Abstract
本發明是有關於一種化妝品組合物,其包括作為有效成分的
曲酸亞甲基二氧肉桂酸酯以及作為穩定化劑的糖脂中的甘露糖赤藻糖醇脂,可以避免具有抗老化、美白、抗氧化等各種功效的曲酸亞甲基二氧肉桂酸酯在劑型中結晶化。
本發明的化妝品組合物即便以高濃度含有有效成分,亦不會於各種化妝品劑型中引起結晶析出。因此,本發明的化妝品組合物能夠將其有效成分的功能性極大化,而且提高皮膚傳達力且改善使用感。
Description
本發明是有關於一種為了提高包含用作化妝品有效成分的曲酸亞甲基二氧肉桂酸酯的組合物的劑型穩定性而使用甘露糖赤藻糖醇脂作為穩定化劑的化妝品組合物。
化妝品組合物為了發揮特定功效而包含各種有效成分。特別是,皮膚直接暴露於外部環境,因此易於因外部刺激而受損。
對此,已知有用以防止皮膚受損且賦予各種功效的多種物質,曲酸亞甲基二氧肉桂酸酯(kojyl methylenedioxycinnamate,商品名為Seletinoid G,Seletinoid GTM)作為此種物質中的一種,特別是於抗老化、抗氧化、美白等方面具有優異效果。
然而,上述曲酸亞甲基二氧肉桂酸酯的結晶性較強而難以於組合物內溶解,於溶解後亦會於較快的時間內再次結晶化或析出而降低製品的皮膚吸收率或使用感。因此種問題而即便曲酸亞甲基二氧肉桂酸酯具有優異的功效,亦難以實質上用作化妝品組合物的有效成分。
為了改善此種曲酸亞甲基二氧肉桂酸酯的難溶性問題,先前已進行多種嘗試。
例如,有於欲溶解結晶性物質的情形時主要使用的方法即利用包含高分子的微膠囊的方法、連同曲酸亞甲基二氧肉桂酸酯一併將結晶性較強的一種以上的另一物質溶解至極性油而提高溶解度的嘗試(韓國公開專利10-2015-0062895)。
然而,於利用包含高分子的微膠囊的情形時,具有如下缺點:有效成分於膠囊內部的溶解度由高分子間的相溶性或高分子本身的結晶度決定,於大部分情形時,在高分子膠囊內獨立地形成結晶而有效成分的釋出速度變得非常緩慢,因此其提供效率下降。
另外,於連同曲酸亞甲基二氧肉桂酸酯一併將結晶性較強的一種以上的另一物質溶解至極性油而提高溶解度的情形時,較為繁瑣、不經濟且無明顯的溶解度改善效果,從而於商用化方面存在極限。
因此,實情為需開發一種降低曲酸亞甲基二氧肉桂酸酯的結晶性且保持組合物內的穩定性的更有效的方法。 [先前技術文獻]
韓國公開專利公報第2015-0062895號“包含兩種具有苯環的化合物的化妝品組合物”。
[發明欲解決的課題]
為了解決上述問題,本申請人選定多種穩定化劑並對上述多種穩定化劑進行篩選而選定可抑制發生曲酸亞甲基二氧肉桂酸酯的結晶化的最佳穩定化劑,於劑型化後,對是否產生結晶進行確認,結果確認到長時間未發生結晶析出。
因此,本發明的目的在於提供一種包含曲酸亞甲基二氧肉桂酸酯及穩定化劑的化妝品組合物。 [解決課題的手段]
為了解決上述問題,本發明提供一種化妝品組合物,其特徵在於包含: 曲酸亞甲基二氧肉桂酸酯(Kojyl methylenedioxycinnamate),其作為有效成分而以下述化學式1表示;及 甘露糖赤藻糖醇脂(Mannosylerythritol lipids,MELs),其作為穩定化劑而以下述化學式2表示: [化學式1][化學式2](於上述化學式2中,R1

此時,特徵在於:上述曲酸亞甲基二氧肉桂酸酯相對於化妝品組合物的總重量包含0.05重量%至0.25重量%。
