Description
本發明係有關含阿斯巴甜的酸性飲料,更詳細係關於經搭配聚合多酚(特別為聚合兒茶素)之阿斯巴甜於保存中甜味降低被抑制之酸性飲料,以及於使用聚合多酚(特別為聚合兒茶素)之含阿斯巴甜的酸性飲料中,抑制阿斯巴甜於保存中甜味降低之方法。
較砂糖具有更強甜味的高甜味度甜味料廣泛使用於減肥食品等(包含飲料)。特別在消費量高的飲料領域中,使用於減少卡路里及零卡路里等低卡路里飲料,以及概念為不使用砂糖之各種飲料。在高甜味度甜味料中,阿斯巴甜因安全性及甜味的性質優異,且亦具有增強香味效果及可減低共存物質之苦味、澀味等利用效果,而廣泛地被使用。然而,阿斯巴甜會有於酸性下產生水解而降低其甜味的問題,目前尚未發現其解決方法。
於專利文件1中記載於阿斯巴甜中添加甜味閥值較低的其他的甜味料,藉此填補因阿斯巴甜分解而引起的阿斯巴甜甜度降低,進而保持掩蓋阿斯巴甜澀味之作用。

本發明之課題係提供抑制阿斯巴甜因於酸性環境下產生分解而降低甜味之方法,以及提供阿斯巴甜於保存中甜味降低被抑制之酸性飲料。
本發明之發明者們為解決上述問題點專心研究後,發現於含阿斯巴甜的酸性飲料中,藉由添加聚合多酚,特別為來自茶類聚合多酚之聚合兒茶素,可抑制保存中阿斯巴甜甜味減低,而完成本發明。
亦即,本發明係關於含有可抑制阿斯巴甜於保存中甜味降低之濃度之聚合多酚,含阿斯巴甜的酸性飲料。特別為關於含有可抑制阿斯巴甜於保存中甜味降低之濃度之聚合多酚,含阿斯巴甜的濃度為0.04重量%以上的酸性飲料。
本發明之一實施方式係含有可抑制阿斯巴甜於保存中甜味降低之濃度之聚合多酚之含阿斯巴甜的酸性飲料,係關於聚合多酚為聚合兒茶素之飲料。
另外本發明係關於含有可抑制阿斯巴甜於保存中甜味降低之濃度之聚合多酚之含阿斯巴甜的酸性且以容器盛裝之飲料,低卡路里之飲料。
進而本發明係關於特徵為使用聚合多酚,於含阿斯巴甜的酸性飲料中,抑制阿斯巴甜於保存中甜味降低之方法。
本發明之一實施方式係關於特徵為使用聚合多酚,於含阿斯巴甜的酸性飲料中,抑制阿斯巴甜於保存中甜味降低之方法,聚合多酚係聚合兒茶素之方法。
阿斯巴甜其化合物名稱係「α-L-天冬氨-L-苯丙氨酸甲酯」,以下述式(I)所示,
具有類似果糖的味道,與砂糖相比雖稍微欠缺醇厚,但其具有砂糖200倍的甜味度。雖與砂糖相同具有約4kcal/g之熱量,但在口腔內齒垢存在下不會產生酸而不會造成蛀牙,且其苦味‧澀味較弱,是美國、日本及其他多數國家承認的安全的食品添加物。阿斯巴甜係以「PAL SWEET」為商品名由味之素(股份有限)公司於市面販售。
搭配本發明飲料之阿斯巴甜的量並無任何限制,但阿斯巴甜為0.04重量%(來自阿斯巴甜的甜度為8)以上時,於長期保存,亦即被填充於容器中,以流通型態以容器盛裝飲料保存時,可明顯確認所伴隨的甜味降低抑制作用。
聚合多酚係來自植物物質(植物化學因子:phytochemical)之一種,係於1分子中具有2個以上的酚性羥基之化合物之總稱。聚合多酚可大致上分為分子量為1,000以下的非聚合多酚,與存在有結合2個以上非聚合多酚之聚合多酚。本發明所使用之聚合多酚即為結合2個以上非聚合多酚者。聚合多酚一般亦稱作單寧。具有代表性的非聚合多酚有黃酮類(黃酮類包含以黃酮、黃酮醇、黃烷酮、黃烷酮醇、異黃酮、花青素、黃烷醇、查耳酮、橙酮為基本架構之化合物)、綠原酸、沒食子酸、鞣花酸等。反之,聚合多酚為結合2個以上的非聚合多酚之化合物,可大致分為藉由碳-碳鍵結而進行聚合的縮合型多酚,與藉由酯鍵結而進行聚合之水解型多酚,多酚可分別舉出具代表性的縮合型多酚為花青素原類,水解型多酚為沒食子單寧、鞣花單寧。
本發明之聚合多酚可使用上述之任一種聚合多酚,市售之各種聚合多酚,例如松樹皮聚合多酚(Flavangenol(註冊商標):東洋新藥股份有限公司)及葡萄種子多酚(龜甲萬股份有限公司)、可可多酚(明治製莫)等之外,可再例舉WO2005/077384號公報所記載之烏龍茶聚合多酚及與其以相同方法製造所得之紅茶聚合多酚等茶聚合多酚(本說明書中亦稱作「聚合兒茶素」)。
上述聚合多酚中以使用茶聚合多酚(亦稱作「聚合兒茶素」)為佳。使用聚合兒茶素不僅顯著獲得可抑制後甜味之作用、降低高甜味度甜味料特有的異味及苦味之作用等後味改善作用,於低卡路里(包含零卡路里)的飲料中,可賦予其稠度感(醇厚感),而可改良飲料所呈現的味道。高甜味度甜味料可用於例如減肥用甜味料等許多種用途(例如過度肥胖時限制卡路里、因糖尿病等疾病需抑制血糖值上升等),雖然具有「低卡路里甜味料」之特徵,但有醇厚感(稠度感、亦可稱之為味道的厚度)不足的問題。本發明於可解除經配合高甜味度甜味料之低卡路里(包含零卡路里)的飲料之醇厚感不足此點可說是極為適用。
聚合兒茶素係藉由以下述條件之HPLC所分析之成分,與茶黃素(栗田研究中心製)具有同樣的溶出時間(參考溶出時間:24分鐘)波形之成分(參照圖1)。

