)十一烷-4-酮、2,3,3-三甲基-2H-茚-1-酮、4-甲基-5-噻唑乙醇、7-甲氧基-3,7-二甲基辛醛、3-甲基-1-苯基-3-戊醇、2-苯基丙醛二甲基縮醛、八氫-2H-1-苯并吡喃-2-酮及2,6,6-三甲基環己-1,3-二烯-1-羰醛中選擇之1種以上之化合物作為有效成分。
Description
本發明係關於一種對昆蟲具有忌避活性之昆蟲忌避劑。
大多數吸血性之昆蟲會以病原體為媒介而對人或動物引起感染症或引起皮膚之炎症。尤其是蚊會傳播登革熱(dengue fever)、寨卡熱(zika fever)、黃熱病(yellow fever)、腦炎、瘧疾等對人而言嚴重之疾病,故而係衛生學上極其有害之昆蟲。
該等昆蟲本身具有感知動物之體溫之溫熱感測器、感知味道之味覺受體、感知體臭等揮發性物質之嗅覺受體、感知作為高揮發性物質之二氧化碳之二氧化碳受體等優異之化學接受系統,並採取多種多樣之行動。例如,已知雌性蚊於產卵前會尋找血,順著動物因呼吸所呼出之二氧化碳或體臭而靠近動物,並利用溫熱感測器感知體溫以發現目標動物並吸血。
近年來,研究有藉由對該昆蟲所具有之化學接受系統施加變化使昆蟲之認知感覺無力化而忌避該昆蟲之方法,並使用N,N-二乙基-3-甲基苯甲醯胺(DEET)或對薄荷烷-3,8-二醇(PMD)等作用於嗅覺受體之物質作為忌避劑。
然而,DEET具有令人不快之氣味,且皮膚滲透力較高,因此存在對幼兒或敏感性皮膚之人之使用受到限制,且持續時間亦僅為2~3小時之問題。

本發明係關於提供一種具有優異之忌避活性且安全性較高之昆蟲忌避劑。
本發明者對抑制昆蟲之熱應答之素材進行了探索,結果發現,特定之香料化合物具有優異之熱接受抑制效果,作為昆蟲忌避劑而有用。又,發現藉由將具有二氧化碳應答抑制作用之香料化合物與該等化合物組合使用,昆蟲之向人之引誘行動被良好地抑制,而作為昆蟲忌避劑更有用。
根據本發明,可提供一種藉由抑制昆蟲之熱接受應答而顯示出優異之忌避效果,與DEET或PMD等普通之忌避劑不同機制且安全性較高之昆蟲忌避劑。