另外,特徵在於:上述甘露糖赤藻糖醇脂相對於化妝品組合物的總重量包含0.25重量%至5.0重量%。
特別是,特徵在於:以1:2.5以上至1:100的重量比包含上述曲酸亞甲基二氧肉桂酸酯與甘露糖赤藻糖醇脂。 [發明效果]
本發明的化妝品組合物提高包含曲酸亞甲基二氧肉桂酸酯的劑型的穩定性,解決先前於劑型內析出結晶的問題而不會發生因上述結晶析出產生的異物感引起的使用感下降,從而可提高製品品質。
因以此方式提高的劑型穩定性而能夠以高濃度於化妝品組合物內含有具有抗老化、抗氧化、美白效果的曲酸亞甲基二氧肉桂酸酯,從而可將上述曲酸亞甲基二氧肉桂酸酯的功能極大化而美白皮膚及增加皮膚彈力且抑制皺紋的產生,藉此防止皮膚老化。
於本發明中,提出一種可於源頭上阻斷於包含作為有效成分的曲酸亞甲基二氧肉桂酸酯的化妝品組合物內發生結晶化及結晶析出的新穎的化妝品組合物。
上述化學式1的曲酸亞甲基二氧肉桂酸酯以國際純化學暨應用化學聯合會(International Union of Pure and Applied Chemistry,IUPAC)命名法命名為2-((3E)-4(2H,3H-苯并[3,4-d]-1,3-二氧戊環-5-基)-2-氧代-3-丁烯氧基)-5-羥基-4H-吡喃-4-酮)(2-((3E)-4(2H,3H-benzo[3,4-d]-1,3-dioxolan-5-yl)-2-oxo-but-3-enyloxy)-5-hydroxy-4H-pyran-4-one),於市場中以商品名Seletinoid G(Seletinoid GTM
)售賣。
上述曲酸亞甲基二氧肉桂酸酯具有抗老化、美白、抗氧化等多種皮膚功效,以各種劑型用作化妝品組合物的有效成分。然而,此種化合物具有結晶性較強而於組合物內的溶解較難,於溶解後亦會隨時間的增加而發生結晶化及析出的問題,從而引起劑型穩定化大幅下降且使用感降低的問題。
因此,於本發明中,使用於糖鍵結有脂肪酸的糖脂的一種即甘露糖赤藻糖醇脂(manosyl erythritol lipids,MELs,以下稱為“MEL”)作為用以防止上述化合物的結晶化的穩定化劑。

上述MEL為具有界面活性的生物界面活性劑(biosurfactants)中的一種。
生物界面活性劑可自微生物製備,生物分解性較高且為低毒性,對人體皮膚無害且具有優異的保濕能力而具有保護皮膚的效果。另外,上述生物界面活性劑為如下物質:最小表面張力為29 dyne/cm,臨界微胞濃度(CMC)為15 μM(10 mg/l)而表現出遵循現有化學合成界面活性劑的程度的表面活性。
於本發明中,上述MEL的製備並無特別限定,可藉由公知的方法生產。作為一例,可自相當於念珠菌屬(Candida sp.)、球擬酵母菌(Torulopsis sp.)、假單胞菌屬(Pseudomonas sp.)、芽孢桿菌屬(Bacillus sp.)、產鹼桿菌屬(Alcaligenes sp.)、不動桿菌屬(Acinetobacter sp.)、黑穗病菌屬(Ustilago sp.)、赤球菌屬(Rhodococcus sp.)中任一種以上的微生物獲得。
於本發明中,連同化學式1的曲酸亞甲基二氧肉桂酸酯一併使用化學式2的MEL作為穩定化劑,藉此防止因上述曲酸亞甲基二氧肉桂酸酯的較低的劑型穩定性而發生結晶化及結晶析出。此種劑型穩定性於低溫、室溫及高溫下亦顯示相同的結果。