‧移動相:A:水:乙晴:三氟乙酸=900:100:0.5。B:水:乙晴:三氟乙酸=200:800:0.5。

‧梯度沖堤條件:自分析開始至5分鐘時使用B液0%,自5分鐘至11分鐘時使用B液8%,自11分鐘至21分鐘時使用B液10%,自21分鐘至22分鐘時使用B液100%,自22分鐘至30分鐘時保持B液100%,自30分鐘至31分鐘為止為0%。
‧檢出:A280nm(收集數據時間為30分鐘),以波形面積進行定量。

.標準物質:烏龍雙黃烷B(簡寫:OHBF-B)。
聚合兒茶素之量係藉由使用標準物質OHBF-B製作檢量線而求出。標準物質之OHBF-B係根據例如Chem. Pharm. Bull 37(12),3255-3563(1989)所記載之方法及特開2005-336117號公報之實施例3之方法所合成者(以純化至純度為98%以上者為佳),亦可使用自茶葉所分離者等。
該聚合兒茶素可例示非聚合之兒茶素類((+)-兒茶素、(-)-表兒茶素、(+)-沒食子兒茶素、(-)-表沒食子酸兒茶素、(-)-兒茶素沒食子酸酯、(-)-表兒茶素沒食子酸酯、(-)-沒食子兒茶素沒食子酸酯、(-)-表沒食子兒茶素沒食子酸酯)之具有藉由來自茶之酵素、酵素、光等連結了複數個構造者,具體而言可舉出以等慣用名稱稱呼之聚合多酚,式(1)之表沒食子兒茶素沒食子酸酯之二聚體,

