Abstract
本發明提供一種膏劑,其係於基布上具備膏體層之膏劑,上述膏體層含有水楊酸二醇酯、水溶性高分子、水及甘油,甘油含量以質量基準計,為水楊酸二醇酯含量之8~15倍。
Description
本發明係有關膏劑。
膏劑係於基布上塗佈含有藥劑之膏體而製造之貼附劑之一種,膏劑一般含有較多水分且膏體較厚。膏劑由於具有此種構成,故可促進有效成分之皮膚透過,減低對皮膚之刺激。
專利文獻1:日本特開平10-114647號公報
然而,含水楊酸二醇酯之膏劑,有於膏體層中之水楊酸二醇酯之保存安定性低,長時間保存後無法充分發揮鎮痛作用之情況。
因此,本發明之目的係提供水楊酸二醇酯之
保存安定性優異之膏劑。
本發明提供一種膏劑,其係於基布上具備膏體層之膏劑,上述膏體層含有水楊酸二醇酯、水溶性高分子、水及甘油,甘油含量以質量基準計,為水楊酸二醇酯含量之8~15倍。
本發明之膏劑中,除以上述成分作為必須以外,亦具有使水楊酸二醇酯與甘油之含有比例成為特定比例之構成。基於此種構成,顯著提高水楊酸二醇酯之保存安定性。
又,藉由使甘油含量以質量基準計,為水楊酸二醇酯含量之8~14倍,可更提高水楊酸二醇酯之保存安定性。
上述膏體層之pH較好為4.7~5.1,更好為4.9~5.1。藉由使pH為4.7以上(更好為4.9以上),有膏體之凝集力更優異之傾向,pH若為5.1以下,有水楊酸二醇酯之保存安定性更優異之傾向。
又,上述膏體層較好含有聚(丙烯酸甲酯/丙烯酸2-乙基己酯)。藉由使膏體層含有聚(丙烯酸甲酯/丙烯酸2-乙基己酯),即使經過長時間亦可維持附著力。
上述水溶性高分子較好含有聚丙烯酸或聚丙烯酸中和物。藉由含有該等成分,可於提高水楊酸二醇酯
之保存安定性之狀態提高附著性。
依據本發明之膏劑,水楊酸二醇酯之保存安定性優異,即使長時間保存後亦可充分發揮鎮痛作用。且,亦確保貼劑所要求之保濕性。
以下顯示一實施形態說明本發明。
本說明書中,所謂水楊酸二醇酯之保存安定性於60℃保存1個月後之水楊酸二醇酯含量與膏劑調製時比較為89%以上。
本發明之一實施形態係一種膏劑,其係於基布上具備膏體層之膏劑,且前述膏體層含有水楊酸二醇酯、水溶性高分子、水及甘油,甘油含量以質量基準計,為水楊酸二醇酯含量之8~15倍。
作為基布,舉例有織布、不織布、樹脂膜、發泡薄片及紙,作為織布舉例為例如編織布。使用織布、不織布或樹脂膜作為基布時,作為其原料舉例為例如聚乙烯、聚丙烯、聚丁烯等之聚烯烴,聚對苯二甲酸乙二酯等聚酯,縲縈、聚胺基甲酸酯及棉,該等可單獨使用1種,亦可組合2種以上使用。且基布可為具有單層構造者,亦可具有多層構造。作為基布之材料,更好為聚酯。
作為基布較好為不織布或織布,最好為具有
特定伸長回復率織不織布或織布。此處,所謂伸長回復率意指根據「JIS L 1096織物及編織物之坯料試驗方法」所測定之值。藉由使用具有伸長回復率之不織布或織布,於貼附於關節等之可動部時,由於基布可根據貼附部位之移動而伸縮故較佳。
基布為不織布時,50%伸長時荷重較好例如縱方向(長軸方向)為1~5N/2.5cm,橫方向(短軸方向)為0.1~3N/2.5cm。又,50%伸長回復率為例如60~99%,較好為65~95%,更好為70~90%。適當之基布的單位面積重量為例如80~120g/m2,較好為90~110g/m2。適當之基布厚度為例如0.5~2mm。且,基布之剛軟度(剛軟度之測定方法係利用JIS L 1096 45°懸臂法)設為例如於縱方向(長軸方向)20~40mm,橫方向(短軸方向)10~35mm,較好於縱方向(長軸方向)25~35mm,橫方向(短軸方向)15~30mm。
