Abstract
本發明提供一種佛手瓜果實細胞原液、 佛手瓜果實細胞原液之製備方法、包含佛手瓜果實細胞原液之護膚除皺組合物及佛手瓜果實細胞原液用於製造護膚除皺組合物之用途。
Description
本發明係揭露一種佛手瓜果實細胞原液、其製備方法、包含其之護膚除皺組合物及其用途。特別係關於以新的製備方法自佛手瓜製取得果實細胞原液、所取得之果實細胞原液、包含該果實細胞原液之護膚除皺組合物及該果實細胞原液於保養品、化妝品及醫藥方面之用途。
人口老化是全球性的趨勢,而老化後最明顯的即是皮膚外觀的變化,因此延緩皮膚老化的相關產品特別受到矚目。皺紋是皮膚老化的特徵之一,而導致皺紋的誘發因素常有: 1、精神因素:情緒不好會導致人體氣血運行不暢,經常悶悶不樂、急躁、孤僻,或是常常在面部表現出愁苦、緊張、拘謹的表情,牽動表情肌就容易產生縱向或橫向的皺紋,使人出現衰老跡象。 2、不良生活習慣:經常睡眠不足、吸煙、缺少運動等,都會使皮膚的新陳代謝及調節功能受損,致使容顏憔悴,引起皮膚衰老、產生皺紋。 3、水分不足:水是皮膚重要的組成部分,也是皮膚進行新陳代謝所不可缺少的物質基礎。一般來說,皮膚含水量若低於10%,就會呈現乾燥狀態,使皮膚粗糙、鬆弛,時間長了皺紋也就產生了 4、陽光暴曬:長時間在陽光下暴曬會使皮膚變乾、變薄、失去彈性,如果使彈力纖維和角質纖維失去了正常功能,那麼皮膚也會逐漸變鬆、起皺。 5、飲食不當:長期飲食不平衡、營養狀況不佳,皮膚及肌肉組織就也會營養不良,引起的皮膚粗糙和鬆弛,會加速皺紋的產生。 6、化妝品使用不當:使用不適當的化妝品會破壞皮膚的質地,導致皮膚纖維減少、皮膚含水量下降、彈性下降、出現皺紋!過厚的粉底液也同樣會使面部出現細密的小皺紋。
從而,為預防及改善皺紋,除了防曬、保持健康生活以外,維持皮膚含水量於10%以上,也是保養皮膚、預防皺紋生成的重要步驟。健康肌膚的含水量應維持在15~20%左右,皮膚含水量過低容易形成敏感性肌膚,而含水量至少要30%以上才能使皮膚達到晶瑩剔透的效果。因此維持皮膚含水量之保溼步驟不論是在防皺、抗老等保養或是在降低皮膚過敏上皆扮演了重要的角色。
此外,由紫外線所造成的老化現象稱為光老化,其會經由有絲分裂原 (mitogen) 活化蛋白激酶 (protein kinase) 途徑之磷酸化作用而增加真皮層之基質金屬蛋白酶 (matrix metalloproteinase,MMP)之量。基質金屬蛋白酶會分解膠原蛋白從而減少皮膚中的膠原蛋白含量。皮膚若少了膠原蛋白的支撐,會變得鬆弛失去彈性,並造成角質層過度增生,皺紋增加。目前已發現的動物膠原蛋白約有二十一種型式,其中以第一型膠原蛋白的含量最多,也是功能最廣的膠原蛋白。依不同組織而有不同的膠原蛋白。於皮膚組織中,第一型膠原蛋白約占80%,而第三型膠原蛋白約占20%。因此,若能抑制基質金屬蛋白酶特別是第一型之活性及/或減少其生成,即可達到改善/增進膚質之功效。
基於安全性及天然性考量,技藝中已嘗試由天然產物中開發抗老化製劑。然依現今潮流,仍有需要發展天然、安全低敏且具更佳效果的護膚除皺產品。
佛手瓜 (Sechium edule
) 在世界各國包括中國各地有栽培,通常食用其果實、嫩葉、芽、莖和塊根。其果實營養豐富,富含蛋白質、脂肪、纖維素、碳水化合物、維生素C、核黃素、鈣、磷、和鐵等,既可做菜,也能當水果生吃。目前針對佛手瓜的各種研究已表明,佛手瓜具有抗菌、抗氧化活性、抗糖尿病、抗潰瘍、抗腫瘤、抗癲癇、保肝及保腎作用、抗高血壓、抗突變、抑制胰蛋白酵素等作用。
其中已有文獻主張含有佛手瓜果實熱水提取物的護膚乳液等可為皮膚加濕達20%以上並具有清除氧自由基作用。而已有文獻也多用水或乙醇等加熱來萃取乾果以作為研究材料。本發明提出一種新的佛手瓜果實細胞原液之製備方法,此方法以低溫及室溫製程,從新鮮佛手瓜中製備出具有更佳的護膚除皺效果之佛手瓜果實細胞原液,以求最大限度地使佛手瓜的天然活性不被破壞。