為了劑型穩定性,可使用先前已知的穩定化劑、界面活性劑、乳化劑、分散劑等各種候補族群的物質,但需考慮有效成分的化學結構,此時無法於所有穩定化劑中確保相同的效果。根據本發明的較佳的實驗例1,連同曲酸亞甲基二氧肉桂酸酯分別一併使用MEL、乙醇、聚山梨糖醇酯20、琥珀酸二乙氧基乙酯、及甘油/聚甘油-6異硬脂酸/山崳酸酯而製備劑型,對該劑型的結晶化傾向進行確認,結果確認到隨時間經過而不僅只於包含MEL的組合物中發生結晶化,而且於其他組成的使用中發生嚴重的結晶化。
另外,根據實驗例2,對溫度的結晶化傾向進行確認,結果於添加MEL的情形時,在低溫、室溫及高溫下均表現出優異的劑型穩定性,於未添加MEL的情形時,發生嚴重的結晶化,該結晶化於低溫下表現出進一步加速化的傾向。
若因結晶化而於劑型內形成結晶,則有效成分難以傳達至皮膚而不僅功效減小,而且於使用時有異物感而成為品質下降的要素,因此防止結晶化非常重要。最終,於本發明中,藉由使用MEL而解決了此種問題。
進而,藉由MEL而提高劑型穩定性,藉此具有能夠以高至0.25重量%的高濃度含有作為有效成分的曲酸亞甲基二氧肉桂酸酯的優點。
另一方面,結晶化是指劑型的穩定性,為此有效成分的含量、MEL的含量、有效成分與MEL的含量比可成為與上述穩定性相關的重要參數。
較佳為,本發明的化妝品組合物於總重量內使用0.05重量%至0.25重量%的曲酸亞甲基二氧肉桂酸酯,較佳為使用0.05重量%至0.15重量%。若上述曲酸亞甲基二氧肉桂酸酯的含量未滿上述範圍,則無法確保藉由使用上述曲酸亞甲基二氧肉桂酸酯而獲得的有效效果、即抗老化、美白、抗氧化效果。與此相反,於超過上述範圍的情形時,有穩定性下降而發生結晶化之虞。
另外,於化妝品組合物的總重量內使用0.25重量%至5.0重量%的MEL,較佳為使用0.25重量%至2.0重量%。若上述MEL的含量未滿上述範圍,則無法實現上述曲酸亞甲基二氧肉桂酸酯的穩定化而會發生結晶化,與此相反,於超過上述範圍的情形時,效果上無較大差異而為非經濟性,因此於上述範圍內適當地使用。
特別是,作為於防止結晶化方面較為重要的參數之一的曲酸亞甲基二氧肉桂酸酯與MEL的重量比大於1:2.5即可,較佳為以1:2.5以上至1:100的重量比使用,更佳為以1:3至1:20的重量比使用。根據本發明的較佳的實驗例2,對是否發生因含量比引起的結晶化進行確認,結果確認到曲酸亞甲基二氧肉桂酸酯與MEL於1:2.5的重量比下產生結晶,於上述1:2.5以上的範圍內不會發生結晶化。
以此方式包含曲酸亞甲基二氧肉桂酸酯與MEL的化妝品組合物於應用至皮膚時,具有抗老化、美白、抗氧化效果而美白皮膚及增加皮膚彈力且抑制皺紋的產生,藉此可防止皮膚老化。
本發明的化妝品組合物可含有化妝品學或皮膚科學上可容許的介質或基劑而劑型化。本發明的化妝品組合物中所包含的成分除作為有效成分的上述組成以外,包含普遍利用於化妝品組合物中的成分,例如包含油脂成分、保濕劑、軟化劑、界面活性劑、有機及無機顏料、有機粉體、紫外線吸收劑、防腐劑、去泡劑、增稠劑、殺菌劑、抗氧化劑、植物萃取物、pH值調整劑、酒精、色素、香料、血行促進劑、冷感劑、止汗劑等。