該本發明之化合物可單獨使用,亦可將2種以上混合使用。
本發明之化合物之中,就昆蟲忌避效果及使用感之方面而言,作為較佳之化合物,例如可列舉:八氫-7-甲基-1,4-甲橋萘-6(2H)-酮:芬美意(Purikaton)、3-甲基-5-丙基-2-環己烯-1-酮(Grabenon)、6-甲基喹啉。
本發明之化合物可與具有二氧化碳應答抑制作用之香料化合物組合使用。
此處,所謂二氧化碳應答抑制,意指使昆蟲所具有之二氧化碳接受應答無力化或減少化,控制對二氧化碳之認知感覺,從而抑制向二氧化碳源之引誘行動。昆蟲之二氧化碳應答可藉由使用公知之單一感覺毛記錄法(Nature 461, 277-281 (10 September 2009)),將電極刺入於感覺毛後確認對二氧化碳刺激之神經放電模式而進行測定。
作為具有二氧化碳應答抑制作用之香料化合物,例如可列舉後述之參考例所示之化合物,即2-甲氧基-4-丙基苯酚、2-異丙基-4-甲基噻唑、異環檸檬醛、9-癸烯-1-醇、1-辛烯-3-醇、1-癸醇、2-乙基己酸乙酯、乙酸反式-2-己烯酯、2-環己基丙酸乙酯、苯乙酸乙酯、2,4,6-三甲基-3-環己烯-1-甲醇、1-辛醇、2,4-二甲基環己-3-烯-1-甲醇、乙酸鄰第三丁基環己酯、2-甲基丁酸、丙酸苄酯、3-甲基-2-戊基環戊-2-烯-1-酮、乙酸順式-3-己烯酯等。其中,較佳為2,4-二甲基環己-3-烯-1-甲醇(FLORALOL)、1-辛醇、乙酸順式-3-己烯酯等,更佳為2,4-二甲基環己-3-烯-1-甲醇。
於本發明中,作為成為對象之昆蟲,只要為具有熱接受應答系統者,則並無特別限定,可為吸血性或非吸血性之任一者。例如可列舉:尖音庫蚊、小型尖音庫蚊、埃及伊蚊、白紋伊蚊、中華按蚊等蚊(雌性)類;家蠅、廄腐蠅、夏廁蠅、麗蠅、麻蠅、灰地種蠅、蔥蠅、實蠅、果蠅、毛蠓、舌蠅等蠅類;及虻類、蚋類、刺蠅類、蠓類、搖蚊科等;其中,較佳為蚊類、蠅類、蚋類、刺蠅類。
如下述實施例所示,本發明之化合物抑制白紋伊蚊之熱接受系統。因此,本發明之化合物可成為昆蟲忌避劑或昆蟲之熱接受抑制,又,可用於製造昆蟲忌避劑或熱接受抑制劑。即,本發明之化合物可用於忌避昆蟲或抑制熱接受。又,於將本發明之化合物與具有二氧化碳應答抑制作用之香料化合物併用之情形時,有效地抑制白紋伊蚊之向人之引誘行動,因此該併用會成為更優異之昆蟲忌避劑,對昆蟲忌避、尤其是蚊之忌避有用。
此處,所謂「昆蟲忌避」,意指不讓昆蟲靠近對象物或者昆蟲不會靠近對象物,所謂「昆蟲之熱接受抑制」,意指使昆蟲所具有之熱接受應答無力化或減少化,抑制對熱源之認知感覺,從而抑制向熱源之引誘行動,與對昆蟲具有殺蟲力而將昆蟲驅除者不同。
再者,熱接受可藉由將昆蟲放至配置有板狀加熱器般之熱源之籠中,並計測被引誘至熱源之昆蟲之數量而進行測定。
本發明之昆蟲忌避劑或熱接受抑制劑可利用塗抹於以人為首之哺乳動物之皮膚、附著或保持於與該皮膚直接或間接地接觸之衣服、隨附品等服飾物品、或者噴灑於室內、車內等固定之空間(包含蒸散或揮散之情形)等方法來忌避昆蟲,且可根據使用態樣適當調配溶劑或各種製劑添加物等其他成分,而以組合物之形式製劑化。又,本發明之昆蟲忌避劑或熱接受抑制劑亦可作為用以對皮膚外用劑、清洗劑、皮膚化妝品、毛髮化妝品、纖維用柔軟劑、布帛處理劑等賦予昆蟲忌避活性之素材而使用。
上述組合物中之本發明之化合物之調配量亦根據製劑形態而有所不同,相對於組合物總量,較佳為0.001質量%以上,更佳為0.01質量%以上,進而較佳為0.1質量%以上,且較佳為80質量%以下,更佳為40質量%以下,進而較佳為10質量%以下,或者較佳為0.001~80質量%,更佳為0.01~40質量%,更佳為0.01~10質量%,進而較佳為0.1~10質量%。
又,於將本發明之化合物與具有二氧化碳應答抑制作用之香料化合物併用之情形時,該香料化合物於組合物中之含量相對於組合物總量,較佳為0.001質量%以上,更佳為0.01質量%以上,進而較佳為0.1質量%以上,且較佳為80質量%以下,更佳為40質量%以下,進而較佳為10質量%以下,或者較佳為0.001~80質量%,更佳為0.01~40質量%,更佳為0.01~10質量%,進而較佳為0.1~10質量%。
溶劑或添加物之種類可根據昆蟲忌避劑或熱接受抑制劑之使用形態或使用目的而適當選擇,作為溶劑,例如可列舉:甲醇、乙醇、丙醇等低級醇或水等。
又,作為添加物,可列舉各種化學處理品、化妝品等通常使用之添加物,具體而言,可列舉:界面活性劑、有機溶劑、油性成分、保濕劑、粉體、助溶劑、增黏劑、樹脂、清洗劑、防腐劑、紫外線吸收劑、無機物、香料、色素、除臭劑、精油、藥劑、植物萃取物、其他忌避劑等。
本發明之昆蟲忌避劑或熱接受抑制劑之製劑形態只要根據使用態樣適當設定即可,可為液體、乳霜、化妝水(lotion)、乳液、凝膠、軟膏劑、粉末、顆粒、噴霧或氣溶噴霧等之任一者。
於將昆蟲忌避劑或熱接受抑制劑噴灑於固定之空間之情形時,可較佳地採取使昆蟲忌避劑或熱接受抑制劑保持於載體或溶解於溶劑中並填充至具有吸液芯之容器中進行加熱或送風蒸散之方法、或者使昆蟲忌避劑或熱接受抑制劑保持於載體並於常溫下蒸散至空間中之方法。於此情形時,作為使昆蟲忌避劑或熱接受抑制劑擔載之載體材料,例如可列舉:紙漿、棉、羊毛、麻、絹等天然纖維;聚丙烯、聚乙烯、聚醯胺、聚對苯二甲酸乙二酯、聚對苯二甲酸丁二酯、聚碸、嫘縈、甲基丙烯酸樹脂、玻璃纖維等合成纖維;及沸石、滑石、白碳、矽藻土、石灰、矽膠、活性碳等多孔質。
又,作為使本發明之昆蟲忌避劑或熱接受抑制劑附著或保持於服飾物品之方法,除將昆蟲忌避劑或熱接受抑制劑塗抹於對象物品以外,可列舉如下等方法:將昆蟲忌避劑或熱接受抑制劑調配於纖維用柔軟劑中,於利用柔軟劑進行處理時使其附著或保持;或者利用昆蟲忌避劑或熱接受抑制劑預先對各種布帛基材進行處理,並使用被賦予了昆蟲忌避活性之該布帛基材製造對象物品。