使用織布尤其是編織布作為基布時,亦包含線圈係經由圓型針織、經編、緯編等集成而成並加工為布狀之編織布。編織布之較佳例舉例為使聚酯系、尼龍系、聚丙烯系、縲縈系等材料之1種或2種以上組合而成之編織布,其中更好為與藥物之相互作用較少之聚酯系之聚對苯二甲酸乙二酯所成之編織布。
尤其,基布為織布時,50%伸長時荷重較好例如縱方向(長軸方向)為1~5N/2.5cm,橫方向(短軸方向)為0.1~3N/2.5cm。又,50%伸長回復率為例如
60~99%,較好為65~95%,更好為70~90%。又,基布之剛軟度可設為例如於縱方向(長軸方向)10~30mm,橫方向(短軸方向)10~30mm,較好於縱方向(長軸方向)15~25mm,橫方向(短軸方向)15~25mm。
含有水之膏體延展於織布上時會通過織布之網眼而有水滲出之虞,但藉由將聚對苯二甲酸乙二酯織布之單位面積重設為80~150g/m2,有可使膏體中所含之水確實通過織布之網眼而不滲出地延展之傾向,且可維持織布與膏體之間之固定性。
又,聚對苯二甲酸乙二酯織布較好縱方向(長軸方向)模數為2~12N/5cm,橫方向(短軸方向)模數為2~8N/5cm(模數之測定方法係利用JIS L 1018)。若為低於2N/5cm(縱方向)或2N/5cm(橫方向)之模數,則塗佈膏體時有黏著劑滲入織布所延伸之網眼中,而使作為膏劑之機能降低之情況。且,若為高於12N/5cm(縱方向)或8N/5cm(橫方向)之模數,則有伸縮性差,應用於彎曲部時難以追隨皮膚伸張之情況。
膏體層中所含之水楊酸二醇酯為非類固醇系消炎鎮痛劑之1種,具有緩和炎症症狀之效果。且,水楊酸二醇酯為具有消炎鎮痛作用之化合物,水溶性比水楊酸甲酯優異。
膏體層含有水溶性高分子,該水溶性高分子意指具有親水性基之高分子。作為親水性基,舉例為例如羥基、羧基、胺基等。藉由含有水溶性高分子,可更長時
間保持膏劑中之水分。
作為水溶性高分子較好包含聚丙烯酸或聚丙烯酸中和物(該等有時稱為「水溶性丙烯酸聚合物」)。藉由使膏體層含有聚丙烯酸或其中和物,可獲得附著性更優異之膏劑。
含有聚丙烯酸作為水溶性高分子時,其含量以膏體層之總質量為基準,較好為1~5質量%,更好為2~6質量%。藉由使水溶性高分子之含量為1質量%以上,有更提高膏體層之成型性及保型性之傾向,藉由使聚丙烯酸之含量為5質量%以下,有膏體層之硬度不易提高,更提高對皮膚之密著性之傾向。
聚丙烯酸中和物可為聚丙烯酸完全中和物,亦可為聚丙烯酸部分中和物,亦可為該等之混合物。作為聚丙烯酸中和物舉例聚丙烯酸鹽,例如可使用鈉鹽、鉀鹽、鈣鹽、銨鹽等。
作為聚丙烯酸中和物,基於初期附著力與經時附著力均提高而言,較好為聚丙烯酸部分中和物。聚丙烯酸部分中和物係於1個聚合物鏈中以任意比例存在源自丙烯酸之構造單位與源自丙烯酸鹽之構造單位者。作為聚丙烯酸部分中和物較好使用1個聚合物鏈中之羧基中20~80莫耳%經中和者。
含有聚丙烯酸中和物作為水溶性高分子時,其含量以膏體層之總質量為基準較好為1~6質量%,更好為2~6質量%。藉由使聚丙烯酸中和物之含量為1質量%
以上,可充分獲得聚丙烯酸中和物之附著力,藉由使聚丙烯酸中和物之含量為6質量%以下,可提高膏體層之成型性及保型性。又,作為水溶性高分子,併用聚丙烯酸與聚丙烯酸中和物(較好為聚丙烯酸部分中和物)時之各較佳含量亦如上述。
