Abstract
El receptor CTLA4 como un ligando para el antígeno B7. Se proporciona la secuencia completa de aminoácido que codifica el gen del receptor CTLA4 humano. Se proporcionan métodos para expresar la CTLA4 como una proteína de fusión de inmunoglobulina, para preparar proteínas de fusión CTLA4 híbridas y para utilizar las proteínas de fusión solubles, fragmentos y derivados de las mismas, incluyendo anticuerpos monoclonales que reaccionan con B7 y CTLA4, para regular las interacciones con las células T y las respuestas inmunitarias mediadas por dichas interacciones. Se reivindica: 1) Una proteína de fusión CTLA4Ig que se enlaza al antígeno B7, teniendo dicha proteína una primera secuencia de aminoácidos que consiste del dominio extracelular de CTLA4 y una segunda secuencia de aminoácidos que consiste del eje, las regiones CH2 y CH3 de la molécula de una inmunoglobulina humana. 2) Una proteína de fusión CTLA4Ig que se enlaza al antígeno B7, teniendo dicha proteína (a) una primera secuencia de aminoácido que comienza con alanina en la posición del aminoácido 1 y que finaliza con el ácido aspártico en la posición de aminoácido 125 de la SEQ ID NO: 13 que representa el dominio extracelular de CTLA4 y (b) una segunda secuencia de aminoácido que consiste del eje, y de las regiones CH2 y CH3 de una molécula de inmunoglobulina humana.The CTLA4 receptor as a ligand for the B7 antigen. The complete amino acid sequence encoding the human CTLA4 receptor gene is provided. Methods are provided for expressing CTLA4 as an immunoglobulin fusion protein, for preparing hybrid CTLA4 fusion proteins, and for using soluble fusion proteins, fragments and derivatives thereof, including monoclonal antibodies reacting with B7 and CTLA4, to regulate T-cell interactions and immune responses mediated by such interactions. The following are claimed: 1) A CTLA4Ig fusion protein that binds to the B7 antigen, said protein having a first amino acid sequence consisting of the extracellular domain of CTLA4 and a second amino acid sequence consisting of the axis, the CH2 and CH3 regions of the molecule of a human immunoglobulin. 2) A CTLA4Ig fusion protein that binds to the B7 antigen, said protein (a) having a first amino acid sequence that begins with alanine at amino acid position 1 and ends with aspartic acid at amino acid position 125 of the SEQ ID NO: 13 representing the extracellular domain of CTLA4 and (b) a second amino acid sequence consisting of the axis, and the CH2 and CH3 regions of a human immunoglobulin molecule.