[實施例]
實施例1 熱接受抑制效果之評價
(1)白紋伊蚊之準備
白紋伊蚊係使用使自Sumitomo Technoservice股份有限公司購買之卵成長而成者。於透明之塑膠鍋中覆上1 cm左右之水,並放入產有卵之濾紙。每天供給熱帶魚用餌料(四胺)作為幼蟲用餌。於大約1週後利用滴管回收蛹,將蛹轉移至20 mL用塑膠杯中,並將塑膠杯轉移至布有網之籠中。供給將10質量%蔗糖添加至25 mL塑膠管中者作為成蟲用之餌。羽化後,將雄性與雌性於同一籠中飼養5天,藉此使其等進行交尾。於飼養5天後,使用吸蟲管收集成蟲,並於冰上麻醉5分鐘後,於目視下將雄性與雌性分開並僅回收雌性。
(2)向熱源之引誘行動抑制效果之評價
將約150-200隻雌性白紋伊蚊轉移至塑膠籠(30×30×30 cm)中。由於係自籠之上部監測雌性蚊之行動,故而將塑膠籠之上表面替換成透明之亞克力板。又,將成為熱源之板狀加熱器(小動物用多面板加熱器16 W,Vivaria製造)以於籠之一角露出5×3 cm之方式自網之外側配置。以可自上表面拍攝到熱源之方式將攝錄影機(Sony製造)配置於上部並進行實驗。使用雙面膠帶將試樣塗抹用之片材固定於熱源。
以使用乙醇將表1所示之香料調整為濃度0.1%而成者作為試樣,並投予該試樣或溶劑乙醇50 μL進行實驗。為了使雌性蚊活化,將二氧化碳1%自上部投予1秒,其後,使熱源與籠接觸。對接觸30秒後之熱源拍攝照片,計數被引誘之蚊之數量。每次分析結束時將熱源所使用之片材廢棄並使用新的片材。又,考慮到試樣對蚊之影響,於一個籠中進行之評價最大設為(7試樣+1對照)。關於抑制效果,與於相同分析(7試樣+1對照)內僅塗抹乙醇溶劑之情形時之降落數進行比較,並根據下述式計算抑制效果率。