作為水溶性高分子,亦可包含水溶性丙烯酸聚合物以外者,例如明膠、聚乙烯醇、聚乙烯吡咯啶酮、褐藻酸鈉、羥基丙基纖維素、羧甲基纖維素鈉(carmellose sodium)、甲基纖維素、卡拉膠。該等可單獨使用1種,亦可組合2種以上使用。作為水溶性高分子,較好為羧甲基纖維素鈉、明膠或聚乙烯醇。又,該等亦可與水溶性丙烯酸聚合物組合使用。
包含水溶性丙烯酸聚合物以外之水溶性高分子作為水溶性高分子時,其含量以膏體層之總質量為基準較好為3~10質量%。水溶性高分子之含量若為3質量%以上,則有膏體層之凝集力易於提高之傾向,若為10質量%以下,則膏體層中所含之水楊酸二醇酯有易於均一分散之傾向。
甘油發揮抑制水分自膏劑之膏體層蒸發之效果。甘油之含量以膏體層質量為基準,較好為3~70質量%,更好為4~60質量%。
且,甘油之含量相對於水楊酸二醇酯之含量,以質量基準,較好為8~15倍,更好為8~14倍,又更好為8~13.6倍。
藉由使膏體層含有水,可提高水楊酸二醇酯之皮膚透過性,不僅可緩和炎症症狀之1種的熱感,亦可更有效地發揮水楊酸二醇酯之消炎鎮痛作用。
水之含量以膏體層之質量為基準,較好為10~90質量%,更好為15~88質量%,又更好為18~85質量%。
膏體層之質量可為例如214~1000g/m2,亦可為400~1000g/m2,又可為400~650g/m2。較好,藉由成為400~650g/m2,可使服貼感良好,可提高更長時間之附著力。膏體層之質量若為上述範圍,可使膏劑全體之厚度減小,易於追隨皮膚,再者,由於貼附時與周緣部之階差變小,故有不易剝離之傾向。
膏體層亦可進而含有聚(丙烯酸甲酯/丙烯酸2-乙基己酯)。以往之膏劑係膏體層之重量小時,水含量易降低,附著力易降低。然而,藉由使膏體層含有聚(丙烯酸甲酯/丙烯酸2-乙基己酯),即使膏體層之質量較小時,即使經過長時間後亦有維持充分附著力之傾向。
作為聚(丙烯酸甲酯/丙烯酸2-乙基己酯),較好為使用水作為媒劑之水性乳液。作為聚(丙烯酸甲酯/丙烯酸2-乙基己酯)乳液,又較好為使用聚氧乙烯壬基苯基醚作為界面活性劑或保護膠體之乳液。且,藉由加熱媒劑之沸點以上(例如於105℃ 3小時)之蒸發殘留物(不揮發分)較好為57~61%。作為此種乳液舉例為NICAZOLE TS-620(商品名,日本CARBIDE工業股份有
限公司製)。依據醫藥品添加劑規格(2013年),將NICAZOLE TS-620於水浴上蒸發乾固後,於105℃乾燥3小時時,蒸發殘留物之量為57~61%。
膏體層之pH較好為4.7~5.1,更好為4.9~5.1。藉由使pH為4.7以上,對皮膚之刺激性較少,藉由使pH為5.1以下,可提高膏劑之成型性及保型性。尤其基布為織布,尤其是編織布時,於形成膏體層時有產生滲出之情況,但於pH為4.9~5.1時,有抑制滲出之傾向。又,pH例如可依據日本藥典一般試驗法之pH測定法,使用玻璃複合電極,以純化水將試料稀釋至20倍而測定。
膏體層中亦可進而添加作為其他成分之其他藥劑、溶解輔助劑、交聯劑、保濕劑、清涼化劑、安定化劑、無機粉體、著色料、著香料、pH調整劑等。