另外,非聚合兒茶素係指兒茶素類((+)-兒茶素、(-)-表兒茶素、(+)-沒食子兒茶素、(-)-表沒食子兒茶素、(-)-兒茶素沒食子酸酯、(-)-表兒茶素沒食子酸酯、(-)-沒食子兒茶素沒食子酸酯、(-)-表沒食子兒茶素沒食子酸酯)之未進行聚合之單體。
本發明之聚合兒茶素係可藉由對茶葉進行溶媒萃取而可得。做為原料的茶葉有未發酵茶之綠茶、半發酵茶之烏龍茶、發酵茶之紅茶,可使用其中之1種或2種以上,然而以使用大量含有聚合兒茶素之半發酵茶或發酵茶之茶葉為佳。萃取溶媒可以水或熱水、甲醇、乙醇、異丙醇、醋酸甲酯等之1種或2種以上之混合物進行萃取。該茶葉之溶媒萃取物可直接使用,亦可使用經濃縮或純化者。
因藉由使用聚合多酚,特別是使用聚合兒茶素可獲得顯著的阿斯巴甜於保存中甜味降低被抑制之作用,自茶葉溶媒萃取物中選擇性地去除單體之兒茶素,以使用聚合兒茶素含有率高者為佳。另外一般因單體之兒茶素及咖啡因有苦澀味,當其搭配量大時,會損及飲食品自身所呈現的味道。因此,以使用自上述茶葉萃取物中,進行選擇性地去除單體之兒茶素以及/或咖啡因之處理之溶媒萃取物為佳。該經進行選擇性地去除單體之兒茶素以及/或咖啡因之處理之溶媒萃取物,可例示如WO2005/077384號公報所記載之含有非聚合兒茶素濃度4倍以上之聚合兒茶素者等。
本發明所使用之聚合多酚之型態可為液狀、或可為藉由噴霧乾燥及凍結破碎等而進行粉末化者。
聚合多酚係以相對於阿斯巴甜之添加重量為0.1~1.0之重量比進行添加為佳,0.1~0.5之重量比進行添加更佳。
本發明之酸性飲料係指pH為2~5(以pH為2.5~4.5更佳)之飲料,但飲料中之阿斯巴甜於保存中進行分解者,則未限定於此。酸性飲料的種類有碳酸飲料、添加果汁飲料、蔬菜飲料、營養飲料等,可適用於任何一種飲料。
本發明係藉由添加聚合多酚(特別係聚合兒茶素),而抑制阿斯巴甜於保存中甜味降低。本發明之飲料中聚合多酚之配合量,可依於利用其之飲料中配合的阿斯巴甜的量,及所使用之聚合多酚之種類適宜地加以選擇,一般,相對於飲料全體,聚合多酚為0.010重量%~0.100重量%。聚合多酚之濃度未達0.010重量%時,無法充分獲得抑制阿斯巴甜於保存中甜味降低之效果。反之,超過0.100重量%時因無法期待更好的效果,且利於經濟面,另外,因聚合多酚之種類而產生來自聚合多酚之澀味等,會引起甜味料甜味度峰值下降。
搭配本發明之聚合多酚(特別為聚合兒茶素)之飲料,不僅可抑制甜味降低,於使用阿斯巴甜之低卡路里飲料中因可彌補醇厚感不足,而適用於低卡路里飲料。於此可適用之低卡路里飲料可舉出可樂、雪碧等碳酸飲料、運動飲料、果汁飲料、乳品飲料、茶系飲料等,特別適用的目標係因大量飲用而產生肥胖問題之碳酸飲料。
可因應需要於本發明之酸性飲料中添加果汁、甜味料、酸味料、香料、其他之添加物等。
本發明之酸性飲料如上所述因可抑制於保存中風味變化,而可適用於填充於容器中流通型態之飲料,亦即以容器盛裝之飲料。以容器盛裝之飲料有藉由加熱而進行殺菌(或滅菌)之步驟,而預期會促進阿斯巴甜的分解,但於本發明之酸性飲料,其亦對因上述加熱步驟所造成之阿斯巴甜甜味降低有所功效。另外,以容器盛裝之飲料係可以紙盒、寶特瓶、鐵罐、玻璃瓶等方式提供飲料者。
以下藉由實施例更詳加說明本發明,但本發明並未因此而有所限定。
使用於溫水(95℃)中經添加0.15重量%小蘇打之7800kg小蘇打溶液,施行600kg之烏龍茶葉之萃取處理,得約7000kg之烏龍茶萃取液。保持該萃取液之液溫於60-65℃,使液體通過400kg之粒狀活性碳(KURARAY公司製GW-H32/60),去除非聚合兒茶素、咖啡因。再將該通過液(經活性碳處理後之液體)進行減壓濃縮,得含Brixl1之聚合兒茶素之烏龍茶濃縮萃取液(以下稱作萃取液A)約900kg。以下述條件之HPLC測定所得之萃取液A中聚合兒茶素、非聚合兒茶素以及咖啡因之濃度。其結果重量基準聚合兒茶素為12000ppm,非聚合兒茶素為800ppm,咖啡因為20ppm。

