Abstract

L'invention concerne des transglutaminases utilisées entre autres pour la réticulation covalente de protéines. On connaît une transglutaminase microbienne, obtenue à partir de Streptomyces mobaraensis, qui présente un faible niveau de stabilité à des températures plus élevées. La transglutaminase commercialisée sous l'appellation d'Activa WM est utilisée principalement dans la technologie alimentaire. Pour des applications ne relevant pas du secteur alimentaire, il faut utiliser des transglutaminases plus thermostables et plus actives pour des réactions de réticulation effectuées à des températures plus élevées. L'invention vise de ce fait à produire des transglutaminases présentant une meilleure thermostabilité. Des transglutaminases modifiées présentant une meilleure stabilité ont pu être produites, par mutagenèse aléatoire, par amplification en chaîne par polymérase (PCR) sujette à erreurs. Les séquences géniques de ces transglutaminases ont été analysées et la séquence d'aminoacide modifiée correspondante a été dérivée. Les régions présentant une meilleure thermostabilité comparativement aux enzymes connues ont pu être identifiées dans la protéine, au moyen de cette information de séquence et des structures cristallines connues. L'invention s'applique à la technologie alimentaire, au secteur pharmaceutique et cosmétique, ainsi qu'à la synthèse polymère.