作為其他藥劑只要具有經皮吸收性即可,舉例為聯苯乙酸(felbinac)、氟比洛芬(flurbiprofen)、待克菲那(diclofenac)、待克菲那鈉、水楊酸甲酯、吲哚美他辛(indometacin)、酮普芬(ketoprofen)、布洛芬(ibuprofen)等之非類固醇系抗炎症劑或該等之酯,二苯胺明(diphenhydramine)、氯苯那敏(chlorpheniramine)等之抗組織胺劑,阿斯匹靈、乙醯胺芬(acetaminophen)、布洛芬、洛索洛芬鈉(loxoprofen sodium)等之鎮痛劑,利多卡因、二丁卡因(dibucaine)等之局部麻醉劑,氯化琥珀膽鹼
(suxamethonium chloride)等之肌肉鬆弛劑,克催馬唑(clotrimazole)等之抗真菌劑,氯壓定(clonidine)等降壓劑,硝基甘油、硝酸異山梨酯等血管擴張劑,維他命A、維他命E(生育酚)、乙酸生育酚、維他命K、奧托硫辛(octothiasine)、丁酸核黃素等維他命類,前列腺素類,東莨若鹼(scopolamine)、芬坦尼(fentanyl)、唐辛子萃取物、壬酸香草醯胺等。

溶解輔助劑是為使膏體層中所含之成分不析出而添加者。作為溶解輔助劑可舉例為例如克羅米通;N-甲基吡咯啶酮;聚乙二醇(PEG)、聚丁二醇等之聚烷二醇;肉豆蔻酸異丙酯、己二酸二乙酯等之脂肪酸酯;單硬脂酸聚乙二醇等之氧伸烷基脂肪酸酯;聚氧伸烷基山梨糖醇酐脂肪酸酯等之脂肪酸酯;聚氧乙烯硬化蓖麻油;聚山梨糖醇酯80等之界面活性劑。該等溶解輔助劑可單獨使
用1種,亦可組合2種以上使用。溶解輔助劑之含量以膏體層之質量為基準,較好為0.1~10質量%。
交聯劑是為了調整聚丙烯酸或其中和物、或聚(丙烯酸甲酯/丙烯酸2-乙基己酯)之交聯反應之進行度而添加者,藉由調整交聯劑之含量,可調整膏劑對皮膚之追隨性。作為交聯劑,可使用作為膏劑一般所使用者。
作為保濕劑只要可抑制隨著時間經過之水分自膏體層之蒸發,則無特別限制。作為保濕劑舉例為例如山梨糖醇、乙二醇、丙二醇、聚乙二醇、液體石蠟、1,3-丙二醇、1,4-丁二醇等之多元醇。該等保濕劑可單獨使用1種,亦可組合2種以上使用。保濕劑之含量以膏體層之質量為基準,較好為3~70質量%。
清涼化劑係於使用膏劑時對使用者賦予冷感、清涼感者,亦可為具有芳香者。作為清涼化劑可舉例為例如百里酚、l-薄荷醇、dl-薄荷醇、l-異番薄荷醇、薄荷油等,較好使用l-薄荷醇。清涼化劑之含量以膏體層之質量為基準,較好為0.5~3質量%。
安定化劑為提高生理活性物質對於光(尤其是紫外光)、熱或氧之保存安定性。作為安定化劑舉例為例如羥苯甲酮(oxybenzone)、二丁基羥基甲苯(BHT)、依地酸鈉、UV吸收劑(例如二苯甲醯基甲烷衍生物)等。安定化劑含量以膏體層之質量為基準較好為0.01~1質量%。
無機粉體系為了調整使用膏劑時之黏住感或
抑制膏體層朝基布側之滲出而添加。作為無機粉體舉例為例如氧化鋁、輕質氧化矽、氧化鈦、合成矽酸鋁等。無機粉體之含量以膏體層之質量為基準,較好為0.1~10質量%。
膏劑亦可具備剝離襯墊。剝離襯墊係以膏體層為中心層合於與基布相反側之面。具備剝離襯墊時,於保存時,可抑制膏體層中之水含量降低,可減低污物等對於膏體層之附著。
作為剝離襯墊之材料並未特別限定,可使用本技藝者一般悉知之襯墊。剝離襯墊之材料舉例為例如聚乙烯、聚丙烯、聚對苯二甲酸乙二酯及紙,可單獨使用1種,亦可組合2種以上使用。
膏劑亦可於鋁箔包內部保存。藉由保存於鋁箔包內部,可抑制膏體中之水含量降低,可減低污物等對膏體層之附著。
本實施形態之膏劑例如可如下般製造。亦即,將水楊酸二醇酯、水溶性高分子(聚丙烯酸及/或其中和物)、水及甘油混合,根據需要添加上述之其他成分而調製膏體液。其次,將所得膏體液均一延展於剝離襯墊上,於其上層合基布,藉由剝離掉剝離襯墊而於基布上形成膏體層,可獲得膏劑。