將結果示於表2。可認為本發明之化合物抑制白紋伊蚊之向熱源之引誘行動,從而抑制熱接受應答。

實施例2 向人體胳膊之引誘行動抑制評價
作為白紋伊蚊,將自Sumitomo Technoservice股份有限公司作為成蟲而購入之出生後5-10日之雌性220隻、雄性80隻放入30 cm×30 cm×30 cm之塑膠籠(bug dorm)中。
作為評價試樣,將2,4-二甲基環己-3-烯-1-甲醇(「FLORALOL」,IFF公司)、八氫-7-甲基-1,4-甲橋萘-6(2H)-酮(「Purikaton」,FIRMENICH公司)利用乙醇分別調整為濃度0.4%。又,將僅有乙醇之試樣作為對照。
將棉100%手套於單隻手上佩戴兩層,利用噴霧器向其中吹送於正背面共計為按壓14次之量(0.6 g)之上述評價試樣及對照試樣。
於賦予試樣後放置1分鐘而使乙醇揮發,然後將佩戴有賦予了試樣之手套之手放入塑膠籠中,每隔1分鐘計數降落至手套之蚊之數量。
計數1分鐘、2分鐘、3分鐘各自之降落數、及其等之總計,並計算將賦予對照試樣之情形設為100%時之降落率。將相同實驗反覆進行3次。將結果示於表3。確認到藉由將本發明之化合物與FLORALOL(2,4-二甲基環己-3-烯-1-甲醇)併用而更有效地抑制蚊之向人體胳膊之引誘行動。

參考例 二氧化碳應答抑制效果之評價
(1)白紋伊蚊之準備
白紋伊蚊係使用使自Sumitomo Technoservice股份有限公司購買之卵成長而成者。於透明之塑膠鍋中覆上1 cm左右之水,並放入產有卵之濾紙。每天供給熱帶魚用餌料(四胺)作為幼蟲用餌。於大約1週後利用滴管回收蛹,將蛹轉移至20 mL用塑膠杯中,並將塑膠杯轉移至布有網之籠中。供給將10質量%蔗糖添加至25 mL塑膠管中者作為成蟲用之餌。羽化後,將雄性與雌性於同一籠中飼養5天,藉此使其等進行交尾。於飼養5天後,使用吸蟲管收集成蟲,並於冰上麻醉5分鐘後,於目視下將雄性與雌性分開並僅回收雌性。對於雌性蚊,於麻醉下卸除翅膀、手腳,並藉由將其背部壓抵至貼附於載玻片上之雙面膠帶而進行固定,將觸角・下顎須配置於容易接入電極之位置。
(2)神經應答記錄用電極之準備
將1 Mol/L氫氧化鉀水溶液注入至50 mL注射器,利用夾子將注射器水平地固定於磁性支架(magnet stand)。將固定於電極保持器之鎢絲自水平方向插入至注射器中,一面於顯微鏡下進行觀察一面藉由電解進行研磨。利用鱷魚嘴夾自交流電源向電極保持器、注射器供給電流。
(3)神經應答記錄
將載玻片上所準備之白紋伊蚊放置於顯微鏡下,並利用×10物鏡以觸角成為視野之中心部之方式調節位置。將基準電極刺入於昆蟲之眼睛。將氣味刺激用之移液管設置於下顎須之附近,並將物鏡轉換為×100。使記錄電極靠近感覺毛,並使用顯微操作儀(micromanipulator)慎重地刺入於感覺毛。可於顯微鏡下確認到白紋伊蚊之二氧化碳感受性之感覺毛具有栓狀之形狀。於將電極刺入於感覺毛後,確認自發性之神經放電模式。利用矽管將調整為0.1%濃度之二氧化碳連接於氣味刺激裝置,並對下顎須進行1秒鐘刺激,確認二氧化碳應答。
(4)二氧化碳應答抑制效果之評價
試樣係使用利用石蠟油將表4所示之香料調整為濃度1%而成者。將15 μL之試樣滴加至濾紙(3 mm×50 mm)上,並將其插入至巴斯德移液管(Pasteur pipette)內。使用乙烯管將巴斯德移液管與刺激裝置連接,一面監測對空氣中之二氧化碳之應答,一面進行1秒鐘之氣味刺激,評價對二氧化碳之應答抑制效果。關於應答抑制效果,計測利用作為溶劑之石蠟油進行刺激後1秒鐘之神經之發電數、利用各種試樣進行刺激後1秒鐘之神經之發電數,基於其比率進行評價(N=1)。

- 如請求項3之蚊類忌避劑,其中具有二氧化碳應答抑制作用之香料化合物係自2,4-二甲基環己-3-烯-1-甲醇、1-辛醇及乙酸順式-3-己烯酯中選擇之1種以上。
- 一種蚊類忌避方法,其係將八氫-7-甲基-1,4-甲橋萘-6(2H)-酮塗抹於哺乳動物之皮膚。
