Abstract

L'invention concerne un gène de nitrilase identifié par extraction et purification de nitrilase d'Arthrobacter sp.NSSC104, par détermination des séquences d'acides aminés N-terminal et internes, par désignation d'une amorce sens et d'une amorce anti-sens, par PCR d'ADN chromosomal à l'aide de ces amorces, par marquage de l'ADN ainsi amplifié avec de la digoxygénine pour obtenir une sonde, et par criblage d'un transformant contenant l'ADN de recombinaison par hybridation sur colonies d'Escherichia coli avec un vecteur plasmidique transféré à l'intérieur, porteur d'un fragment d'ADN contenant un gène de nitrilase obtenu par digestion d'ADN chromosomal avec des enzymes de restriction et intégré, à l'aide de la sonde mentionnée ci-dessus.
