、分散させることにより、水に接触させた際の自己乳化性を目視により観察し、以下の評価基準により評価した。結果を表1に併記する。
A:微細に自己乳化した
B:油滴の合一なく、自己乳化した
C:自己乳化はするが油滴の合一が見られる、もしくは、濁りが大きい(分散径が大きい)
D:全く自己乳化しない
・油脂A:構成脂肪酸の70質量%以上がドコサヘキサエン酸、平均脂肪酸エステル化数:約2.7以上である油脂含有原料(商品名:アルガトリウム(ブルディーテクノロジー社製))(特定グリセリン脂肪酸エステル)
・油脂B:含有脂肪酸の46質量%がドコサヘキサエン酸、平均脂肪酸エステル化数:約2.8以上である油脂含有原料(商品名:DHA-46A、(株)マルハニチロ食品製)(特定グリセリン脂肪酸エステル)
・油脂C:含有脂肪酸の22質量%がドコサヘキサエン酸、平均脂肪酸エステル化数:約2.8以上である油脂含有原料(商品名:DHA-22、(株)マルハニチロ食品製)
・油脂D:脂肪酸がカプリル酸(飽和脂肪酸C8:ドコサヘキサエン酸非含有)のトリグリセリドである油脂含有原料、平均脂肪酸エステル化数:約2.9以上(商品名:ココナードRK、花王(株)製)
・グリセリン(食添用グリセリン、花王(株)製)(成分(B))
・乳化剤A:脂肪鎖がステアリン酸(C18飽和脂肪酸)のデカグリセリン脂肪酸エステル(商品名:サンソフトQ-18S、太陽化学(株)製)
・乳化剤B:脂肪鎖がオレイン酸(C18不飽和脂肪酸)のデカグリセリン脂肪酸エステル(商品名:ポエムJ-0381V、理研ビタミン(株)製)(特定乳化剤)
・乳化剤C:脂肪鎖ラウリン酸(C12飽和脂肪酸)のデカグリセリン脂肪酸エステル(商品名:サンソフトQ-12S、太陽化学(株)製)
・乳化剤D:ポリソルベート80(商品名:ウィルサーフTF-80、日油(株))
・乳化剤E:脂肪鎖がオレイン酸のショ糖脂肪酸エステル(商品名:Q-1570Y、三菱化学フーズ(株)製)

Claims (3)
- 少なくとも、(A)油性成分、(B)グリセリン、及び(C)乳化剤を含有する油脂含有組成物であって、
前記(A)油性成分として構成脂肪酸の30質量%以上75質量%以下がドコサヘキサエン酸であるグリセリン脂肪酸エステルを含み、
前記(C)乳化剤として構成脂肪酸がオレイン酸であるデカグリセリン脂肪酸エステルを含み、且つ、
前記(A)油性成分、(B)グリセリン、及び(C)乳化剤の含有比率が、前記(A)油性成分、(B)グリセリン、及び(C)乳化剤の合計質量に対して以下の範囲内にある油脂含有組成物。
(A)油性成分:55質量%以上78質量%以下
(B)グリセリン:10質量%以上35質量%以下
(C)乳化剤:5質量%以上13質量%以下 - 前記構成脂肪酸の30質量%以上75質量%以下がドコサヘキサエン酸であるグリセリン脂肪酸エステルは、グリセリン骨格が有するヒドロキシ基の平均脂肪酸エステル化数が2.5以上3.0以下のグリセリン脂肪酸エステルである請求項1に記載の油脂含有組成物。
- 請求項1又は請求項2に記載の油脂含有組成物を含有する経口製剤。