有鑑於上述習知技藝之問題,本發明之目的就是在提供一種佛手瓜果實細胞原液之製備方法、利用此種製備方法可得到具有較佳護膚除皺效果之新鮮佛手瓜果實細胞原液、包含其之護膚除皺組合物及所述佛手瓜果實細胞原液於製備護膚除皺組合物之用途。
根據本發明之一目的,提出一種製備佛手瓜果實細胞原液之方法,其包含:榨取佛手瓜,以取得新鮮佛手瓜汁液;對佛手瓜汁液進行第一次過濾以除去細小果渣;將經過濾之佛手瓜汁液於室溫下靜置沈澱;以及對佛手瓜汁液進行第二次過濾以獲得佛手瓜果實細胞原液。
較佳者,其中第一次過濾係以64目的尼龍濾布執行,而第二次過濾係以濾紙或離心過濾之方式執行。
較佳者,其中靜置沈澱之時間約為24小時以上。
根據本發明之另一目的,提出一種佛手瓜果實細胞原液其係由如上所述之佛手瓜果實細胞原液之製備方法所製得。
較佳者,其中該佛手瓜果實細胞原液以下列條件進行高壓液相層析(HPLC)分析:儀器:Agilent 1200;注射體積:50微升 ;分析管柱:Phenomenex Luna C18(2), 250 X 4.5 毫米,5微米;分析時間:45分鐘;管柱溫度:30°C;流動相:溶劑A:水與10mM NH4OAc,溶劑B: ACN與 0.1% 甲酸,梯度: A (於45分鐘內從100.0 %降至0.0 % ),B (於45 分鐘內從0.0 %升至100.0 %);流速:1.0 ml/min;測定波長:254 nm,所得之HPLC圖譜於2-15分鐘區間具有滯留時間約2.506分鐘、約2.754分鐘、約2.986分鐘、約4.048分鐘、約6.438分鐘、約8.358分鐘等的吸收峰。
根據本發明之再一目的,提出一種佛手瓜果實細胞原液於製造護膚除皺組合物之用途,其中佛手瓜果實細胞原液具有如上所述之特徵之HPLC圖譜。
根據本發明之又一目的,提出一種護膚除皺組合物,其包含20-98.5%重量百分比之具有如上所述之特徵之HPLC圖譜之佛手瓜果實細胞原液,以及保養品、化妝品或藥學上可接受之載劑。
較佳者,其中載劑可為常用於面膜、乳霜、精華液、化妝水等的稀釋劑、賦形劑等,且載劑可包含溶液類、乳液類(例如,微乳液與奈米乳液)、凝膠類、固體類與脂質體類,但不限於此。
為了讓上述目的、技術特徵以及實際實施後之增益性更為明顯易懂,於下文中將係以較佳之實施範例輔佐對應相關之圖式來進行更詳細之說明。
除非本文中另外定義,否則本文中所使用之用語應以與本發明所屬技術領域之通常知識者的一般理解相同之含義進行解釋。且除非上下文另有表示,否則本說明書中所使用之單數用語亦包含複數。
本文中所使用之「重量百分比」,係指在施用之前,相對於添加載體、其他活性成分後之組合物的重量百分數。
第1圖係為描繪依據本發明實施例之佛手瓜果實細胞原液之製備方法之流程圖。
如第1圖所示,本發明實施例之佛手瓜果實細胞原液之製備方法包含:榨取佛手瓜汁液 (步驟101);對新鮮佛手瓜汁液進行第一次過濾(步驟103);於室溫下靜置經過濾之佛手瓜汁液(步驟105);對佛手瓜汁液進行第二次過濾(步驟107)以獲得果實細胞原液。
於步驟101中,將新鮮之佛手瓜果實用水洗淨後切塊,較佳為切成約2公分寬的長條,放入家用或工業級果汁機中以榨取佛手瓜汁液。於一較佳實施例中,所吐出的果渣可重新榨取2-3次以提高收率。
於步驟103中,將得自步驟101之佛手瓜汁液以64目的尼龍濾網進行第一次過濾,以除去佛手瓜汁液中之細小殘渣。
於步驟105中,將已經過步驟103濾除細小殘渣之佛手瓜汁液於室溫下靜置24小時以上,待佛手瓜汁液出現沈澱。