本發明亦提供膏劑之製造方法,其係於基布上具備膏體層之膏劑之製造方法,其包含自將水楊酸二醇酯、水溶性高分子、水及甘油混合而得之膏體液形成膏體
層之步驟,且甘油含量以質量基準計,為水楊酸二醇酯含量之8~15倍。
甘油含量以質量基準計,較好為水楊酸二醇酯含量之8~15倍,更好為8~14倍,又更好為8~13.6倍。且較好將膏體層之pH調整為4.7~5.1,更好調整為4.9~5.1。膏體液亦可含有聚(丙烯酸甲酯/丙烯酸2-乙基己酯)。水溶性高分子較好包含聚丙烯酸或聚丙烯酸中和物。
進而本發明亦提供一種方法,其係於基布具備膏體層之水楊酸二醇酯之保存安定性之提高方法,係將水楊酸二醇酯、水溶性高分子、水及甘油混合所得之膏體液形成膏體層,且甘油含量以質量基準計,為水楊酸二醇酯含量之8~15倍。
甘油含量以質量基準計,較好為水楊酸二醇酯含量之8~15倍,更好為8~14倍,又更好為8~13.6倍。且較好將膏體層之pH調整為4.7~5.1,更好調整為4.9~5.1。膏體液亦可含有聚(丙烯酸甲酯/丙烯酸2-乙基己酯)。水溶性高分子較好包含聚丙烯酸或聚丙烯酸中和物。
以下顯示實施例及比較例,更具體說明本發明。
依據表1記載之組成,充分混合各成分,調製膏體液。將所得膏體液均一延展於剝離襯墊上,於其上層合基布,藉由剝離掉剝離襯墊,分別獲得實施例1~8及比較例1、2之膏劑。
依據日本藥典之記載,測定所得膏劑之膏體層之pH,其值示於表2。又,各膏劑於60℃保存1個月後,膏體層中所含之水楊酸二醇酯之含量以氣體層析法進行測定。相對於調製時之膏劑中水楊酸二醇酯之含量(初期值)之保存後之膏劑之水楊酸二醇酯之含量示於表2。
比較實施例1~3之膏劑時,即使甘油含量相同之膏劑,若酒石酸含量多,則膏體層之pH變低,水楊酸二醇酯之保存安定性變高。且,比較實施例1、4、6、8之貼劑時,甘油含量降低,水楊酸二醇酯之保存安定性
提高。另一方面,膏體層中所佔之甘油含量為35質量%之比較例1之膏劑,水楊酸二醇酯之保存安定性降低。且,膏體層中所佔之甘油含量為10質量%之比較例2之膏劑,水楊酸二醇酯之保存安定性雖高,但保濕性於作為膏劑時並不充分。
依據表3記載之組成,充分混合各成分,調製膏體液。將所得膏體液均一延展於剝離襯墊上,於其上層合基布,藉由剝離掉剝離襯墊,分別獲得實施例9~12之膏劑。
依據日本藥典之記載,測定所得膏劑之膏體層之pH,其值示於表4。又,各膏劑於60℃保存1個月後,膏體層中所含之水楊酸二醇酯之含量以氣體層析法進行測定。相對於調製時之膏劑中水楊酸二醇酯之含量之保存後之膏劑之水楊酸二醇酯之含量示於表4。
實施例11之膏劑與實施例9之膏劑比較時,甘油之含量低,水楊酸二醇酯之安定性更改善。該傾向於實施例10及12之膏劑中亦同樣。
Claims (5)
- 一種膏劑,其係於基布上具備膏體層之膏劑,且前述膏體層含有水楊酸二醇酯、水溶性高分子、水及甘油,甘油含量以質量基準計,為水楊酸二醇酯含量之8~15倍。
- 如請求項1之膏劑,其中前述甘油含量以質量基準計,為前述水楊酸二醇酯含量之8~14倍。
- 如請求項1或2之膏劑,其中前述膏體層之pH為4.7~5.1。
- 如請求項1~3中任一項之膏劑,其中前述膏體層含有聚(丙烯酸甲酯/丙烯酸2-乙基己酯)。
- 如請求項1~4中任一項之膏劑,其中前述水溶性高分子包含聚丙烯酸或聚丙烯酸中和物。