本發明的化妝品組合物可製備成本領域中通常製備的任一劑型,例如可劑型化成溶液、懸浮液、乳濁液、糊劑、凝膠、乳霜、潤膚乳、粉劑、香皂、含有界面活性劑的洗面奶、油、粉末粉底、乳濁液粉底、蠟狀粉底及噴霧等,但並不限定於此。更詳細而言,可製備成柔軟化妝水、營養化妝水、營養乳霜、按摩乳霜、精華素、眼霜、洗面膏、泡沫型洗面奶、洗面水、面膜、噴霧或粉的劑型。
於本發明的劑型為糊劑、乳霜或凝膠的情形時,可利用動物油、植物油、蠟、石蠟、澱粉、黃芪、纖維素衍生物、聚乙二醇、矽、膨潤土、二氧化矽、滑石或氧化鋅等作為載體成分。
於本發明的劑型為粉或噴霧的情形時,可利用乳糖、滑石、二氧化矽、氫氧化鋁、矽酸鈣或聚醯胺粉作為載體成分,特別是於噴霧的情形時,可追加包含如含氯氟烴、丙烷/丁烷或二甲醚的促進劑。
於本發明的劑型為溶液或乳濁液的情形時,可利用溶劑、溶解化劑或乳濁化劑作為載體成分,例如有水、乙醇、異丙醇、碳酸乙酯、乙酸乙酯、丙三醇、卡波姆、卡醇、苯甲酸苯甲酯、丙二醇、1,3-丁二醇、甘油脂肪族酯、辛酸癸酸甘油酯、氫化聚癸烯、鯨蠟硬脂基葡糖苷、山梨糖醇硬脂酸酯、聚乙二醇、鯨蠟硬脂醇等。
於本發明的劑型為懸浮液的情形時,可利用如水、乙醇或丙二醇的液狀稀釋劑、如乙氧基化異硬脂醇、聚山梨糖醇酯及聚氧乙烯脫水山梨糖醇酯的懸浮劑、微小結晶性纖維素、偏氫氧化鋁、膨潤土、瓊脂或黃芪等作為載體成分。
於本發明的劑型為含界面活性劑的洗面奶的情形時,可利用脂肪族醇硫酸酯、脂肪族醇醚硫酸酯、磺基琥珀酸單酯、羥乙基磺酸酯、咪唑啉衍生物、甲基牛磺酸酯、肌胺酸酯、脂肪酸醯胺醚硫酸酯、烷基醯胺甜菜鹼、脂肪族乙醇、脂肪酸甘油酯、脂肪酸二乙醇醯胺、植物油、羊毛酯衍生物或乙氧基化甘油脂肪酸酯等作為載體成分。
作為一例,本發明的化妝品組合物可為乳濁液、即經乳化的劑型。
乳液是化妝品產業中最具代表性的劑型,其指利用乳化技術均勻地分散油相與水相的類型。此種乳液可根據連續相的組成而製備成各種形態,可為油包水(W/O,water-in-oil)、水包油(O/W)、水包油包水(W/O/W)或油包水包油(O/W/O)型乳液,於本發明中並不特別限定其形態。
此時,水相部連同水一併使用多元醇,油相部使用油,可將如上所述的各種添加劑、穩定化劑、界面活性劑、紫外線吸收劑等添加至各相中。
水相部中所使用的水為蒸餾水,較佳為使用經去離子化的蒸餾水,且以整體重量%滿足100重量%的方式用作殘部。
水相部中所使用的多元醇是為了保濕而使用,折射率的範圍為1.3至1.5,使用選自由丙三醇、丙二醇、丁二醇、赤藻糖醇、木糖醇、麥芽糖醇、山梨糖醇、聚丙三醇、聚乙二醇、戊二醇、異戊二
醇及其等的組合所構成的族群中的一種。上述多元醇的含量可於組合物整體內使用0.1重量%~20.0重量%。
於本發明中,並不特別限定油相部中所使用的油,只要為於本技術領域內普遍使用的油,即可使用任一種油。代表性地,可使用包含聚癸烯及石蠟油的烴類油;包含鯨蠟醇乙基己酸酯、三辛酸甘油酯、肉豆蔻酸辛基十二烷酯、棕櫚酸異丙酯、肉豆蔻酸異丙酯、棕櫚酸乙基己酯的酯類合成油;包含環甲聚矽氧烷、二甲聚矽氧烷的矽油;動物油;包含芒果油、乳木果油、古樸阿蘇果油、澳洲堅果籽油的植物油;乙氧基化烷基醚類油;膽固醇;膽固醇硫酸酯;植物鞘氨醇;鞘脂質;選自由鯊肝醇、山崳醇、鯨蠟硬脂醇、鯨蠟醇、十八烷醇等C10~C40脂肪醇、辛酸/癸酸三甘油酯、神經醯胺及其等的混合物所構成的族群中的一種。