‧檢出:A280nm
‧標準物質:烏龍雙黃烷B(OHBF-B)。
‧聚合兒茶素之滯留時間:約25分鐘時之波峰與茶黃素之波峰一致。
將100g之紅茶葉加入2000g之熱水(90℃以上)內,保持1小時(90℃以上)後,得紅茶萃取液。將該萃取液冷卻後,進行離心分離(6000rpm,5分鐘),去除不溶物質,再進行與製造例1相同之活性碳處理,得含聚合兒茶素之紅茶萃取液(Brix2),再進行減壓濃縮而製造含聚合兒茶素之紅茶濃縮萃取液(萃取液B)。與製造例1同樣地以HPLC進行分析後,重量基準聚合兒茶素為2000ppm,非聚合兒茶素為200ppm,咖啡因為5ppm。
所使用之阿斯巴甜係PAL SWEET DIET(味之素股份有限公司),聚合多酚係製造例1所得之萃取液A(烏龍茶聚合多酚),製造例2所得之萃取液B(紅茶聚合多酚),葡萄種子聚合多酚(龜甲萬股份有限公司),松樹皮聚合多酚(東洋新藥股份有限公司)。
於0.1%阿斯巴甜水溶液中,分別調製添加萃取液A,使聚合多酚重量基準為100、200、300、500ppm者,以及添加萃取液B,使聚合多酚重量基準為300ppm者,添加葡萄種子聚合多酚重量基準為300ppm者,添加松樹皮聚合多酚重量基準為300ppm者,再加入磷酸調整pH為3.5。將其進行加熱殺菌(95℃,30秒)後,分別填充至350mL之寶特瓶內,保存於4℃或45℃中。1個月後,以4人評鑑小組進行甜味的官能評價。保存於45℃下1個月相當於進行保存10個月。官能評價係以0.1%阿斯巴甜水溶液(pH 3.5)(未添加聚合多酚)之甜味計點為5點,將0.1%阿斯巴甜水溶液(pH 3.5)(未添加聚合多酚)保存於45℃下1個月者(控制組)之甜味為1點,以5點法評價甜味的強度,再求出其平均值(表1)。可得知藉由添加聚合多酚,特別係藉由添加烏龍茶聚合多酚及紅茶聚合多酚,可抑制阿斯巴甜的甜味消失。另外,保存於4℃下添加聚合多酚之阿斯巴甜水溶液,無論聚合多酚之種類,其甜味計點均為5。
調製如表2所示配合之碳酸飲料,進行加熱殺菌(95℃,30秒)後,封入碳酸,分別填充至350mL之寶特瓶內,保存於45℃下1個月。保存後,以5人評鑑小組進行官能評價。以進行保存前之甜味計點為5點,以5點法評價甜味的強度,再求出其平均值。其結果試作品1(未添加烏龍茶聚合多酚)為2.8點,試作品2(添加烏龍茶聚合多酚)為4.1點,可得知藉由添加烏龍茶聚合多酚可抑制甜味消失。
於含阿斯巴甜之酸性飲料中藉由添加聚合多酚,特別為來自茶類的聚合多酚之聚合兒茶素,可抑制保存中阿斯巴甜之甜味降低。

Claims (13)
- 一種酸性飲料,係含阿斯巴甜的濃度為0.04重量%以上、聚合多酚的濃度為0.010重量%~0.100重量%、pH為2~5的酸性飲料,其特徵係前述聚合多酚為從烏龍茶聚合多酚、紅茶聚合多酚、葡萄種子聚合多酚及松樹皮聚合多酚中選擇出一種以上。
- 如申請專利範圍第1項之酸性飲料,其中pH為2.5~4.5。
- 如申請專利範圍第1項之酸性飲料,其中前述聚合多酚為從烏龍茶聚合多酚、紅茶聚合多酚中選擇出一種以上之聚合兒茶素。
- 如申請專利範圍第3項之酸性飲料,其中進而含有非聚合兒茶素,且聚合兒茶素較非聚合兒茶素含有較高濃度。
- 如申請專利範圍第4項之酸性飲料,其係含有非聚合兒茶素濃度之4倍以上之聚合兒茶素。
- 如申請專利範圍第3~5項中任一項之酸性飲料,其中聚合兒茶素係來自半發酵茶或發酵茶。
- 如申請專利範圍第1~5項中任一項之酸性飲料,其係填充於經加熱殺菌保存之容器之容器盛裝之飲料。
- 如申請專利範圍第1~5項中任一項之酸性飲料,其係低卡路里之飲料。
- 一種方法,其係在包含阿斯巴甜之酸性飲料中抑制阿斯巴甜於保存中甜味的降低,其特徵為使用0.010重 量%~0.100重量%之濃度之聚合多酚。
