Abstract
The invention relates to virology and biotechnology and can be used in virological investigations for the purpose of isolation of leaf-rolling mosaic virus from plant material into pure culture, long-term storage of virus isolates, investigation of physico-chemical, biological, serological properties and reproduction features of leaf-rolling mosaic virus. A method for cultivation of leaf-rolling mosaic virus in subinoculated lines of mammalian cell cultures provides preparation of partially clarified virus preparation by homogenization of potato plants in phosphate buffer solution, emulsification with chloroform, and centrifugation. The preparation decontamination is carried out with antibiotics. The produced suspension is introduced into vials with cell culture CHEB or VNK-1. After virus contact with monolayer of mammalian cells the latter is washed out and introduced into cell maintenance medium. Inoculated vials with cell culture are incubated in thermostat till appearance of features of virus cytopathic action. The proposed invention allows isolating virus into pure culture during 1-5 days.