水相部及油相部中的任一種以上的相可更包含增稠劑。代表性地,可為選自由丙烯酸鈉共聚物、卡波姆、交聯或非交聯丙烯酸聚合物、特別是藉由多價作用基劑交聯而成的聚丙烯酸、例如由GOODRICH公司市售的商標名為“CARBOPOL”的製品、纖維素衍生物、例如甲基纖維素、羥基甲基纖維素、羥基乙基纖維素、羥基丙基甲基纖維素、羧基甲基纖維素、羧基甲基纖維素的鈉鹽、鯨蠟硬脂醇、含有氧化乙烷33莫耳的經氧乙烯化的鯨蠟硬脂醇的混合物、瓜爾豆膠、黃原膠及其等的組合所構成的族群中的一種。上述增稠劑的含量可於組合物整體內使用0.01重量%~10.0重量%。
水相部及油相部中的任一種以上的相可更包含界面活性劑。
例如,可為山梨糖醇硬脂酸酯、山梨糖醇月桂酸酯、山梨糖醇棕櫚酸酯、甘油硬脂酸酯、聚甘油硬脂酸酯、聚氧乙烯硬脂醇醚、聚氧乙烯油酸酯、聚氧乙烯山梨糖醇硬脂酸酯、聚氧乙烯山梨糖醇月桂酸酯、聚氧乙烯山梨糖醇油酸酯、聚氧乙烯山梨糖醇六油酸酯、聚氧乙烯氫化蓖麻油等天然油的聚氧乙烯醚類化合物、或泊洛沙姆168(Poloxamer 168)、泊洛沙姆407(Poloxamer 407)等聚氧丙烯的聚氧乙烯醚類化合物、硬脂酸鈉、硬脂酸鉀、月桂酸鈉、月桂酸硫酸鈉、月桂醇醚硫酸鈉(sodium laureth sulfate)、月桂醇醚硫酸銨、鯨蠟醇磷酸酯鉀、PEG-100二甲聚矽氧烷、聚乙二醇單異硬脂酸、橄欖油鯨蠟醇酯、橄欖油山梨糖醇酐、經氫化的卵磷脂(Hydrogenated Lecithin)、花生醇葡糖苷、鯨蠟硬脂醇、鯨蠟硬脂基葡糖苷、聚山梨糖醇酯(Polysorbate)80及聚山梨糖醇酯(Polysorbate)60等。上述界面活性劑相對於組合物的總重量使用10重量%以下。
除此之外,可視需要更添加本技術領域所公知的水溶性/油溶性生物活性成分、保濕劑、防腐劑、pH值調節劑、脂肪酸、乳化劑、抗氧化劑、紫外線遮斷劑、顏料、染料、香料、穩定化劑等添加劑,此時上述添加劑的種類及含量可由業者適當地選擇。
乳液劑型是於製備以水為連續相的水相部,製備以油為連續相的油相部後,變更上述水相部與油相部的混合順序而製備成各種形態。具體而言,水相部以加熱成50℃~90℃、較佳為60℃~80℃的方式製備,油相部以加熱成50℃~80℃、較佳為65℃~75℃以便其他成分可充分地溶解至油的方式製備。
此時,於真空乳化槽內,利用均質混合機以2000 rpm~4000 rpm、較佳為3000 rpm的速度執行3分鐘~10分鐘的攪拌。
本發明的曲酸亞甲基二氧肉桂酸酯與MEL可添加至上述水相部或油相部中的任一相中,或於製備最終乳液後添加,於本發明的一實施例中,在混合水相部與油相部的步驟後進行添加。
如上所述,以此方式製備的化妝品組合物將曲酸亞甲基二氧肉桂酸酯的有效功能極大化,且於劑型化後不會發生結晶化,藉此可確保提高配方穩定化且提高使用感的效果。
以下,根據本發明的實施例,進一步詳細地對本發明的毛髮管理用組合物的製備方法進行說明。當然,本發明並不限制於該等實施例。
實驗例 1 :根據穩定化劑的種類進行的析出試驗 ( 1 )護膚乳液組合物的製備
利用下述表1中提出的組成製備護膚用乳液。首先,混合原料1至4並於70℃下進行溶解而製備水相部,之後於70℃下溶解原料5至11而製備油相部。其次,將上述油相部添加至水相部而利用均質混合機進行攪拌,於進行一次乳化後,添加原料12而增稠。最後,去除氣泡並添加原料13及14,之後冷卻至室溫而製備乳液組合物。此時,使用SurfMellow BBG(manufactured by Toyobo Co., Ltd.)作為MEL。 [表1] ( 2 )析出試驗