於步驟107中,利用濾紙過濾、或是細砂芯漏斗過濾、或離心過濾的方法進行第二次過濾,濾除於步驟105中沈澱之澱粉、葉綠素、細胞壁等非水溶性物質,以獲得為澄清之佛手瓜果實細胞原液。於一實施例中,第二次過濾係以濾紙過濾或離心之方式執行。於一實施例中,濾紙過濾係利用2號濾紙,以抽氣或重力過濾執行。於一實施例中,第二次過濾係在10,000轉/分的條件下,離心過濾10分鐘。
本發明之實施例亦提供一種佛手瓜果實細胞原液,該佛手瓜果實細胞原液係以如上所述之佛手瓜果實細胞原液之萃取方法所製得。其中該佛手瓜果實細胞原液於儀器為Agilent 1200 HPLC system;注射體積為50微升;分析管柱為Phenomenex Luna C18(2), 250 X 4.5 毫米,5微米;分析時間為45分鐘;管柱溫度為30°C;流動相為溶劑 A: 水與10mM NH4OAc,溶劑B: 乙腈(acetonitrile)與 0.1% 甲酸(formic acid),梯度: A (於45分鐘內從100.0 %降至0.0 % ),B (於45 分鐘內從0.0 %升至100.0 %);流速為1.0 ml/min;測定波長為254 nm之條件下,進行高壓液相層析(HPLC)分析,可得到如第2圖所示之HPLC圖譜,其於滯留時間約2.506分鐘、約2.754分鐘、約2.986分鐘、約4.048分鐘、約6.438分鐘、約8.358分鐘具有吸收峰。
以下利用膠原蛋白酶之螢光受質(螢光多胜肽受質I,Fluorogenic peptide substrate I),評估根據本發明之佛手瓜果實細胞原液對膠原蛋白酶的抑制效果。於此所使用之膠原蛋白酶為利用基因重組所獲得之廣效性膠原蛋白酶。
首先,將480微升之水注入至微量離心管中,再添加80微升之10倍稀釋緩衝溶液(以5毫升之1莫耳濃度氨基丁三醇(Tris)(pH7.8)、1毫升之1莫耳濃度氯化鈣、3.75毫升之4莫耳濃度氯化鈉、及0.25毫升之水配製而成)、80微升之佛手瓜果實細胞原液(SE-fresh)、80微升之膠原蛋白酶(0.01毫克/毫升)、及80微升之螢光受質(螢光多胜肽受質I,10微莫耳濃度(μM))。將所得之溶液混合均勻後,將其置入37℃之培養箱中反應20小時。接著,以BioTek Synergy HTX多功能化學分析儀(激發光:490奈米;放射光:520奈米)測試該溶液之吸收值。最後利用以下公式計算佛手瓜果實細胞原液對膠原蛋白酶之抑制率。以相同步驟進行三次試驗後,計算抑制率之平均值及標準偏差。

實驗結果如第3圖所示,佛手瓜果實細胞原液相較對照物對MMP-1有更為顯著的抑制活性。由實驗結果可知,本發明實施例提供之佛手瓜果實細胞原液對基質金屬蛋白酶-1 ( Matrix Metalloproteinase-1, MMP-1) 的活性有顯著抑制作用。
本發明之實施例亦提供一種佛手瓜果實細胞原液於製造具有護膚及除皺功能之組合物之用途。於本實施例中,具有如第2圖所示之HPLC圖譜之佛手瓜果實細胞原液可直接、經室溫減壓濃縮、冷凍乾燥或以任何其他習知之方法調配成具有護膚除皺功能的組合物。
根據本發明實施例之組合物可包括上述之佛手瓜果實細胞原液以及保養品、化妝品或藥學上可接受之適用於皮膚之載劑。本發明中所使用之載劑無特別限制,只要所用之載劑為無毒性且與本發明中之佛手瓜果實細胞原液不發生反應之成惰性載劑即可。載劑可為常用於面膜、乳霜、精華液、化妝水等的稀釋劑、賦形劑等。此種載劑可包含溶液類、乳液類(例如,微乳液與奈米乳液)、凝膠類、固體類與脂質體類,但不限於此。舉例而言,載劑可包括水(例如,去離子水)、植物油、礦物油、酯類、醇類、烴油、矽油、蠟類、或上述物質之組合或混合物等。
載劑可包含溶液類、乳液類(例如,微乳液與奈米乳液)、凝膠類、固體類與脂質體類,但不限於此。適用之乳液類之非限制性實例包括油包水乳液、水包油乳液、水包聚矽氧乳液、聚矽氧烷包水乳液、水包蠟乳液、水-油-水三相乳液或呈乳霜、凝膠或微乳液狀之類似物質。乳液可包括乳化劑,如非離子性、陰離子或兩性表面活性劑。
