Abstract

L'invention concerne la détermination de séquences nucléotidiques par séquençage. La synthèse amorcée d'un deuxième brin complémentaire d'un brin complémentaire dans des ensembles répétés d'étapes, chaque étape consistant à fournir une ou plusieurs des classes de complémentarité nucléotidiques à incorporer dans le brin synthétisé, et chaque ensemble d'étapes consistant é fournir les quatre classes complémentaires nucléotidiques possibles. Trois des quatre classes complémentaires nucléotidiques peuvent d'abord être fournies pour être incorporées dans le brin synthétisé, puis séparément la quatrième classe de complémentarité nucléotidique séparément. L'invention porte également sur une molécule d'ADN formée d'une partie tige et de première et seconde parties boucles, la partie tige comprenant un premier et un second brin, les premier et second brins étant de longueur équivalente, complémentaires et recuits ensemble, la première boucle reliant l'extrémité 3' du premier brin à l'extrémité 5' du second brin et la seconde boucle reliant l'extrémité 3' du second brin à l'extrémité 5' du premier brin, de sorte que la molécule d'ADN ne possède plus d'extrémité 5' ou 3' libre, et sur son utilisation.
