Abstract

Die Erfindung bezieht sich auf Varianten des rekombinanten humanen Interferon-η (rhu-IFN-η) mit erhöhter thermischer Stabilität. Ausgehend von rhu-IFN-η, wurden in der Aminosäuresequenz des Polypeptidmonomers paarweise mindestens eines der folgenden Aminosäurenpaare zu Cystein verändert: Glu8 und Ser70, Ala18 und His112, Lys81 und Leu121 beziehungsweise Gln49 und Leu96. Zuzätzlich kann das C-terminale Ende des Polypeptids um 1 bis 10 Aminosäuren verkürzt werden. Die neuartigen, erfindungsgemäßen Proteine sind ohne Verlust der biologischen Aktivität thermisch stabiler als das unveränderte rhu-IFN-η. Die Erfindung betrifft weiterhin die Herstellung der zur Erzeugung der erfindungsgemäßen Proteine benötigten Oligonucleotide, Vektoren und Mikroorganismen. Weiterhin wird ein Verfahren zur Herstellung und Verwendungen der Proteine vorgestellt.
