Description
本發明為關於3-去氧基衍生物及其醫藥組成物。
肝失調症廣泛出現於人群且為早期致命的風險因子。舉例言之,非酒精性脂肪肝病(NAFLD)乃在美國影響數以百萬計成人的失調症,且指飲用極少量酒精或不飲酒者的肝臟累積過量脂肪的病況。最常見的NAFLD形式為稱作肝脂肪變性(脂肪肝)的非重症病況,其中脂肪堆積於肝細胞內。NAFLD最常見於帶有稱作代謝症候群的眾多風險因子的個體,代謝症候群的特徵為空腹血漿葡萄糖升高,對飯後血糖有或沒有不耐症,體重過重或肥胖,高血脂諸如膽固醇及三酸甘油酯類及高密度脂蛋白膽固醇濃度過低,及高血壓;但並非全部病人皆具有代謝症候群的全部表癥。肥胖被視為NAFLD的最常見起因,有些專家估計約三分之二的肥胖成人及約半數肥胖兒童可能患有脂肪肝。

糖尿病(Diabetes mellitus)俗稱為糖尿病(diabetes),係指通常以葡萄糖的製造上及利用上有代謝性缺陷為特徵,結果導致無法維持體內適當的血糖濃度的疾病或病況。

癌症的適當治療取決於腫瘤衍生自其中的細胞類型、惡性腫瘤的期別及嚴重度,及促成腫瘤的基因異常。
癌症分期系統描述癌症進行的程度。一般而言,分期系統描述腫瘤的擴散程度,及將具有相似的預後及治療的病人分類於相同的期別群組。一般而言,已經變成侵犯性或轉移性腫瘤的預後較差。
於一型分期系統中,病例被分成四期,以羅馬數字I至IV標示。於第I期,癌症經常為侷限性且通常可治癒。第II期及第IIIA期癌症通常較為惡化,可能已經侵犯周遭組織及蔓延到淋巴結。第IV期癌症包括已經蔓延到淋巴結外側部位的轉移癌。

為了評估化合物1調控FXR標靶基因的能力,進行定量RT-PCR分析試驗(參考第1圖)。HepG2細胞被選用為相關細胞系來測定本發明化合物是否能調節內生性FXR基因網絡。本發明化合物誘導FXR標靶基因的能力係藉從使用1 μΜ比較化合物及本發明化合物處理隔夜的細胞分離總RNA加以評比。已確立化合物A為強力FXR選擇性促效劑,及已確立化合物B為FXR/TGR5雙重強力促效劑。

為了評估化合物1的試管內細胞毒性,採用兩種不同的分析試驗方法。該等分析試驗藉測量ATP濃度而評估細胞存活力及藉測量LDH釋放而評估細胞毒性。腺苷三磷酸(ATP)核苷酸表示在基礎分子層面的能量來源,原因在於ATP乃多功能分子,在每個細胞內用作為輔酶,且是粒線體DNA的整合一體部分(Kangas, et al., Medical Biology, 1984, 62, 338-343;Crouch, et al., J Immunol. 為了評估化合物1的藥物-藥物交互作用的可能,調查研究六種主要CYP450同質異形體(CYP1A2, CYP2C9, CYP2C19, CYP2D6, CYP2E1, CYP3A4) (Obach, et al., J Pharmacol. Exp. Ther, 2006, 316, 336-348)。

得自以ERG鉀通道穩定地轉染的中國倉鼠卵巢細胞的膜製備品用來利用Predictor™螢光極化分析試驗評估本發明化合物對此通道的潛在抑制效果。因hERG鉀通道抑制的結果,膜極化的減低係與螢光極化(FP)的減低有直接關係。
分析試驗係藉使用受試化合物及陽性對照E-4031及塔莫西芬的16-點劑量-反應以三重重複進行。針對E-4031獲得15 nM (AmP = 163)及針對塔莫西芬獲得1.4 μΜ (ΔητΡ = 183)的IC50