在一實施例中,本發明之組合物視情況可包含,本發明所屬之技術領域之通常知識者所習知之用於保養品、化妝品及醫藥上之其他添加物,包括但不限於:填充劑、乳化劑、抗氧化劑、表面活性劑、成膜劑、螯合劑、膠凝劑、增稠劑、柔軟劑、保濕劑、濕潤劑、維生素、礦物質、黏度及/或流變改質劑、防曬劑、角質溶解劑、去色素劑、類視色素、激素化合物、α-羥基酸、α-酮酸、抗真菌劑、抗微生物劑、抗病毒劑、止痛劑、脂質化合物、抗過敏劑、H1或H2抗組織胺、消炎劑、抗刺激劑、抗增生藥、免疫系統加強劑、免疫系統抑制劑、除痤瘡劑、麻醉劑、消毒劑、驅蟲劑、皮膚冷卻化合物、皮膚保護劑、皮膚滲透增強劑、去角質劑、潤滑劑、芳香劑、著色劑、去色素劑、色素淡化劑、防腐劑、安定劑、藥劑、光安定劑、中和劑、膠原酶抑制劑、彈性蛋白酶抑制劑、皮膚豐滿劑、光散射劑、剝離劑及其混合物。
在另一實施例中,本發明之組合物可進一步包含植物性萃取物或動物性萃取物之其他萃取物。動物性萃取物之實例包含,但不限於:膠原蛋白、水解膠原蛋白、水解蛋白、彈力蛋白、胎盤素等。植物性萃取物包含取自蘆薈、洋甘菊、銀杏、金縷梅、甘草根、鼠尾草、海藻、印度冷杉、綠茶、米、當歸川芎、當歸靈芝、酪梨、夾竹桃等。
在一實施例中,本發明之組合物包含98.5重量百分比之如上所述之佛手瓜果實細胞原液及1重量百分比之載劑。較佳者,本發明之組合物包含20重量百分比之如上所述之佛手瓜果實細胞原液及80重量百分比之載劑。最佳者,本發明之組合物包含20重量百分比之如上所述之佛手瓜果實細胞原液及80重量百分比之載劑。
其中,本發明之組合物可為,例如洗劑、乳霜、漿液、噴霧、氣霧劑、餅狀、軟膏、香精、凝膠、糊狀物、貼片、筆狀物、濕紙巾、面膜、棒狀、泡沫物、酏劑、濃縮液等之形式。更具體的,可為用以局部施用於具有皺紋及/或細紋之皮膚上之乳液或乳霜之形式。較佳地,組合物可直接施用於細紋及/或皺紋上。該組合物適用於任何皮膚表面上之細紋與/或皺紋,包括,但不限於,臉部、頸部、與/或手部之皮膚 。
以下之非限制性之實例有助於本發明所屬技術領域中之通常知識者實施本發明。此些實例不應視為對本發明之限制。本發明所屬技術領域之通常知識者可在不背離本發明之精神或範疇的情況下對本文所述之實例進行修改及變化,而仍屬於本發明之範圍。 實例 實例1:組合物1之製備
步驟1. 製備佛手瓜果實細胞原液
將2,508克的新鮮之佛手瓜果實用水洗淨後,切成約2公分寬的長條,放入Hurom HR808 慢磨料理機榨取佛手瓜汁液,所吐出的果渣重新榨取2次以提高收率。將所得之佛手瓜汁液以64目的尼龍濾布過濾去除殘渣後,得到佛手瓜汁液2,277克,在室溫下靜置24小時,待佛手瓜汁液中之澱粉、葉綠素、細胞壁等不溶於水之物質充分沈澱之後,利用2號濾紙濾除沈澱,獲得澄清之佛手瓜果實細胞原液2,240克。
步驟2. HPLC分析
(2)待測樣品: 未稀釋之澄清佛手瓜果實細胞原液
(3)HPLC條件
儀器:Agilent 1200 HPLC system;注射體積:50微升;分析管柱:Phenomenex Luna C18(2), 250 X 4.5 毫米,5微米;分析時間:45分鐘;管柱溫度:30°C;流動相:溶劑 A:水與10mM NH4OAc,溶劑 B: 乙腈與 0.1% 甲酸,梯度: A (於45分鐘內從100.0 %降至0.0 % ),B (於45 分鐘內從0.0 %升至100.0 %);流速:1.0 ml/min;測定波長:254 nm。
HPLC分析之結果如第2圖所示。其於滯留時間約2.506分鐘、約2.754分鐘、約2.986分鐘、約4.048分鐘、約6.438分鐘、約8.358分鐘具有吸收峰。
