Claims (1)

Изобретение относится к области молекулярной биологии и биотехнологии, а именно - к способам размножения, экспрессии, клонирования и диагностики нуклеиновых кислот в бесклеточных системах. Преимущественной областью использования является препаративный синтез индивидуальных нуклеиновых кислот, клонирование нуклеиновых кислот и обнаружение специфических нуклеиновых кислот в анализируемом материале. Целью изобретения является способ размножения и экспрессии нуклеиновых кислот, при котором потомство каждой молекулы (клон) и продукты экспрессии остаются в той зоне реакционного объема, где исходно находилась матричная молекула. Размножение и/или экспрессию нуклеиновых кислот осуществляют в среде, которую иммобилизуют путем использования органической и/или неорганической твердой основы, пронизывающей среду и имеющей пористую, волокнистую, сетчатую, капиллярную, складчатую или слоистую структуру, которая включает компоненты бесклеточной ферментной системы экспоненциального размножения нуклеиновых кислот и/или компоненты бесклеточной ферментной системы экспрессии нуклеиновых кислот. Предпочтительно использование твердой основы, способной к обратимым взаимодействиям с нуклеиновыми кислотами, а также использование иммобилизованной среды, сформованной в виде по крайней мере одного тонкого слоя. Способ позволяет клонировать нуклеиновые кислоты вне клетки, а также детектировать одиночные молекулы нуклеиновых кислот в исследуемом образце.The invention relates to the field of molecular biology and biotechnology, namely to methods of reproduction, expression, cloning and diagnosis of nucleic acids in cell-free systems. The aim of the invention is a method of reproduction and expression of nucleic acids, in which the progeny of each molecule (clone) and expression products remain in the zone of the reaction volume, where the matrix molecule was originally located. The reproduction and / or expression of nucleic acids is carried out in a medium that is immobilized by using an organic and / or inorganic solid base that penetrates the medium and has a porous, fibrous, reticular, capillary, folded or layered structure that includes components of a cell-free enzyme system of exponential reproduction of nucleic acids and / or components of a cell-free nucleic acid expression system. It is preferable to use a solid base capable of reversible interactions with nucleic acids, as well as the use of an immobilized medium formed in the form of at least one thin layer. The method allows to clone nucleic acids outside the cell, as well as to detect single nucleic acid molecules in the sample under study.