於剛剛製備後、第3週及第10週利用光學顯微鏡(×500)及偏光顯微鏡(×500)根據下述內容觀察於上述實施例及比較例中製備的乳液組合物是否產生結晶,將其結果示於下述表2。 <判斷基準> ×:未觀察到結晶 ▲:觀察到微小尺寸的結晶 ○:觀察到結晶 ◎:觀察到較大尺寸的結晶 [表2]
參照上述表2,於除含有MEL的實施例1以外的所有條件下,均於3週內發生結晶析出現象。
特別是,比較例1、比較例3及比較例5的組合物自第3週開始明顯地觀察到結晶,比較例3及比較例4於第3週微小地觀察到結晶,但於第10週明顯地觀察到結晶而確認到不穩定。
可藉由光學顯微鏡及偏光顯微鏡的觀察而確認到上述結果,將其結果示於圖1及圖2。
圖1及圖2是於利用光學顯微鏡(×500)及偏光顯微鏡(×500)於製備後觀察與剛剛製備後、第3週、第10週對應的各組合物是否產生結晶、及結晶的尺寸的所得的結果。
參照圖1,可確認到不含MEL的比較例1的組合物於剛剛製備後未產生結晶,但自3週後開始觀察到結晶,於10週後,結晶的尺寸 變得非常大。
相反地,於圖2的實施例1的組合物的情形時,確認到藉由使用MEL,於10週後亦不會產生結晶而保持穩定的狀態。
實驗例 2 :根據 MEL 的含量進行的析出試驗
為了確認用作穩定化劑的MEL的最佳含量,使用與上述實施例1的組合物相同的配方製備乳液組合物而執行析出試驗。此時,改變精製水的含量而調節100重量%。 [表3]
參照上述表3,可知只有於整體化妝品組合物內添加0.25重量%以上的MEL才可抑制曲酸亞甲基二氧肉桂酸酯的結晶化,特別是只有添加0.5重量%以上的MEL才可於10週後亦可保持劑型穩定性。
此種MEL的含量相當於曲酸亞甲基二氧肉桂酸酯的含量為0.1重量%的情形,於之後的實驗中進行MEL與曲酸亞甲基二氧肉桂酸酯的含量比的實驗。
實驗例 3 :根據曲酸亞甲基二氧肉桂酸酯與 MEL 的含量比進行的析出試驗
為了確認是否根據曲酸亞甲基二氧肉桂酸酯與MEL的含量比發生結晶化,使用與上述實施例1的組合物相同的配方製備乳液組合物而執行析出試驗。此時,改變精製水的含量而調節100重量%。 [表4]
參照上述表4,若添加MEL,則至3週為止未發生結晶化,於10週後,在第12次及第13次試驗中觀察到結晶。
根據此種結果可知,只有按照最小超過2.5倍之比添加曲酸亞甲基二氧肉桂酸酯與MEL,才可抑制結晶化。
實驗例4:根據曲酸亞甲基二氧肉桂酸酯與MEL的溫度進行的析出試驗
為了確認曲酸亞甲基二氧肉桂酸酯與MEL的各溫度下的穩定性,利用上述實施例1及比較例1的乳液組合物確認是否根據溫度及時間發生結晶化。
參照上述表5,實施例1的組合物於包含室溫在內的低溫及高溫下亦未發生結晶化,於10週亦保持該結晶化而表現出較高的劑型穩定性。
與此相比,可知比較例1的組合物因未使用MEL而亦未於室溫下發生結晶化,於低溫下更嚴重地發生此種現象而低溫下的析出現象加速。

Claims (5)
- 如申請專利範圍第1項所述的方法,其特徵在於:所述曲酸亞甲基二氧肉桂酸酯相對於所述化妝品組合物的總重量為0.05重量%至0.25重量%。
- 如申請專利範圍第1項所述的方法,其特徵在於:所述甘露糖赤藻糖醇脂相對於所述化妝品組合物的總重量為0.25重量%至5.0重量%。