步驟3. 完成組合物1之製造
取98.5克之上述佛手瓜果實細胞原液,加入0.75克的卡波姆(carbomer) 使其充分漲潤後,加入 0.75 克的三乙醇胺(triethanolamine),攪拌均勻以製備組合物1。觀察志願者使用組合物 1 後膚質 改善之功效
如第4圖和第5圖所示,志願者使用組合物1後可明顯消除手部、 面部、頸部等處的皺紋, 恢復皮膚彈性,改善皮膚整體外觀,超聲波皮膚掃描顯示可顯著增加皮下膠原蛋白的密度和皮膚厚度。 實例2:組合物2之製備
將20克佛手瓜細胞原液,加入76.75克的水稀釋後,再加入1克的戊二醇(pentylene glycol)、0.25克的三仙膠(xanthan gum)與2克的Leuconostoc/Radish Root Ferment Filtrate,攪拌均勻,製成佛手瓜面膜液。將布膜摺疊置入鋁袋中,再倒入上述製備的佛手瓜面膜液25克,以完成佛手瓜面膜(組合物2)的製作。觀察志願者使用佛手瓜面膜 ( 組合物 2) 後膚質 改善之功效
志願者(女,33歲) 洗完全臉後,敷佛手瓜面膜15分鐘,撕下面膜並等待5分鐘,直到面膜液完全吸收。使用前與使用後分別使用全臉型分析儀(IRV Derma) 與超聲波皮膚掃描儀 (Cortex Dermascan USB) 檢測。結果如第6圖和第7圖所示,志願者使用組合物2後可明顯縮小皮膚毛孔、減少紅斑和深層斑、消除皺紋,超聲波皮膚掃描顯示可顯著增加皮下膠原蛋白的密度。
從而可得知含有依據本發明之萃取方法獲得之佛手瓜果實細胞原液之組合物,具有顯著之護膚除皺效果。
以上所述僅為舉例性,而非為限制性者。任何未脫離本發明之精神與範疇,而對其進行之等效修改或變更,均應包含於後附之申請專利範圍中。

Claims (9)
- 一種佛手瓜果實細胞原液之萃取方法,其包含以下步驟: 榨取一佛手瓜,以取得一佛手瓜汁液;對該佛手瓜汁液進行一第一次過濾;將經過濾之該佛手瓜汁液於室溫下靜置沈澱一時間;以及 對該佛手瓜汁液進行一第二次過濾以獲得一佛手瓜果實細胞原液。
- 如申請專利範圍第1項所述之萃取方法,其中該第一次過濾係以64 目的濾網執行。
- 如申請專利範圍第1項所述之萃取方法,其中該第二次過濾係以濾紙過濾或離心之方式執行。
- 如申請專利範圍第1項所述之萃取方法,其中靜置沈澱之該時間約為24小時以上。
- 一種佛手瓜果實細胞原液,其係經由如申請專利範圍第1項所述之方法製得。
- 如申請專利範圍第5項所述之佛手瓜果實細胞原液,其中該佛手瓜果實細胞原液以下列條件進行高壓液相層析(HPLC)分析:儀器:Agilent 1200 HPLC system;注射體積:50微升;分析管柱:Phenomenex Luna C18(2), 250 X 4.5 毫米,5微米;分析時間:45分鐘;管柱溫度:30°C;流動相:溶劑 A:水與10mM NH4OAc,溶劑 B: 乙腈與 0.1% 甲酸,梯度: A :於45分鐘內從100.0 %降至0.0 %,B :於45 分鐘內從0.0 %升至100.0 %;流速:1.0 ml/min;測定波長:254 nm,所得之HPLC圖譜於2-15分鐘區間具有滯留時間約2.506分鐘、約2.754分鐘、約2.986分鐘、約4.048分鐘、約6.438分鐘、及約8.358分鐘的吸收峰。
- 一種佛手瓜果實細胞原液於製造護膚除皺組合物之用途,其中該佛手瓜果實細胞原液係如申請專利範圍第6項所述之佛手瓜果實細胞原液。
- 一種護膚除皺組合物,其包含20%至98.5%重量百分比之如申請專利範圍第6項所述之佛手瓜果實細胞原液,以及保養品、化妝品或藥學上可接受之一載劑。
- 如申請專利範圍第8項所述之護膚除皺組合物,其中該載劑包含溶液類、乳液類(例如,微乳液與奈米乳液)、凝膠類、固體類與脂質體類。
