Abstract

La présente invention concerne un procédé et un système optiques permettant de détecter un analyte par une immobilisation en phase solide, des marqueurs d'analyte spécifiques aptes à produire un signal et des procédés d'utilisation de ces derniers. Le procédé de détection par numération de l'invention permet de rétrécir la zone d'observation du signal et, par conséquent, d'améliorer le rapport signal détectable/fond. Le système comprend une plate-forme/support permettant d'immobiliser une platine d'échantillon sur laquelle est lié un échantillon marqué (complexe d'analyte), une source de rayonnement, un appareil optique destiné à générer et à diriger le rayonnement vers l'échantillon et un moyen de collecte et d'analyse des données. Au moment où le système est enclenché, l'échantillon génère un signal pouvant se différencier du signal de fond, les deux signaux étant collectés et mis en image grâce à un détecteur de signal qui transmet une image de l'échantillon à un appareil de traitement des données. Ledit appareil reçoit les mesures du signal et, à son tour, effectue la numération des événements de liaison individuels. Il est possible de renforcer le signal généré par une augmentation choisie de la masse. Le procédé d'analyse par numération de l'invention, qui permet de détecter des événements de liaison individuels, peut être utilisé, par exemple, dans des analyses visant à détecter un analyte ou à confirmer des résultats dans des applications de recherche, des applications commerciales ou aux endroits où des soins sont dispensés.
