Abstract

 腸の上皮細胞におけるGsdmdおよびそれをコードするGsdmd遺伝子の機能を解明するための、モデルマウスを提供する。染色体上でGsdmdをコードするGsdmd遺伝子がノックアウトされたホモ接合体を有し、Gsdmdが非発現であるマウスを、モデルマウスとする。まず、マウスゲノムDNAにおけるGsdmd遺伝子のエキソン2を含む断片を有するターゲティングベクターをマウス胚幹細胞に導入し、相同組換えによりGsdmd遺伝子がノックアウトされた胚幹細胞からキメラ胚細胞を作製する。これを仮親マウスに移植して、生まれたGsdmd遺伝子がノックアウトされたキメラマウスを交配して、ヘテロ接合体マウスを選択し、前記へテロ接合体マウス同士を交配することによって、Gsdmd遺伝子がノックアウトされたホモ接合体のモデルマウスが得られる。
