Abstract

L'invention se rapporte à un réactif unique stable et à des procédés automatisables utilisant ce réactif pour obtenir le titrage du calcium dans le sérum et dans des échantillons aqueux, tel que l'urine, les échantillons aqueux ne contenant que peu ou pas de protéine. Le réactif renferme de l'arsénazo III, de la 8-hydroxyquinoline, un tampon qui n'entre pas en concurrence avec l'arsénazo III pour se lier au calcium et qui confère au réactif un pH compris entre 8,5 environ et 10 environ, le tampon étant en quantité suffisante pour maintenir le pH du réactif à l'intérieur d'une plage de +/- 0,1 unités de pH, lorsque le réactif est ajouté à un échantillon de teneur en calcium inconnue, le réactif renfermant en outre du sulfoxyde diméthylique et de l'eau. Le sulfoxyde diméthylique a pour fonction importante de permettre l'incorporation de la 8-hydroxyquinoline dans un réactif unique stable, qui se trouve à un pH alcalin optimal pour la mesure du calcium par le complexe calcium-arsénazo III. La 8-hydroxyquinoline élimine les interférences de titre dues au ion magnésium. On peut également ajouter au réactif au moins un agent tensio-actif non ionique en quantité suffisante pour réduire au minimum la turbidité des lipides. Les mesures spectrales du complexe calcium-arséanzo III sont effectuées à des longueurs d'ondes qui réduisent au minimum les interférences provenant de la lipémie, de l'hémolyse et de la bilirubinémie. Les interférences spectrales provenant de la lipémie et de l'hémolyse sont en outre réduites grâce à l'utilisation de mesures bichromatiques du complexe calcium-arsénazo III et grâce à l'utilisation de mesures à blanc du sérum.
